Objective This study aimed to clarify the intervention effect of salidroside(SAL)on lung injury caused by PM_(2.5) in mice and illuminate the function of SIRT1-PGC-1ɑ*** Specific pathogen-free(SPF)grade male C57BL/6 mice were randomly assigned to the following groups:control group,SAL group,PM_(2.5) group,SAL+PM_(2.5) *** the first day,SAL was given by gavage,and on the second day,PM_(2.5) suspension was given by intratracheal *** whole experiment consist of a total of 10 cycles,lasting 20 *** the end of treatment,blood samples and lung tissues were collected and *** of pathological changes in lung tissue using inverted microscopy and transmission electron *** expression of inflammatory,antioxidants,apoptosis,and SIRT1-PGC-1ɑproteins were detected by Western *** Exposure to PM_(2.5) leads to obvious morphological and pathologica changes in the lung of ***_(2.5) caused a decline in levels of antioxidant-related enzymes and protein expressions of HO-1,Nrf2,SOD2,SIRT1 and PGC-1ɑ,and an increase in the protein expressions of IL-6,IL-1β,Bax,caspase-9 and cleaved ***,SAL reversed the aforementioned changes caused by PM_(2.5) by activating the SIRT1-PGC-1α *** SAL can activate SIRT1-PGC-1ɑ to ameliorate PM2.5-induced lung injury.

OBJECTIVE: To investigate the underlying protein molecular mechanisms of "Qi stagnation and blood stasis syndrome"(QS) and "Qi deficiency and blood stasis syndrome"(QD), as two subtypes of coronary artery disease(CAD) in Traditional Chinese Medicine(tcm),following percutaneous coronary intervention(PCI).METHODS: In this study, a total of 227 CAD patients with QS and 211 CAD patients with QD were enrolled; all participants underwent PCI. Label-free quantification proteomics were employed to analyze the changes in serum in two subtypes of CAD patients before and 6 months after PCI, aiming to elucidate the intervention mechanism of PCI in treating CAD characterized by two different tcm ***: Biochemical analysis revealed significant changes in tumor necrosis factor-α, high density lipoprotein cholesterol, blood stasis clinical symptoms observation, and Gensini levels in both patient groups post-PCI; Proteomic analysis identified 79 and 95 differentially expressed proteins in the QS and QD patient groups, respectively, compared to their control *** C8 alpha chain, complement factor H,apolipoprotein H, apolipoprotein B, plasminogen,carbonic anhydrase 2, and complement factor Ⅰ were altered in both comparison groups. Furthermore,enrichment analysis demonstrated that cell adhesion and connectivity-related processes underwent changes in QS patients post-PCI, whereas lipid metabolism-related pathways, including the peroxisome proliferator-activated receptor signaling pathway and extracellular matrix receptor interaction, underwent changes in the QD *** protein-protein interaction network analysis further enriched 52 node proteins, including apolipoprotein B,lipoprotein(a), complement C5, apolipoprotein A4,complement C8 alpha chain, complement C8 beta chain,complement C8 gamma chain, apolipoprotein H,apolipoprotein A-Ⅱ, albumin, complement C4-B,apolipoprotein C3, among others. OBJECTIVE: To investigate the mechanisms of the effect of Actinidia chinensis polysaccharide (ACPS) on the invasion and metastasis of gastric cancer cells. METHODS: BGC-823-Luc gastric cancer cells stably transfected with a luciferase gene were used to establish an in situ transplanted tumor mouse model. A live mouse imaging system was used to observe tumor growth， and hematoxylin and eosin staining was applied to analyze tissue histopathology. Transwell and scratch wound assays were performed to examine the invasive and migratory ability of BGC-823 cells. Immunofluorescence， confocal microscopy， immunohistochemistry， and Western blot assays were used to analyze the expressions of the nuclear transcription factor-κB (NF-κB) signaling pathway and epithelial-mesenchymal transition (EMT)-related proteins. RESULTS: ACPS significantly inhibited the growth of subcutaneously transplanted BGC-823-Luc gastric cancer tumors in nude mice and reduced inflammatory cell infiltration in tumor tissues. ACPS inhibited Epidermal Growth Factor-induced invasion， migration， and morphological changes in the cytoskeleton of BGC-823 cells. ACPS inhibited gastric cancer EMT and decreased the expression of MMP-9， N-cadherinandp-NF-κB p65 in transplanted tumor tissues. ACPS promoted p65-NF-κB nuclear translocation， inhibited the expression of matrix metalloproteinases and vascular adhesion factors in BGC-823 cells， promoted the NF-κB inflammatory pathway metastasis-associated protein expression of phospho-IKK alpha/beta (p-IKKα/β) and p65， and inhibited the protein expression of IKKκα. CONCLUSIONS: ACPS inhibited gastric cancer invasion and metastasis both in vivo and in vitro， which evidenced the inhibition of gastric cancer EMT via regulating the NF-κB inflammatory pathway.

