The expression ofβ-lactamase,particularly metallo-β-lactamase(MBL)in bacteria has caused significant resistance to clinically importantβ-lactam antibiotics,including life-saving carbapenems.Antimicrobial peptides(AMPs)have emerged as promising therapeutic agents to combat antibiotic resistance.However,the cytotoxic AMPs has been one of the major concerns for their applications in clinical practice.Herein,we report a novel cephalosporin-caged AMP,which shows significantly reduced cytotoxicity,hemolytic activity,and antibacterial activity but turns highly active against bacteria upon specific hydrolysis by the antimicrobial resistance-causativeβ-lactamase.Further investigations demonstrate thisβ-lactamaseactivatable AMP selectively inactivates resistant bacterial pathogens over susceptible bacteria.This strategy should be applicable to other AMPs as a potential solution for the treatment of infectious diseases caused byβ-lactamase-expressing pathogenic bacteria.

Understanding of the nephrotoxicity induced by drug candidates is vital to drug discovery and development.Herein,an in situ metabolomics method based on air flow-assisted desorption electrospray ionization mass spectrometry imaging(AFADESI-MSI)was established for direct analysis of metabolites in renal tissue sections.This method was subsequently applied to investigate spatially resolved metabolic profile changes in rat kidney after the administration of aristolochic acid I,a known nephrotoxic drug,aimed to discover metabolites associated with nephrotoxicity.As a result,38 metabolites related to the arginine-creatinine metabolic pathway,the urea cycle,the serine synthesis pathway,metabolism of lipids,choline,histamine,lysine,and adenosine triphosphate were significantly changed in the group treated with aristolochic acid I.These metabolites exhibited a unique distribution in rat kidney and a good spatial match with histopathological renal lesions.This study provides new insights into the mechanisms underlying aristolochic acids nephrotoxicity and demonstrates that AFADESI-MSI-based in situ metabolomics is a promising technique for investigation of the molecular mechanism of drug toxicity.

The plants of the genus Melodinus (Apocynaceae) are widely distributed, and have long been used in folk medicine for the treatment of various ailments such as meningitis in children and rheumatic heart diseases, hernia, infantile malnutrition, dyspepsia and testitis. Over 100 alkaloids together with flavonoids, lignans, steroids, terpenoids and coumarins have been identified in the genus, and many of these have been evaluated for biological activity. This review presents comprehensive information on the chemistry and pharmacology of the genus together with the traditional uses of many of its plants. In addition, this review discusses the structure-activity relationship of different compounds as well as recent developments and the scope for future research in this aspect.

10-（4-Formylphenyl）-5,15-bis（pentafluorophenyl）corrole 1 and its gallium complex 1-Ga were used to investigate their photocytotoxicity in liver cancer （BEL-7402）, lung cancer （A549） and cervical cancer （Siha, Hela） cell lines. Among all the tested tumor cells, Siha tumor cells were the most sensitive to photodynamic therapy （PDT） treatment by 1 and 1-Ga. PDT IC50 of 1 and 1-Ga toward Siha tumor cell reaches 1.3 and 0.8 gmol/L respectively, which is 10-fold higher than that of cisplatin. 1 and 1-Ga might pass across the tumor cell membrane smoothly without the help of carrier protein, and mainly localized in the cytoplasm of tumor cell. After PDT treatment, the intracellular reactive oxygen species （ROS） level increased and the mitochondrial membrane potential decreased significantly, which would finally result in the apoptosis of tumor cells.

Objective: Information on the comparative efficacy is important for drug development as well as drug therapy. Up to now, the relative efficacy of approved biologics and many agents under investigation in ankylosing spondylitis（AS） are still unclear. Several meta-analyses have been conducted to discover the potential difference among the available treatments for AS. Yet, no statistical difference among these treatments was observed and their relative clinical efficacy remains unknown. The common drawback of previous meta-analyses were that, primarily focusing on the single-end point data, they failed to take into account the varied time points of outcomes which many RCTs reported, thus leading to the inadequate utilization of the data. The objective of this study was to quantify the relative efficacy and time-course of various treatments measured by Ankylosing Spondylitis Assessment Study group response criteria 20（ASAS20）scores, change from baseline in Bath Ankylosing Spondylitis Disease Activity Index（BASDAI）and Bath Ankylosing Spondylitis Functional Index（BASFI）.Method: 34 double-blinded trials of 10 biologics and small molecules（adalimumab,certolizumab, etanercept, golimumab, infliximab, secukinumab, tocilizumab, sarilumab,tofacitinib, apremilast） encompassing 5,339 patients with AS were included in this analysis. Three mathematical models（ASAS20 model, ?BASDAI（change from baseline in BASDAI） and?BASFI（change from baseline in BASFI） model） with non-parametric placebo estimations were used to describe the longitudinal profile for the three efficacy measures.Baseline characteristics with relative rich information（percentage of male, disease duration, age, BASDAI, BASFI, CRP（C-reactive protein）） were included in our model as covariates to investigate their association with the drug effects. Model selection was based on the log likelihood ratio at an acceptance P value of 0.05.Results: The results detected significant differences among included treatments.Generally, the efficacy trend is similar across the three m

