Ultra‑High Sensitivity Anisotropic Piezoelectric Sensors for Structural Health Monitoring and Robotic Perception

Monitoring minuscule mechanical signals,both in magnitude and direction,is imperative in many application scenarios,e.g.,structural health monitoring and robotic sensing ***,the piezoelectric sensor struggles to satisfy the requirements for directional recognition due to the limited piezoelectric coefficient matrix,and achieving sensitivity for detecting micrometer-scale deformations is also ***,we develop a vector sensor composed of lead zirconate titanate-electronic grade glass fiber composite filaments with oriented arrangement,capable of detecting minute anisotropic *** as-prepared vector sensor can identify the deformation directions even when subjected to an unprecedented nominal strain of 0.06%,thereby enabling its utility in accurately discerning the 5μm-height wrinkles in thin films and in monitoring human pulse *** ultra-high sensitivity is attributed to the formation of porous ferroelectret and the efficient load transfer efficiency of continuous lead zirconate titanate ***,when integrated with machine learning techniques,the sensor’s capability to recognize multi-signals enables it to differentiate between 10 types of fine textures with 100%*** structural design in piezoelectric devices enables a more comprehensive perception of mechanical stimuli,offering a novel perspective for enhancing recognition accuracy.

Hard carbon(HC)is widely used in sodium-ion batteries(SIBs),but its performance has always been limited by lowinitial Coulombic efficiency(ICE)and cycling *** compensation agent is a favorable strategy to make up for the loss of active sodium ions consumed *** it lacks agent that effectively decomposes to increase the active sodium ions as well as regulate carbon defects for decreasing the irreversible sodium ions ***,we propose 1,2-dihydroxybenzene Na salt(NaDB)as a cathode compensation agent with high specific capacity(347.9 mAh g^(-1)),lower desodiation potential(2.4–2.8 V)and high utilization(99%).Meanwhile,its byproduct could functionalize HC with more C]O groups and promote its reversible ***,the presodiation hard carbon(pHC)anode exhibits highly reversible capacity of 204.7 mAh g^(-1) with 98%retention at 5 C rate over 1000 ***,with 5 wt%NaDB initially coated on the Na3V2(PO4)3(NVP)cathode,the capacity retention of NVP t NaDBjHC cell could increase from 22%to 89%after 1000 cycles at 1 C *** work provides a new avenue to improve reversible capacity and cycling performance of SIBs through designing functional cathode compensation agent.

Study on Substrate Specificity at Subsites for Severe Acute Respiratory Syndrome Coronavirus 3CL Protease

Autocleavage assay and peptide-based cleavage assay were used to study the substratespecificity of 3CL protease from the severe acute respiratory syndrome *** was found that therecognition between the enzyme and its substrates involved many positions in the substrate,at least includingresidues from P4 to P2′.The deletion of either P4 or P2′residue in the substrate would decrease its cleavageefficiency *** contrast to the previous suggestion that only small residues in substrate could beaccommodated to the S1′subsite,we have found that bulky residues such as Tyr and Trp were also *** addition,based on both peptide-based assay and autocleavage assay,Ile at the P1′position could not behydrolyzed,but the mutant L27A could hydrolyze the Ile peptide *** suggested that there was astereo hindrance between the S1′subsite and the side chain of Ile in the *** 20 amino acids exceptPro could be the residue at the P2′position in the substrate,but the cleavage efficiencies were *** specificity information of the enzyme is helpful for potent anti-virus inhibitor design and usefulfor other coronavirus studies.

Nanog gene plays a key role in maintaining pluripotency of ES cells and early embryonic cells.A 5’ flank sequenceof the Nanog gene has been reported to be regulated differentially,and two regulatory elements within the Nanog pro-moter,namely Oct-4 and Sox-2 binding sites,have been identified to regulate the transcriptional activity ofNanog *** this report,we identified the role of two putative Sp 1 binding sites located in the Nanog gene 5’-flanking region inregulation of murine Nanog gene *** studies showed that the two sites were essential for the Nanogpromoter *** shift and supershift analysis showed that both sites specifically bind Spl and ***,overexpression of dominant-negative Sp 1 or Sp3 mutants significantly inhibits Nanog promoter *** resultssuggest that the transcription factor Spl and Sp3 are important for Murine Nanog gene expression.

Identification of γ1 subunit of GABAA receptor in rat testis

The isoform type ofγsubunits of GABAA receptor is a molecular determinant of its pharmacological characteristics. At present, the existence of GABAA receptor in mammalian sperm is still a controversy. By using degenerate primers designed according to highly conserved region in all threeγ(γ1, γ2 andγ3) subunits cloned in rat brain, we performed reverse transcription polymerase chain reaction (RT-PCR) to examine the expression pattern of -γsubunits of GABAA receptor in rat testis. Only one 370 bp fragment was obtained from RT-PCR in rat testis and sequencing results showed that it representedγ1 subunit, but notγ2 orγ3 subunit. Using the cloned fragment as probe, a 3.8 kb transcript which in size as same asγ1 ubunit in rat brain was detected in rat testis mRNA by performing Northern blot assay. Furthermore, results of in situ hybridization assay confirmed thatγ1 subunit was expressed in round spermatids and spermatozoa, maybe also in secondary spermatocyte. These evidences proved that 71 subunit of GABAA receptor is exclusively expressed in rat testis and this feature may be the structural basis of the specific function of GABAA receptors in sperm acrosome reaction.