Angiotensin-converting enzyme 2(ACE2)has proven beneficial in attenuating diabetic cardiomyopathy(DCM)but has been found to be a substrate of a disintegrin and metalloprotease protein-17(ADAM17).However,whether ADAM17 plays a role in the pathogenesis and intervention of DCM is *** this study,we created cardiomyocyte-specific knockout of ADAM17(A17^(α-MHCKO))mice,and left ventricular dimension,function,pathology and molecular biology were assessed in ADAM17^(fl/fl) control,A17^(α-MHCKO) control,ADAM17^(fl/fl) diabetic and A17^(α-MHCKO) diabetic *** differentiated H9c2 cells and neonatal rat cardiomyocytes(NRCMs)were used to explore the molecular mechanisms underlying the effect of ADAM17 on *** results showed that protein expression and activity of ADAM17 were upregulated whereas the protein expression of ACE2 was downregulated in the myocardium of diabetic ***-specific knockout of ADAM17 mitigated cardiac fibrosis and cardiomyocyte apoptosis and ameliorated cardiac dysfunction in mice with *** analyses detected a number of genes enriched in metabolic pathways,in particular the AMPK signaling pathway,expressed differentially between the hearts of A17^(α-MHCKO)and ADAM17^(fl/fl)diabetic *** mechanism may involve activated AMPK pathway,increased autophagosome formation and improved autophagic flux,which reduced the apoptotic response in *** addition,hypoxia-inducible factor-1α(HIF-1α)might act as an upstream mediator of upregulated ADAM17 and ADAM17 might affect AMPK signaling viaα1 A-adrenergic receptor(ADRA1A).These results indicated that ADAM17 activity and ACE2 shedding were enhanced in DCM,which was reversed by cardiomyocyte-specific ADAM17 ***,inhibition of ADAM17 may provide a promising approach to the treatment of DCM.

AIM:To investigate the effect of electroacupuncture(EA)on the mitochondria-dependent apoptotic signaling pathway in the ciliary muscle of guinea pigs with negative lensinduced myopia(LIM).METHODS:Guinea pigs were randomly divided into normal control(NC)group,LIM group,LIM+SHAM acupoint(LIM+SHAM)group,and LIM+EA *** in the NC group received no intervention,while those in other three groups were covered with-6.0 diopter(D)lenses on right ***,animals in the LIM+EA group received EA at Hegu(LI4)combined with Taiyang(EX-HN5)acupoints,while those in the LIM+SHAM group were treated at sham *** treatments for 1,2,and 4wk,morphological changes in ciliary muscles were observed with hematoxylin and eosin(H&E)staining and nick end labeling(TUNEL),and the expression of the mitochondrial apoptotic signaling pathway-related molecules in ciliary muscles was measured by real-time quantitative polymerase chain reaction(qPCR)and Western ***,the adenosine triphosphate(ATP)contents were also determined in ciliary ***:Axial length increased significantly in the LIM and LIM+SHAM groups and decreased in the LIM+EA *** ciliary muscle fibers were broken and destroyed in both LIM and LIM+SHAM groups,whereas those in the LIM+EA group improved *** assay showed the number of apoptotic cells increased in the LIM and LIM+SHAM groups,whereas reduced in the LIM+EA *** contents showed a significant decrease in the LIM and LIM+SHAM groups,whereas increased after EA *** with the NC group,the dynamin-related protein 1(DRP1),Caspase3,and apoptotic protease activator 1(APAF1)levels were significantly increased in the LIM group and decreased in the LIM+EA ***:The results provide evidence of EA inhibiting the development of myopia by regulating the mitochondrial apoptotic signaling pathway.
