Probing the processing of facial expressions in monkeys via time perception and eye tracking

Accurately recognizing facial expressions is essential for effective social ***-human primates(NHPs)are widely used in the study of the neural mechanisms underpinning facial expression processing,yet it remains unclear how well monkeys can recognize the facial expressions of other species such as *** this study,we systematically investigated how monkeys process the facial expressions of conspecifics and humans using eye-tracking technology and sophisticated behavioral tasks,namely the temporal discrimination task(TDT)and face scan task(FST).We found that monkeys showed prolonged subjective time perception in response to Negative facial expressions in monkeys while showing longer reaction time to Negative facial expressions in *** faces also reliably induced divergent pupil contraction in response to different expressions,while human faces and scrambled monkey faces did ***,viewing patterns in the FST indicated that monkeys only showed bias toward emotional expressions upon observing monkey ***,masking the eye region marginally decreased the viewing duration for monkey faces but not for human *** probing facial expression processing in monkeys,our study demonstrates that monkeys are more sensitive to the facial expressions of conspecifics than those of humans,thus shedding new light on inter-species communication through facial expressions between NHPs and humans.

Integrating multisensory inputs to generate accurate perception and guide behavior is among the most critical functions of the *** regions such as the amygdala are involved in sensory processing including vision and audition,yet their roles in multisensory integration remain *** this study,we systematically investigated the function of neurons in the amygdala and adjacent regions in integrating audiovisual sensory inputs using a semi-chronic multi-electrode array and multiple combinations of audiovisual *** a sample of 332 neurons,we showed the diverse response patterns to audiovisual stimuli and the neural characteristics of bimodal over unimodal modulation,which could be classified into four types with differentiated regional *** the hierarchical clustering method,neurons were further clustered into five groups and associated with different integrating functions and ***,regions distinguishing congruent and incongruent bimodal sensory inputs were ***,visual processing dominates audiovisual integration in the amygdala and adjacent *** findings shed new light on the neural mechanisms of multisensory integration in the primate brain.

Polyetheretherketone （PEEK） is regarded as one of the most potential candidates for replacing current implant applications. To obtain good bone-implant interfaces, many modification methods have been developed to enable PEEK and PEEK- based composites from bio-inert to bioactive. Among them, physical methods have aroused significant attention and been widely used to modify PEEK for orthopedic implants. This review summarizes current physical modification techniques of PEEK for orthopedic applications, which include composite strategies, surface coating methods and irradiation treatments. The positive consequences of those modification methods will encourage continuing investigations and stimulate the wide range of applications of PEEK-based implants in orthopedics.

Retrograde adeno-associated viruses(AAVs)are capable of infecting the axons of projection neurons and serve as a powerful tool for the anatomical and functional characterization of neural ***,few retro-grade AAV capsids have been shown to offer access to cor-tical projection neurons across different species and enable the manipulation of neural function in non-human primates(NHPs).Here,we report the development of a novel retro-grade AAV capsid,AAV-DJ8R,which efficiently labeled cortical projection neurons after local administration into the striatum of mice and *** addition,intrastriatally injected AAV-DJ8R mediated opsin expression in the mouse motor cortex and induced robust behavioral ***,AAV-DJ8R markedly increased motor cortical neuron firing upon optogenetic light stimulation after viral delivery into the macaque *** data demonstrate the usefulness of AAV-DJ8R as an efficient retrograde tracer for cortical projection neurons in rodents and NHPs and indicate its suitability for use in conducting functional interrogations.