Stem cell niche is a specialized microenvironment crucial to *** testis in Drosophila contains two dif-ferent types of stem cells,the germline stem cells and the somatic cyst stem cells that are sustained by their respec-tive niche signals,thus is a good system for studying the interaction between the stem cells and their hosting *** JAK-STAT and BMP pathways are known to play critical roles in the self-renewal of different kinds of stem cells,but the roles of several other pathways have emerged recently in a complex signaling network in the testis *** of independent observations from three research groups have uncovered an important role of Hedgehog(Hh)in the Drosophila testis *** this review,we summarize these recent fi ndings and discuss the interplay between the Hh signaling mechanisms and those of the JAK-STAT and BMP *** also dis-cuss directions for further investigation.

Effect of thermal injury on LPS-mediated Toll signaling pathways by murine peritoneal macrophages:inhibition of DNA-binding of transcription factor AP-1 and NF-κB and gene expression of c-fos and IL-12p40

Thermal injury is able to induce the immune functional inhibition in the host, but the mechanisms are largely unknown. The available evidence indicates that macrophages play an important role in immune regulations and host defenses. Thus, studies of the effect of thermal injury on macrophages are essential. Transcriptional factors of AP-1 (activator protein-1) and NF-кB (nuclear factor-кB) are critical members of signal transduction pathways. These transcriptional factors have been shown to be necessary for a variety of expressions of cytokine genes. In this investigation, the effect of thermal injury on AP-1 and NF-кB was studied using a model for the murine thermal injury. Our findings indicated that thermal injury mediated the inhibition of AP-1 and NF-кB DNA binding activities and c-fos and IL-12 p40 gene expressions in response to LPS.

Adenovirus-mediated expression of pig α(1,3) galactosyltransferase reconstructs Gal α(1,3) Gal epitope on the surface of human tumor cells

Gal α(1, 3) Gal (gal epitope) is a carbohydrate epitope and synthesized in large amount by α(1, 3) galactosyltransferase [α(1, 3) GT] enzyme on the cells of lower mammalian animals such as pigs and mice. Human has no gal epitope due to the inactivation of α(1, 3) GT gene but produces a large amount of antibodies (anti-Gal) which recognize Gal α(1, 3) Gal structures specifically. In this study, a replication deficient recombinant adenoviral vector Ad5sGT containing pig α(1, 3) GT cDNA was constructed and characterized. Adenoviral vector-mediated transfer of pig α(1, 3) GT gene into human tumor cells such as malignant melanoma A375, stomach cancer SGC-7901, and lung cancer SPC-A-1 was reported for the first time. Results showed that Gal epitope did not increase the sensitivity of human tumor cells to human complement-mediated lysis, although human complement activation and the binding of human IgG and IgM natural antibodies to human tumor cells were enhanced significantly after Ad5sGT transduction. Appearance of gal epitope on the human tumor cells changed the expression of cell surface carbohydrates reacting with Ulex europaeus I (UEA I) lectins, Vicia villosa agglutinin (VVA), Arachis hypogaea agglutinin (PNA), and Glycine max agglutinin (SBA) to different degrees. In addition, no effect of gal epitope on the growth in vitro of human tumor cells was observed in MTT assay.

Bcl-2 over-expression and activation of protein kinase C suppress the Trail-induced apoptosis in Jurkat T cells

Trail, a tumor necrosis factor-related apoptosis-inducing ligand, is a novel potent endogenous activator of the cell death pathway through the activation of cell surface death receptors Trail-R1 and Trail-R2. Its role, like FasL in activation-induced cell death (AICD), has been demonstrated in immune system. However the mechanism of Trail induced apoptosis remains nuclear. In this report, the recombinant Trail protein was expressed and purified. The apoptosis-inducing activity and the regulation mechanism of recombinant Trail on Jurkat T cells were explored in vitro. Trypan blue exclusion assay demonstrated that the recombinant Trail protein actively killed Jurkat T cells in a dose-dependent manner. Trail-induced apoptosis in Jurkat T cells were remarkably reduced by Bcl-2 over expression in BcL-2 gene transfected cells. Treatment with PMA (phorbol 12-myristate 13-acetate), a PKC activator, suppressed nail-induced apoptosis in Jurkat T cells. The inhibition of apoptosis by PMA was abolished by pretreatment with Bis, a PKC inhibitor. Taken together, it was suggested that Bcl-2 over-expression and PMA activated PKC actively down-regulated the Trail-mediated apoptosis in Jurkat T cell.
