Transarterial embolization is a widely recognized clinical treatment method for liver *** that the soft and easily damaged features of embolic particles may limit tumor embolization efficiency,the present study carries out an attempt of fabricating tough and elastic microspheric gel for promoting embolization *** promote the toughness of hydrogel,poly(ethylene glycol)-co-poly(ε-caprolactone)-co-poly(ethylene glycol)(PPP)and PPP with two terminal double bonds(PPPDA)are co-assembled into nano-micelles,which are connected with methacrylated chitosan(CSMA)to fabricate microspheric gels via microfluidic *** double bond density of micelles promotes the freedom degree of micelles,significantly enhancing hydrogel *** compensate for the strength loss caused by the decrease of double bond density of micelles,phytic acid(PA)are employed to interact with CS to form a physical network,further improving hydrogel strength and *** CS-PPPDA&PPP-PA microspheric gels exhibit higher blocking effect in vitro.A rabbit VX2 liver metastasis tumor model is prepared to verify the embolization efficacy of CS-PPPDA&PPP-PA microspheric *** with clinical used microspheres,fewer CS-PPPDA&PPP-PA microspheric gels can achieve enough embolization *** embolization for 14 days,CS-PPPDA&PPP-PA microspheric gels exhibit improved tumor necrosis rate and promoted tumor cells apoptosis with reduced inflammation in surrounding tissues,confirming advanced embolic efficiency of tough microgels.

Objective:Traditional Chinese medicine plays a significant role in the treatment of the pandemic of coronavirus disease 2019(COVID-19).Tanreqing Capsule(TRQC)was used in the treatment of COVID-19 patients in the shanghai Public Health clinical *** study aimed to investigate the clinical efficacy of TRQC in the treatment of ***:A retrospective cohort study was conducted on 82 patients who had laboratory-confirmed mild and moderate COVID-19;patients were treated with TRQC in one designated *** treatment and control groups consisted of 25 and 57 cases,*** treatment group was given TRQC orally three times a day,three pills each time,in addition to conventional Western medicine treatments which were also administered to the control *** clinical efficacy indicators,such as the negative conversion time of pharyngeal swab nucleic acid,the negative conversion time of fecal nucleic acid,the duration of negative conversion of pharyngeal-fecal nucleic acid,and the improvement in the level of immune indicators such as T-cell subsets(CD3,CD4 and CD45)were ***:COVID-19 patients in the treatment group,compared to the control group,had a shorter negative conversion time of fecal nucleic acid(4 vs.9 days,P=0.047)and a shorter interval of negative conversion of pharyngeal-fecal nucleic acid(0 vs.2 days,P=0.042).The level of CD3+T cells increased in the treatment group compared to the control group([317.09±274.39]vs.[175.02±239.95]counts/l L,P=0.030).No statistically significant differences were detected in the median improvement in levels of CD4+T cells(173 vs.107 counts/l L,P=0.208)and CD45+T cells(366 vs.141 counts/l L,P=0.117)between the treatment and control ***:Significant reductions in the negative conversion time of fecal nucleic acid and the duration of negative conversion of pharyngeal-fecal nucleic acid were identified in the treatment group as compared to the control group,illustrating the potentia

Human pluripotent stem cells provide an inexhaustible model to study human embryogenesis in *** studies have provided diverse models to generate human blastoids by self-organization of different pluripotent stem cells or somatic reprogramming ***,whether blastoids can be generated from other cell types or whether they can recapitulate postimplantation development in vitro is ***,we develop a strategy to generate human blastoids from heterogeneous intermediates with epiblast,trophectoderm,and primitive endoderm signatures of the primed-to-naïve conversion process,which resemble natural blastocysts in morphological architecture,composition of cell lineages,transcriptome,and lineage differentiation *** addition,these blastoids reflect many features of human peri-implantation and pregastrulation development when further cultured in an in vitro 3D culture *** summary,our study provides an alternative strategy to generate human blastoids and offers insights into human early embryogenesis by modeling peri-and postimplantation development in vitro.

The current coronavirus disease 2019(COVID-19)pandemic caused by severe acute respiratory syndrome coronavirus(SARS-CoV-2)remains a threat to pregnant ***,the impact of early pregnancy SARS-CoV-2 infection on the maternal-fetal interface remains poorly ***,we present a comprehensive analysis of single-cell transcriptomics and metabolomics in placental samples infected with SARS-CoV-2 during early *** to control placentas,SARS-CoV-2 infection elicited immune responses at the maternal-fetal interface and induced metabolic alterations in amino acid and phospholipid profiles during the initial weeks ***,subsequent immune cell activation and heightened immune tolerance in trophoblast cells established a novel dynamic equilibrium that mitigated the impact on the maternal-fetal ***,the immune response and metabolic alterations at the maternal-fetal interface exhibited a gradual decline during the second *** study underscores the adaptive immune tolerance mechanisms and establishment of immunological balance during the first two trimesters following maternal SARS-CoV-2 infection.

Cardiac biological pacing(BP)is one of the future directions for bradyarrhythmias ***,cardiac pacemaker cells(PCs)used for cardiac BP are mainly derived from pluripotent stem cells(PSCs).However,the production of high-quality cardiac PCs from PSCs remains a ***,we developed a cardiac PC differentiation strategy by adopting dual PC markers and simulating the developmental route of ***,two PC markers,Shox2 and Hcn4,were selected to establish Shox2:EGFP;Hcn4:mCherry mouse PSC reporter ***,by stepwise guiding naïve PSCs to cardiac PCs following naïve to formative pluripotency transition and manipulating signaling pathways during cardiac PCs differentiation,we designed the FSK method that increased the yield of SHOX2^(+);HCN4^(+)cells with typical PC characteristics,which was 12 and 42 folds higher than that of the embryoid body(EB)and the monolayer M10 methods *** addition,the in vitro cardiac PCs differentiation trajectory was mapped by single-cell RNA sequencing(scRNA-seq),which resembled in vivo PCs development,and ZFP503 was verified as a key regulator of cardiac PCs *** PSC-derived cardiac PCs have the potential to drive advances in cardiac BP technology,help with the understanding of PCs(patho)physiology,and benefit drug discovery for PC-related diseases as well.

One of the most significant findings in recent stem cell research is the establishment of the induced pluripotent stem （iPS） cells, because they could have critical implications in both regenerative and repro- ductive medicine. Male gametes play a crucial role in transmitting genetic information to subsequent generations, and notably there are more and more patients with azoospermia, due to genetic and environmental factors. Recent advancements on generation of male gametes from human iPS cells would bring great promise to produce patient own male gametes for treating male infertility and provide an excellent platform for unveiling molecular mechanisms of male germ cell development.