In vitro and in vivo evaluation of bone morphogenetic protein- 2 （BMP-2） immobilized collagen-coated polyetheretherketone （PEEK）

Polyetheretherketone （PEEK） is regarded as one of the most potential candidates of biomaterials in spinal implant applications. However, as a bioinert material, PEEK plays a limited role in osteoconduction and osseointegration. In this study, recombinant human bone morphogenetic protein-2 （rhBMP-2） was immobilized onto the surface of collagen-coated PEEK in order to prepare a multi-functional material. After adsorbed onto the PEEK surface by hydroph（bic interaction, collagen was cross-linked with N-（3-dimethylaminopropyl）-N＇-ethyl carbodiimide hydrochloride （EDC） and N-hydro xysuccinimide （NHS）. EDCINHS system also contributed to the immobilization of rhBMP- 2. Water contact angle tests, XPS and SEM clearly demonstrated the surface changes. ELISA tests quantified the amount of rhBMP-2 immobilized and the release over a period of 30 d. In vitro evaluation proved that the osteogenesis differentiation rate was higher when cells were cultured on modified PEEK discs than on regular ones. In vivo tests were conducted and positive changes of major parameters were presented. This report demonstrates that the rhBMP-2 immobilized method for PEEK modification increase bioactivity in vitro and in vivo, suggesting its practicability in orthopedic and spinal clinical applications.

Backgrounds:Streptozotocin(STZ)-induced diabetic monkey is a wide used preclinical animal model for the investigation of diabetes such as islet transplantation and development of diabetic *** are serious side effects of this method,including nausea,emesis,weight loss,liver damage,renal failure,and metabolic *** order to reduce the side effects,diabetic monkeys were induced using clinicalgrade ***,clinical-grade STZ is not available in ***,we establised a method by using 100 mg/kg analytical-grade STZ to induce complete diabetes in cynomolgus monkey without generating adverse effects to liver and ***:Three cynomolgus monkeys were used in this study.100 mg/kg STZ dissolved in normal saline and infused through vein line in 5 minutes after indwelling catheter in the carotid artery and jugular *** the STZ administration,blood glucose levels were examined every 1 or 2 hours in the first 48 ***,blood glucose levels were examined twice per day during the first week after the STZ *** and C-peptide levels were measured by *** chemistry of hepatic and renal function tests were *** and glucagon expression in the islet of diabetic monkey and normal monkey were examined by immunohistochemistry ***:The stimulated C-peptide level(Intravenous glucose tolerance test)which is less than 0.5 ng/mL,the triphasic blood glucose response and the destroyed β cell suggested the complete induction of diabetes *** apparent adverse effects were observed including no signs of vomiting and toxicity after STZ ***:In summary,we established a safe and reproducible STZ-induced diabetic cynomolgus monkey model for islet transplantation which will be used to develop novel approaches for the treatment of diabetes.

The outbreak of the COVID-19 pandemic was partially due to the challenge of identifying asymptomatic and presymptomatic carriers of the virus, and thus highlights a strong motivation for diagnostics with high sensitivity that can be rapidly deployed. On the other hand, several concerning SARS-Co V-2 variants,including Omicron, are required to be identified as soon as the samples are identified as ‘positive’.Unfortunately, a traditional PCR test does not allow their specific identification. Herein, for the first time,we have developed MOPCS(Methodologies of Photonic CRISPR Sensing), which combines an optical sensing technology-surface plasmon resonance(SPR) with the ‘gene scissors’ clustered regularly interspaced short palindromic repeat(CRISPR) technique to achieve both high sensitivity and specificity when it comes to measurement of viral variants. MOPCS is a low-cost, CRISPR/Cas12a-systemempowered SPR gene-detecting platform that can analyze viral RNA, without the need for amplification,within 38 min from sample input to results output, and achieve a limit of detection of 15 fM. MOPCS achieves a highly sensitive analysis of SARS-Co V-2, and mutations appear in variants B.1.617.2(Delta),B.1.1.529(Omicron) and BA.1(a subtype of Omicron). This platform was also used to analyze some recently collected patient samples from a local outbreak in China, identified by the centers for Disease Control and Prevention. This innovative CRISPR-empowered SPR platform will further contribute to the fast, sensitive and accurate detection of target nucleic acid sequences with single-base mutations.
