Effects of “Bioactive” amino acids leucine, glutamate, arginine and tryptophan on feed intake and mRNA expression of relative neuropeptides in broiler chicks

Feed intake control is vital to ensuring optimal nutrition and achieving full potential for growth and development in poultry. The aim of the present study was to investigate the effects of L-leucine, L-glutamate, L-tryptophan and L-arginine on feed intake and the mRNA expression levels of hypothalamic Neuropeptide involved in feed intake regulation in broiler chicks. Leucine, glutamate, tryptophan or arginine was intra-cerebroventricularly （ICV） administrated to 4d-old broiler chicks respectively and the feed intake were recorded at various time points. Quantitative PCR was performed to determine the hypothalamic mRNA expression levels of Neuropeptide Y （NPY）, agouti related protein （AgRP）, pro-opiomelanocortin （POMC）, melanocortin receptor 4 （MC4R） and corticotrophin releasing factor （CRF）. Our results showed that ICV administration of L-leucine （0.15 or 1.5 μmol） significantly （P〈0.05） increased feed intake up to 2 h post-administration period and elevated both hypothalamic NPY and AgRP mRNA expression levels. In contrast, ICV administration of L-glutamate （1.6 μmol） significantly （P 〈 0.05） decreased feed intake 0.25, 0.5 and 2 h post-injection, and increased hypothalamic CRF and MC4R mRNA expression levels. Meanwhile, both L-tryptophan （10 or 100 μg） and L-arginine （20 or 200 μg） had no significant effect on feed intake. These findings suggested that L-leucine and L-glutamate could act within the hypothalamus to influence food intake, and that both orexigenic and anorexigenic Neuropeptide genes might contribute directly to these effects.

LC-MS determination and pharmacokinetic study of salidroside in rat plasma after oral administration of suspensions of traditional Chinese medicine Erzhi Wan and Fructus Ligustri lucidi

A simple,rapid and sensitive liquid chromatography-mass spectrometry（LC-MS）method was developed for the determination of salidroside in rat plasma and study of its pharmacokinetics after oral administration of suspension of Erzhi Wan and Fructus Ligustri lucidi into Wistar *** sample of 200 μL was extracted with acetic ether-isopropanol（2∶1）and the extraction was performed on a Kromasil C18 column（150 mm×4.6 mm,5 μm）with the mobile phase of methanol-water（41∶59,v/v）within a run time of 6.0 *** analyte was monitored with positive electrospray ionization（ESI）by selected ion monitoring（SIM）*** target ions were m/z 323.05 for salidroside and m/z 411.05 for internal standard（IS）geniposide.A good linear relationship was obtained over the range of 5.0-500.0 ng/mL and the lower limit of quantification was 5.0 ng/*** validated method was successfully applied to the pharmacokinetic study of salidroside in rat plasma after oral administration of suspension of Erzhi Wan and Fructus Ligustri lucidi.

Determination and pharmacokinetic study of catechin in rat plasma by HPLC

A high performance liquid chromatographic method was developed and validated for the quantitative determination of catechin in rat plasma and its pharmacokinetic study after intragastric administration of Catechu and Xiongdanjiangre Wan into SD rats. Plasma samples were prepared by protein precipitation using methanol-5% aqueous zinc sulfate （70：30, v/v） as precipitant. Chromatographic separation was achieved on Hypersil Cl8 column （250 mm~ 4.6 mm, 10 pm） with acetonitrile-water-triethylamine （6：94：0.3, v/v/v, pH 4.0＋0.1, adjusted with phos- phoric acid） as mobile phase, followed by a UV detection at 207 nm. Good linearity was obtained over the range of 0.143-7.15 mg/L of catechin, with correlation coefficient of 0.9992. The method was simple, sensitive, accurate and reproducible and＇ has been successfully applied to the pharmacokinetic study of catechin in rat plasma.

Aridity index reflects the exchanges of energy and water between the land surface and the atmosphere,and its variation can be used to forecast drought and flood patterns,which makes it of great significance for agricultural *** ratio of potential evapotranspiration and precipitation is applied to analyse the spatial and temporal distributions of the aridity index in the Belt and Road region under the 1.5℃and 2.0℃global warming scenarios on the basis of outputs from four downscaled global climate *** results show that:(1)Under the 1.5℃warming scenario,the area-averaged aridity index will be similar to that in 1986-2005(around 1.58),but the changes vary *** aridity index will increase by more than 5%in Central-Eastern Europe,north of West Asia,the monsoon region of East Asia and northwest of Southeast Asia,while it is projected to decrease obviously in the southeast of West *** the seasonal scale,spring and winter will be more arid in South Asia,and the monsoon region of East Asia will be slightly drier in summer compared with the reference ***,West Asia will be wetter in all seasons,except winter.(2)Relative to 1986-2005,both areal averaged annual potential evapotranspiration and precipitation are projected to increase,and the spatial variation of aridity index will become more obvious as well at the 2.0℃warming *** the aridity index over the entire region will be maintained at approximately 1.57 as that in 1.5℃,the index in Central-Eastern Europe,north of West Asia and Central Asia will grow rapidly at a rate of more than 20%,while that in West Siberia,northwest of China,the southern part of South Asia and West Asia will show a declining *** the seasonal scale,the increase of the aridity index in Central-Eastern Europe,Central Asia,West Asia,South Asia and the northern part of Siberia in winter will be obvious,and the monsoon region in East Asia will be drier in both summer and autumn.(3)Under the scena

