Molecular detection of virulence genes in Campylobacter species isolated from livestock production systems in South Africa

Campylobacter species are a major cause of foodborne bacterial infections in both developed and developing countries worldwide.Campylobacter jejuni is responsible for the majority of infectio ns.This study was con ducted to identify virule nceassociated genes in Campylobacter species isolated from livestock production systems in South Africa.A total of 250 fecal samples consisting of cattle(n=50),chickens(n=50),goats(n=50),sheep(n=50)and pigs(n=50)were randomly collected from livestock in Eastern Cape and KwaZulu-Natal provinces of South Africa between April and October 2018.The samples were an a lyzed for the presence of virule nee genes in Campylobacter species using molecular PCR-based methods.It was found that 77 and 23%of Campylobacter jejuni and Campylobacter coli respectively were isolated from all the livestock samples.There were positive significant(P<0.05)correlations amongst all the virulenee genes that were investigated.Chisquare and Fisher's exact tests were implemented to test for the effect of livestock species on the presenee or absenee of virule nee gen es.The study dem on strated that most of livestock species can pote ntially cause zoonotic infecti on s and food pois oning due to the high prevale nee of Campylobacter.The high prevale nee of virule nee genes highlights the sign ifica nee of Campylobacter\r\livestock production systems in South Africa.This requires the implementation of one-health approaches to reduce the impact of foodborne and zoonotic diseases for the welfare of human and animal health.

背景与目的:丙型肝炎(HCV)的传统治疗是应用聚乙二醇干扰素(FEG-IFN)和病毒唑,但副作用明显。IL28B基因多态性可以预测治疗反应,CC基因型者效果更好。ITPA基因缺失能对治疗引起的临床贫血起到保护作用。本研究目的是检测接受FEG-IFN和病毒唑治疗的1型HCV患者中IL28B基因多态性和ITPA基因突变情况,并分析其与持续病毒学应答(SVR)的相关性。方法:通过病历记录查询过去10年所有接受FEG-IFN和病毒唑治疗的1型HCV患者,通过信函邀请其参加本次研究。采集患者血液标本用以IL28B和ITPA基因检测。从病历记录中提取治疗反应、贫血发生及治疗中断情况。结果:共有61例1型HCV患者接受了FEG-IFN和病毒唑治疗,其中42例同意参加本次研究。治疗时平均年龄为(45.6±12.9)岁,男性患者占83.3%。33例(78.6%)检测为IL28B CC基因型,其中25例(75.8%)获得SVR,而9例非CC基因型患者中仅3例(33.3%)获得SVR(P=0.041)。11例患者检测为ITPA AC基因型,30例为CC基因型。ITPA AC与CC基因型患者的临床贫血发生率的差异无统计学意义(45.5%vs.63.3%,P=0.302)。即便在出现临床贫血的患者中,仍有70.8%最终获得了SVR。治疗中断对SVR亦无影响。结论:对于IL28B CC基因型的1型HCV患者,在考虑新的治疗选择前,应告知其FEG-IFN和病毒唑具有良好的治疗效果。

Simultaneous Quantification of κ-Carrageenan Oligosaccharides of DP 3, 5 and 7 by LC-MS/MS: Application to an in vitro Absorption Study

Carrageenans are widely utilized in many commercial applications such as the food and pharmaceutical industry, due to their excellent functional properties. In this study, a sensitive LC-MS/MS method was developed to determine κ-3, κ-5, and κ-7 carrageenan oligosaccharides simultaneously. Optimum MRM transitions for κ-3, κ-5, and κ-7 carrageenan oligosaccharides were(645.079→565.111, [M-H]-),(515.137→474.946, [M-2H]^2-), and(471.484→445, [M-3H]^3-), respectively. Chromatographic separation was performed on an Amide column coupled with a guard column operated at 60℃ under stepwise gradient elution. The linearity of the LC-MS/MS method for κ-3, κ-5, and κ-7 carrageenan oligosaccharides, evaluated over the concentration range of 0.10- 20.0 μmol L^-1, was excellent. The precisions of the method for κ-3, κ-5, and κ-7 carrageenan oligosaccharides were from 0.91% to 9.66%, and the inter-day precisions were from 0.92% to 10.5%. Validation of the LC-MS/MS method indicated that the method was precise and in line with the CFDA guidance. This method has been successfully applied to an in vitro absorption study.

Rapid detection and identification of Escherichia coli(E.coli)is essential to prevent its quickly spread.In this study,a novel fluorescence probe based on ZnTe quantum dots(QDs)modified by mannose(MAN)had been prepared for the determination of E.coli.The results showed that the obtained QDs showed excellent selectivity toward E.coli,and presented a good linearity in range of 1.0×10~5~1.0×10~8 CFU/mL.The optimum fluorescence intensity for detecting E.coli was found to be at pH 7.0 with a temperature of25℃and incubation time of 20 min.Under these optimum conditions,the detection limit of E.coli was4.6×10~4 CFU/mL.The quenching was discussed to be a static quenching procedure,which was proved by the quenching efficiency of QDs decreased with the temperature increasing.

