Gastric cancer(GC)is the third most common cause of cancer death globally and a large portion of patients are diagnosed at advanced stages with cancer invasion and metastasis1,***,the mechanisms underlying the invasion and metastasis of GC remain to be *** plays critical roles in cell mobility via cytoskeleton regulation in various cell types.3 In this study,we further reported that ZYX promoted migration,invasion,and metastasis of GC ***,ZYX promoted WNK1 activation and SNAl1 up-regulation,inducing epithelial-mesenchymal transition(EMT)to enhance the mobility of GC *** of WNK1 impaired the mobility of GC ***,ZYX/WNK1 could be potential therapeutic targets for GC treatment.

Study on the Mechanism of Glutamine Promoting the Synthesis of Colonic Mucin and Maintaining the Stability of Intestinal Microbiota in Septic Mice

Colonic mucus and gut microbiota closely linked through mutual regulatory effect. While their alterations during sepsis and whether glutamine can maintain the colonic mucus barrier and gut microbiota stability are unclear. Cecal ligation and puncture-induced sepsis in mice was utilized to observe changes in colonic mucus， gut microbiota， and their interaction with glutamine intervention. Our findings indicated that glutamine mitigated sepsis-induced intestinal damage and restores colonic mucus barrier function by augmenting mucin synthesis. Further analysis revealed that goblet cells were under oxidative stress after sepsis， resulting in anterior gradient-2 (AGR2)， the key mucin-modifying enzyme， being dissynthesized， and inhibiting mucin2 (MUC2) maturation. glutamine could ameliorates this situation by promoting the key enzyme glucose-6-phosphate dehydrogenase (G6PD) glycosylation in the pentose phosphate pathway， increasing the reduced nicotinamide adenine dinucleotide phosphate (NADPH) synthesis， reducing endoplasmic reticulum stress， and accelerating AGR2 synthesis and MUC2 maturation. Additionally， glutamine aided in maintaining gut microbiota stability during sepsis， up-regulating mucin-associated bacteria such as Akkermansia and Alistipes. These bacteria， intimately linked to mucin synthesis and degradation， may impact intestinal mucus stability. In conclusion， glutamine can maintain goblet cell redox balance， promotes AGR2 synthesis and MUC2 maturation， shields the mucus barrier， and potentially maintains gut microbiota stability by regulating the interaction between bacteria and mucus， thus alleviating sepsis-induced intestinal damage.

BACKGROUND Multidrug-resistant Gram-negative bacteria,exacerbated by excessive use of antimicrobials and immunosuppressants,are a major health *** To study the clinical efficacy and safety of colistin sulfate in the treatment of carbapenem-resistant Gram-negative bacilli-induced pneumonia,and to provide theoretical reference for clinical diagnosis and *** This retrospective analysis involved 54 patients with Gram-negative bacilli pneumonia admitted to intensive care unit of The General hospital of the Northern Theater Command of the People's Liberation army of China from August 2020 to June *** bacteriological culture,the patients'airway secretions were collected to confirm the presence of Gram-negative *** patients were divided into the experimental and control groups according to the medication *** research group consisted of 28 patients who received polymyxin sulfate combined with other drugs through intravenous,nebulization,or intravenous combined with nebulization,with a daily dosage of 1.5–3.0 million *** control group consisted of 26 patients who received standard dosages of other antibiotics(including sulbactam sodium for injection,cefoperazone sodium sulbactam for injection,tigecycline,meropenem,or vaborbactam).RESULTS Of the 28 patients included in the research group,26 patients showed improvement,treatment was ineffective for two patients,and one patient died,with the treatment efficacy rate of 92.82%.Of the 26 patients in the control group,18 patients improved,treatment was ineffective for eight patients,and two patients died,with the treatment efficacy rate of 54.9%;significant difference was observed between the two groups(P<0.05).The levels of white blood cell(WBC),procalcitonin(PCT),and C-reactive protein(CRP)in both groups were significantly lower after treatment than before treatment(P<0.05),and the levels of WBC,PCT,and CRP in the research group were significantly lower than those in the control group
