In the present study, 28 Chinese medicinal herbs belonging to traditional Chinese medicine(TCM) for the treatment of type 2 diabetes were selected to explore the application of network pharmacology in developing new Chinese herbal medicine formulae for the treatment of type 2 diabetes mellitus(T2DM). These herbs have the highest appearance rate in the literature, and their compounds are listed. The human protein–protein interaction network and the T2DM disease protein interaction network were constructed. Then, the related algorithm for network topology was used to perform interventions on the interaction network of disease proteins and normal human proteins to test different Chinese herbal medicine compound combinations, according to the information on the interaction of compounds–targets in two databases, namely TarN et and the Medicinal Plants Database. Results of the intervention scores indicate that the method proposed in this study can provide new effective combinations of Chinese herbal medicines for T2DM. Network pharmacology can effectively promote the modernization and development of TCM.

Epimedium pubescens *** a well-known traditional Chinese medicinal herb with flavonol glycosides as the major pharmaceutically active ***-glycosyltransferases(UGTs)are a group of enzymes responsible for the glycosylation of flavonoid *** this study,a genome-wide analysis was performed to identify UGT family genes in *** a result,a total of 339 putative UGT genes were identified,which represents the largest UGT gene family known thus far,implying a significant expansion of the UGT gene family in *** EpUGTs were unevenly distributed across six chromosomes,and they were classified into 17 major *** expression profiles showed that UGT genes were differentially expressed in roots,leaves,flowers,shoots and *** particular,several EpUGTs were highly induced by high light intensity,which was consistent with the accumulation level of bioactive flavonoids in *** UGT79 genes that were preferentially expressed in roots or leaves were successfully expressed in ***,and only the recombinant EpGT60 protein was found to be active toward 8-prenylkaempferol and icaritin to produce the key bioactive compounds baohuoside II and baohuoside *** optimal temperature,pH,k_(m) and V_(max) were determined for the recombinant EpGT60 *** addition,expression of recombinant EpGT60 in *** cell culture led to successful production of baohuoside II when fed *** study provides a foundation for further functional characterization of UGT genes in *** and provides key candidate genes for bioengineering bioactive flavonoids in ***.

In the present study, we developed and validated a rapid analytical method using high performance liquid chromatography coupled with quadrupole time of flight mass spectrometry(HPLC-Q-TOF/MS) to investigate the metabolic profile of Guge Fengtong tablet(GGFTT), a traditional Chinese medicine. The urine and bile samples were collected with 24 h after oral administration of GGFTT. A total of 34 compounds, including 11 parent compounds and 23 metabolites were unambiguously or tentatively identified. Our results indicated that glucuronidation, oxidation and methylation were the major metabolic pathways of the constituents in GGFTT. In addition, the results of this work also demonstrated the feasibility of HPLC-ESI-Q-TOF/MS for reliable characterization of the in vivo metabolites from herbal preparations.

Gastrodia elata (G. elata ) is a traditional Chinese herbal medicine. In recent years, G. elata has been widely used in food and pharmaceutical industries because of its excellent health benefits. However, the stability and bioactivity of phenolic compounds from G. elata after in vitro gastrointestinal digestion remain unknown. In this study, the aqueous extract of G. elata rhizomes was used to simulate gastrointestinal digestion and colonic fermentation in vitro . This study aimed to evaluate the bioaccessibility during the simulation of gastrointestinal and colonic fermentation and to clarify the effects of different digestion stages on the anti-oxidant and enzyme inhibitory activities and intestinal health of samples in vitro . The results indicated that the total polyphenol and flavonoid contents after colonic fermentation for 72 h were 5.48- and 3.24- fold higher than those without digestion, respectively. Except for gastrodigenin and ferulic acid, the contents and bioaccessibility of the other phenols after fermentation were higher than those in the small-intestine stage. Antioxidant activity was positively correlated with the total polyphenol content. The inhibitory activities of pancrelipase and cholinesterase were enhanced by gastric digestion and colonic fermentation. The sample promoted the production of Lactobacillus and decreased the abundance of pathogenic bacteria. Moreover, the content of short-chain fatty acids also exhibited an upward trend. Therefore, G. elata has the potential to develop into a health food with anti-oxidant, anti-obesity, and intestinal health functions.

The steamed Panax ginseng C. A. Mey., Panax notoginseng(Burk.) F. H. Chen, and Panax quinquefolium L. not only facilitate storage, but also increase the content of rare saponins and enhance their clinical application value. The traditional steaming process has high energy consumption, low efficiency, and lacks standardized operating procedures and unified standards. This paper retrieves the research literature on the steaming process parameters, rare saponin increments, and efficacy enhancement results of ginseng plants. By summarizing the effects of different steaming processes on rare saponins and pharmacodynamics in P. ginseng, P. notoginseng and P. quinquefolium, we explored new steaming methods and established a new quality evaluation system to provide guarantee for the effectiveness and safety of clinical medication and provide scientific basis for its rational use.

The quality standards for Fructus Comi have been established based on the effects of the manufacturing *** critical process parameters(CPPs)of extraction,filtration,and concentration to prepare Fructus Comi concentrate were identified by Plackett-Burman design with a single batch of Fructus Corni,which were heating medium temperature,extraction time,and water *** yield,loganin yield,and dry matter yield were process critical quality attributes(CQAs).CPPs arranged with a Box-Behnken design were applied to treat different batches of Fructus Comi After constructing a model that included CPPs,material propertie s,and process CQAs,loganin content was found to be the critical material attribute(CMA).The design space was calculated with a probability *** to the limits of process CQAs,the minimum content of loganin in Fructus Corni was calculated with an error propagation method,which was 6.92 mg·g^(-1).When the content of loganin in Fructus Corni reaches up to 6.92 mg·g^(-1),the material is considered high-quality and is most suitable for the ***-quality material can be used for production of Fructus Comi *** method can also be used to set material quality standards for other Chinese medicines.