Tailored nanocarriers and bioconjugates for combating glioblastoma and other brain tumors

Worldwide,the incidence of primary brain tumors is on the rise.Unfortunately,noninvasive drug therapy is hampered by poor access of most drugs to the brain due to the insurmountable blood-brain barrier(BBB).Nanotechnology holds great promise for noninvasive therapy of severe brain diseases.Furthermore,recent bioconjugation strategies have enabled the invasion of the BBB via tailored-designed bioconjugates either with targeting moieties or alterations in the physicochemical and/or the pharmacokinetic parameters of central nervous system(CNS)active pharmaceutical ingredients.Multifunctional systems and new entities are being developed to target brain cells and tumor cells to resist the progression of brain tumors.Direct conjugation of an FDA-approved drug with a targeting moiety,diagnostic moiety,or pharmacokinetic-modifying moiety represents another current approach in combating brain tumors and metastases.Finally,genetic engineering,stem cells,and vaccinations are innovative nontraditional approaches described in different patents for the management of brain tumors and metastases.This review summarizes the recent technologies and patent applications in the past five years for the noninvasive treatment of glioblastoma and other brain tumors.Till now,there has been no optimal strategy to deliver therapeutic agents to the CNS for the treatment of brain tumors and metastases.Intensive research efforts are ongoing to bring novel CNS delivery systems to potential clinical application.

Study on a Specific Site Tyr^(444) on a Hyperthermophilic Enzyme APE1547

Hyperthermophilic enzyme APE1547 is an extremely thermostable recombinant protein from thermophilic archaeon Aeropyrumpernix K1. The Tyr444 located in the catalytic domain adjacent to the catalytic amino acid Ser445 and formed hydrogen bond with Ile567. To study the effect of Tyr444 on the activity of APE1547, site-directed mutagenesis was applied. Two mutant enzymes T444S and T444G were created. Comparison of the mutant enzymes with wide enzyme, the thermostability of mutants T444S and T444G decreased by 10%-20%, but the catalytic efficiency of mutants toward pNPC8 and Ac-Leu-pNA increased 1.33 and 1.75 fold respectively. Molecular modeling shows that the elimination of hydrogen bond between Tyr444 and Ile567 is the cause of the decrease in thermostability and increase in catalytic efficiency. These observations suggest that Tyr444 plays an important role in the catalytic ability and thermostability of this enzyme.

Background:It seems to be numerous unclear black-box mechanisms of Chinese Medicines(CMs)with multiple bioactivities in the real-world clinical practice.Meanwhile,prior prediction is necessary before the implementation of pharmacodynamics-pharmacokinetics-based researches.With emergent ML techniques for TCM domain,Bayesian Network(BN)has shown its potentials for CM-bioactivity prediction and syndromes identification in Traditional Chinese Medicine(TCM),benefited from many advantages,such as flexibility in addressing,data-driven and probability-based inference under complex uncertainty.Although BN has been extensively used in TCM,the scarcity of researches on refining methodological features of BN-modelling for optimization poses a significant challenge.Our goal is to present methodological overview of BN-modelling for CM-bioactivities prediction towards pharmacology,which tends to acquire a sequence of intimations for boosting in-depth and optimized CM-BN collaboration based on detected gaps.Methods:We performed systematic search of 13 databases from their inception to November 10th 2022 regardless of language written,which excluded unindexed journals and clinical trial registries,using the 3 keywords(CM,Pharmacology,BN).And full-text original researches with the given subject were under consideration.Afterwards,selection of eligible studies,data refinement and inspection were totally conducted by 6 review authors.Results:A total of 7 studies involving 17 BN models were included for synthesis and refinement,based on existing literatures and databases with 2 modelling functions:regression and tagging.There were 3 prediction patterns:property-bioactivity,efficacy-bioactivity and constituent-bioactivity inference,covering 8 feature-utilized efficacies,5 feature-utilized properties and 10 feature-utilized constituents.Thereafter,without an independent validation dataset,established BNs were mostly utilized to predict the root-node probabilities of unknown data.Indeed,incomplete report on modelling samples,directed ac

Dental caries is one of the major oral health problems in most countries,affecting more than 50%of school children and majority of adults.Normally,we can prevent the disease by utilization of suitable antiseptics,e.***.alcohols and chlorhexidine[1].However,most antiseptics cause irritation or straining of the mucous membrane and teeth.Therefore,finding a more safe and efficient antiseptics is still a matter of interest to eradicate the causative pathogen.Volatile oils from plants including spearmint oil(SMO)could reduce the number of Streptococcus mutans counts in plaques and saliva.
