Objective To recover broad-neutralizing monoclonal antibodies(Bn Abs)from avian influenza A(H5N1)virus infection cases and investigate their genetic and functional *** We screened the Abs repertoires of expanded B cells circulating in the peripheral blood of H5N1 *** genetic basis,biological functions,and epitopes of the obtained Bn Abs were assessed and *** Two Bn Abs,2-12 D5,and 3-37 G7.1,were respectively obtained from two human H5N1 cases on days 12 and 21 after disease *** Abs demonstrated cross-neutralizing and Ab-dependent cellular cytotoxicity(ADCC)*** derived from distinct Ab lineages,i.e.,V^H1-69-D2-15-JH^4(2-12D5)and V^H1-2-D3-9-JH^5(3-32 G7.1),the Bn Abs were directed toward CR6261-like epitopes in the HA stem,and HA2 I45 in the hydrophobic pocket was the critical residue for their *** motifs for binding with the HA stem,namely,IFY in VH1-69-encoded Abs and LXYFXW in D3-9-encoded Abs,were also observed in 2-12D5 and 3-32 G7.1,*** Cross-reactive B cells of different germline origins could be activated and re-circulated by avian influenza *** HA stem epitopes targeted by the Bn Abs,and the two Ab-encoding genes usage implied the VH1-69 and D3-9 are the ideal candidates triggered by influenza virus for vaccine development.

English newspaper reading course plays a positive effects on English Teaching in Higher Vocational Colleges. However it does not obtain enough attention at present. The application and problems of English newspaper reading in English Teaching in Higher Vocational Colleges are discussed in terms of English teaching material selection＇s appropriateness, teaching 1 diversity and the major problems. The summary of experience lesson is made and teaching suggestions are put forward, which makes a constructive effect on English newspaper reading development in English Teaching in Higher Vocational Colleges.

Short-wavelength infrared(SWIR)sensors have attracted keen attention due to the increasing necessity in a variety of scientific and industrial applications,including biomedical and information technology *** conventional SWIR sensors are made of inorganic materials with rigid and brittle characteristics,organic materials with a discrete SWIR absorption are required for flexible SWIR sensors in the flexible electronics ***,we demonstrate that a polytriarylamine,poly[N,N′-bis(4-butylphenyl)-N,N′-bis(phenyl)benzidine](PolyTPD),can absorb almost full range of SWIR wavelength(λ=1000–3200 nm)after 48 h doping with tris(pentafluorophenyl)borane(BCF).The spectroscopic characterization disclosed that an electron transfer from PolyTPD to BCF created a new low energy level(gap)state leading to the SWIR absorption in the BCF-doped PolyTPD *** phototransistors(OPTRs)with the BCF-doped PolyTPD films as a gate-sensing layer could detect the SWIR light with a reasonable photoresponsivity of~538 mAW^(−1)(λ=1500 nm),~541 mAW^(−1)(λ=2000 nm),and~222 mAW^(−1)(λ=3000 nm).The present breakthrough SWIR-OPTR technology can pave a way for further advances in SWIR-absorbing organic materials and flexible SWIR sensors.

Background: The in vivo concentration of bicarbonate(HCO3-), one of the essential sperm capacitating effectors,varies greatly in the different environments sperm go through from cauda epididymis to the fertilisation site. On the contrary, porcine in vitro sperm capacitation and fertilisation media usually contains a standard concentration of25 mmol/L, and one of the main problems presented is the unacceptable high incidence of polyspermy. This work hypothesised that by modifying the HCO3-concentration of the medium, the output of in vitro sperm capacitation and fertilisation could be ***: Once exposed to the capacitation medium, the intracellular pH(pHi) of spermatozoa increased immediately even at low concentrations of HCO3-, but only extracellular concentrations of and above 15 mmol/L increased the substrates protein kinase A phosphorylation(pPKAs). Although with a significant delay, 15 mmol/L of HCO3-stimulated sperm linear motility and increased other late events in capacitation such as tyrosine phosphorylation(Tyr-P) to levels similar to those obtained with 25 mmol/L. This information allowed the establishment of a new in vitro fertilisation(IVF)system based on the optimization of HCO3-concentration to 15 mmol/L, which led to a 25.3% increment of the viable zygotes(8.6% in the standard system vs. 33.9%).Conclusions: Optimising HCO3-concentrations allows for establishing an IVF method that significantly reduced porcine polyspermy and increased the production of viable zygotes. A concentration of 15 mmol/L of HCO3-in the medium is sufficient to trigger the in vitro sperm capacitation and increase the fertilisation efficiency in porcine.