Liriope(Liliaceae) species have been used as folk medicines in Asian countries since ancient times. From Liriope plants(8 species), a total of 132 compounds(except polysaccharides) have been isolated and identified, including steroidal saponins, flavonoids, phenols, and eudesmane sesquiterpenoids. The crude extracts or monomeric compounds from this genus have been shown to exhibit anti-tumor, anti-diabetic, anti-inflammatory, and neuroprotective activities. The present review summarizes the results on phytochemical and biological studies on Liriope plants. The chemotaxonomy of this genus is also discussed.

Objective： To conduct a study on attitudes, knowledge, and use of complementary and altemative medicine （CAM） therapies in Serbia. Available data about CAM therapies in the region are scarce, opinions lacking from health sector. Balkan region countries had a delay in issuing national policies on CAM therapies. Methods： The questionnaire used was based on previously validated CAM Health Belief Questionnaire （CHBQ）, formulated as 5-item Likert type scale, adjusted for local environment. Health care students and professionals were evaluated. The questionnaire comprehended 10 closed questions on attitudes, knowledge and use of CAM therapies. This survey was conducted in eight cities of Serbia, January 2010-July 2011. A total of 797 participants was included. The second group of participants was 145 healthcare professionals （50 academic staff, 64 clinical staff, 19 pharmacists, 6 other clinical branch specialists and 6 nurses）. Data were collected by an interview. Examinees could acquire maximum of 70 points, 35 representing neutral attitude. Restdts： Students of dentistry （54.65 ± 6.07） were better informed on CAM therapies than medicine students （50.26 ± 7.92）. pharmacy students （51.16 ± 7.10） accepted low-scientific CAM. Pharmacists scored better than university professors （55.12±6.55 vs. 50.29± 9.50）. Primary health care professionals had better awareness than pharmacists in dispensing pharmacies. Both groups of participants preferred use of vitamins over any other CAM therapy. Conclusion： These pioneering efforts in the region exposed weaknesses in CAM attitudes of current and future health care professionals. Nevertheless, awareness on alternative medicine treatment choices is growing among Balkan prescribers. Supportive legal framework would facilitate dissemination of CAM medical practices.

Roscoea is an alpine or subalpine genus from the pan-tropical family Zingiberaceae,which consists of two disjunct groups in geography,namely the"Chinese"clade and the"Himalayan"clade.Despite extensive research on the genus,Roscoea species remain poorly defined and relationships between these species are not well resolved.In this study,we used plastid genomes of nine species and one variety to resolve phylogenetic relationships within the"Chinese"clade of Roscoea and as DNA super barcodes for species discrimination.We found that Roscoea plastid genomes ranged in length from 163,063 to 163,796 bp,and encoded 113 genes,including 79 protein-coding genes,30 tRNA genes,four rRNA genes.In addition,expansion and contraction of the IR regions showed obvious infraspecifc conservatism and interspecific differentiation.Plastid phylogenomics revealed that species belonging to the"Chinese"clade of Roscoea can be divided into four distinct subclades.Furthermore,our analysis supported the independence of R.cautleoides var.pubescens,the recovery of Roscoea pubescens Z.Y.Zhu,and a close relationship between R.humeana and R.cautloides.When we used the plastid genome as a super barcode,we found that it possessed strong discriminatory power(90%)with high support values.Intergenic regions provided similar resolution,which was much better than that of protein-coding regions,hypervariable regions,and DNA universal barcodes.However,plastid genomes could not completely resolve Roscoea phylogeny or definitively discriminate species.These limitations are likely related to the complex history of Roscoea speciation,poorly defined species within the genus,and the maternal inheritance of plastid genomes.

AIM: To compare rabeprazole (RPZ; 10 mg) with Lansoprazole orally disintegrating tablets (LPZ; 30 mg OD) in terms of antisecretory activity and blood drug concentration after a single dose. METHODS: Eight H pylori-negative cytochrome P450 (CYP) 2C19 extensive metabolizers were assigned to receive a single oral dose of RPZ 10 mg or LPZ 30 mg OD. Twelve hour intragastric pH monitoring was perform- ed on the day of treatment. Blood samples were also collected after the administration of each drug. RESULTS: LPZ 30 mg OD induced a significantly earlier rise in blood drug concentration than RPZ 10 mg; consequently, LPZ 30 mg OD induced a significantly earlier rise in median pH in the third and fourth hours of the study. CONCLUSION: In H pylori-negative CYP2C19 extensive metabolizers, LPZ 30 mg OD induced a significantly faster inhibition of gastric acid secretion than RPZ 10 mg.