Application of prolonging small feeding volumes early in life to prevent of necrotizing enterocolitis in very low birth weight preterm infants

Objective:To study the effects of prolonging small feeding volumes early in life on the incidence of necrotizing enterocolitis(NEC)in very low birth weight(VLBW)preterm ***:A total of 128 VLBW infants who could not be breastfed were assigned into the experimental group(63 cases)and the control group(65 cases)using a random number *** experiment group was fed 12 mL/(kg·d)on day 1 which was increased to 24 mL/(kg·d)for the first 10 study *** control group was fed 12 mL/(kg·d)for the first 14e48 ***,the feeding volume increased by 24-36 mL/(kg·d)up to 140e160 mL/(kg·d)and maintained until the 10th day after *** incidence of feeding intolerance and NEC,duration of hospitalization,time to full enteral feedings,incidence of intrahepatic cholestasis,and the levels of gastrin and motilin in serum were ***:The incidence of feeding intolerance was significantly lower in the experimental group compared with the control group(15.87% vs.33.84%).There was a significant reduction in the incidence of NEC between the experimental and control groups(7.9% vs.16% in the control group).Conclusion:A protocol that prolongs small feeding volumes early in life can reduce the incidence and severity of NEC,but still warrants further study.

ANALYSIS OF INDICES FOR SEVERE CONVECTIVE WEATHER UNDER THE INFLUENCE OF TROPICAL CYCLONES IN JIANGSU

By statistical research on the occurrence pattern of severe convective weather in Jiangsu province under the influence of tropical cyclones within a 10-year period(from 2001 to 2010),this paper discovers that among different severe convective weather,the occurrence frequency of short-range heavy precipitation is the highest,thunderstorms and gales come in second,and general thunderstorms rarely happen,while hailstorms and tornadoes never *** statistical results also showed that within the research period there are 21 tropical cyclones(TCs) affecting the Jiangsu area and most of them are in the stage of weakening to tropical ***,through studying indices for relevant cases of severe convection,it is discovered that their thresholds are lower than that of previous research,which indicated that convective instability and energy accumulation can easily lead to severe convective weather eventually due to the influence of TCs.

Establishment and analysis of global gridded Tm-Ts relationship model

In ground-based GPS meteorology, Tm is a key parameter to calculate the conversion factor that can convert the zenith wet delay（ZWD） to precipitable water vapor（PWV）. It is generally acknowledged that Tm is in an approximate linear relationship with surface temperature Ts, and the relationship presents regional variation. This paper employed sliding average method to calculate correlation coefficients and linear regression coefficients between Tm and Ts at every 2°× 2.5° grid point using Ts data from European Centre for Medium-Range Weather Forecasts（ECMWF） and Tm data from ＂GGOS Atmosphere＂, yielding the grid and bilinear interpolation-based Tm Grid model. Tested by Tm and Ts grid data, Constellation Observation System of Meteorology, Ionosphere, and Climate（COSMIC） data and radiosonde data, the Tm Grid model shows a higher accuracy relative to the Bevis Tm-Ts relationship which is widely used nowadays. The Tm Grid model will be of certain practical value in high-precision PWV calculation.

Dynamic monitoring of plant cover and soil erosion often uses remote sensing data, especially for estimating the plant cover rate (vegetation coverage) by vegetation index. However, the latter is influenced by atmospheric effects and methods for correcting them are still imperfect and disputed. This research supposed and practiced an indirect, fast, and operational method to conduct atmospheric correction of images for getting comparable vegetation index values in different times. It tries to find a variable free from atmospheric effects, e.g., the mean vegetation coverage value of the whole study area, as a basis to reduce atmospheric correction parameters by establishing mathematical models and conducting simulation calculations. Using these parameters, the images can be atmospherically corrected. And then, the vegetation index and corresponding vegetation coverage values for all pixels, the vegetation coverage maps and coverage grade maps for different years were calculated, i.e., the plant cover monitoring was realized. Using the vegetation coverage grade maps and the ground slope grade map from a DEM to generate soil erosion grade maps for different years, the soil erosion monitoring was also realized. The results show that in the study area the vegetation coverage was the lowest in 1976, much better in 1989, but a bit worse again in 2001. Towards the soil erosion, it had been mitigated continuously from 1976 to 1989 and then to 2001. It is interesting that a little decrease of vegetation coverage from 1989 to 2001 did not lead to increase of soil erosion. The reason is that the decrease of vegetation coverage was chiefly caused by urbanization and thus mainly occurred in very gentle terrains, where soil erosion was naturally slight. The results clearly indicate the details of plant cover and soil erosion change in 25 years and also offer a scientific foundation for plant and soil conservation.