Distribution and relative activity of matrix metalloproteinase-2 in human coronal dentin

The presence of matrix metalloproteinase-2 （MMP-2） in dentin has been reported, but its distribution and activity level in mature human coronal dentin are not well understood. The purpose of this study was to determine the MMP-2 distribution and relative activity in demineralized dentin. Crowns of twenty eight human molars were sectioned into inner （ID）, middle （MD）, and outer dentin （OD） regions and demineralized. MMP-2 was extracted with 0.33 mol·L-1 EDTA/2 mol·L-1 guanidine-HCl, pH 7.4, and MMP-2 concentration was estimated with enzyme-linked immunoabsorbant assay （ELISA）. Further characterization was accomplished by Western blotting analysis and gelatin zymography. The mean concentrations of MMP-2 per mg dentin protein in the dentin regions were significantly different （P=0.043）： 0.9 ng （ID）, 0.4 ng （MD）, and 2.2 ng （OD）, respectively. The pattern of MMP-2 concentration was OD〉ID〉MD. Western blotting analysis detected -66 and -72 kDa immunopositive proteins corresponding to pro- and mature MMP-2, respectively, in the ID and MD, and a -66 kDa protein in the OD. Gelatinolytic activity consistent with MMP-2 was detected in all regions. Interestingly, the pattern of levels of Western blot immunodetection and gelatinolytic activity was MD〉ID〉OD. The eoneentration of MMP-2 in human coronal dentin was highest in the region of dentin that contains the dentinoenamel junction and least in the middle region of dentin. However, levels of Western blot immunodetection and gelatinolytic activity did not correlate with the estimated regional concentrations of MMP-2, potentially indicating region specific protein interactions.

Although melanoma is notorious for its high degree of heterogeneity and plasticity1,2,the origin and magnitude of cell-state diversity remains poorly ***,it is unclear whether growth and metastatic dissemination are supported by overlapping or distinct melanoma ***,by combining mouse genetics,single-cell and spatial transcriptomics,lineage tracing and quantitative modelling,we provide evidence of a hierarchical model of tumour growth that mirrors the cellular and molecular logic underlying the cell-fate specification and differentiation of the embryonic neural *** show that tumorigenic competence is associated with a spatially localized perivascular niche,a phenotype acquired through an intercellular communication pathway established by endothelial *** with a model in which only a fraction of cells are fated to fuel growth,temporal single-cell tracing of a population of melanoma cells with a mesenchymal-like state revealed that these cells do not contribute to primary tumour growth but,instead,constitute a pool of metastatic initiating cells that switch cell identity while disseminating to secondary *** data provide a spatially and temporally resolved map of the diversity and trajectories of melanoma cell states and suggest that the ability to support growth and metastasis are limited to distinct pools of *** observation that these phenotypic competencies can be dynamically acquired after exposure to specific niche signals warrant the development of therapeutic strategies that interfere with the cancer cell reprogramming activity of such microenvironmental cues.

DNA nanotechnology utilizes DNA double strands as building units for self-assembly of DNA *** specific base-pairing interaction between DNA molecules is the basis of these *** decades of development,this technology has been able to construct complex and programmable *** the increase in delicate nature and complexity of the synthesized nanostructures,a characterization technology that can observe these structures in three dimensions has become necessary,and developing such a technology is considerably *** assemblies have been studied using different characterization methods including atomic force microscopy(AFM),scanning electron microscopy(SEM),and transmission electron microscopy(TEM).However,the three-dimensional(3D)DNA assemblies always collapse locally due to the dehydration during the drying *** electron microscopy(cryo-EM)can overcome the challenge by maintaining three-dimensional morphologies of the cryogenic samples and reconstruct the 3D models from cryogenic samples accordingly by collecting thousands of two-dimensional(2D)projection images,which can restore their original morphologies in ***,we have reviewed several typical cases of 3D DNA-assemblies and highlighted the applications of cryo-EM in characterization of these *** comparing with some other characterization methods,we have shown how cryo-EM promoted the development of structural characterization in the field of DNA nanotechnology.

Prostate-specific antigen(PSA)is the most widely used biomarker for the early diagnosis of prostate *** methods for PSA detection are burdened with some limitations and require ***,we developed a novel microfluidic–electrochemical(μFEC)detection system for PSA ***,we constructed an electrochemical biosensor based on screen-printed electrodes(SPEs)with modification of gold nanoflowers(Au NFs)and DNA tetrahedron structural probes(TSPs),which showed great detection ***,we fabricated microfluidic chips by DNA TSP-Au NF-modified SPEs and a PDMS layer with designed dense meandering ***,theμFEC detection system was achieved based on microfluidic chips integrated with the liquid automatic conveying unit and electrochemical detection ***μFEC system we developed acquired great detection performance for PSA detection in PBS *** PSA assays in spiked serum samples of theμFEC system,we obtained a linear dynamic range of 1–100 ng/mL with a limit of detection of 0.2 ng/mL and a total reaction time<25 *** serum samples of prostate cancer patients presented a strong correlation between the“gold-standard”chemiluminescence assays and theμFEC *** terms of operation procedure,cost,and reaction time,our method was superior to the current methods for PSA detection and shows great potential for practical clinical application in the future.
