Objective Multiple sclerosis（MS） is a multifocal, inflammatory demyelinating disease of the central nervous system. The regulation of inflammation is pivotal for preventing the development or reoccurrence of MS. The bias ratio of high-M1 versus low-M2 polarized microglia is a major pathological feature implicated in MS. The present study is to explore the role of long nonco.ing RNAs（lncRNAs） in regulating microglial polarization. Methods We used microarray screening to identify potential lncRNAs and verified the function of selected ones by q PCR, western and ELISA analysis after lentivirus mediated over-expression and interference. RNA immunoprecipitation, RNA pull-down and Ch IP were used to elucidate the mechanism. Results Our work showed that lncRNA GAS5 suppressed microglial M2 polarization and affected the survival and differentiation of co.cultured oligodendrocyte progenitor cells. Interference with GAS5 in transplanted microglia attenuated the progression of experimental autoimmune encephalomyelitis（EAE） and promoted remyelination in a lysolecithin-induced demyelination model. Higher levels of GAS5 were found in the microglia of aged mice and in amoeboid-shaped microglia in MS patients. Functional studies demonstrated that GAS5 suppressed IRF4 transcription by binding to PRC2 and inhibiting M2 polarization, thereby revealing a novel mechanism for the GAS5 function. co.clusion We identified GAS5 as the first lncRNA participating in regulating microglial polarization and involved in the pathogenesis of EAE and MS.

The HINT project is a four-co.ntry co.lective of Global heritage interpretation. HINT is a two-year project part funded Geoparks researching the uses and benefits of new technologies in by the European Leader Programme. The partners are Geopark Shetland （Sco.land）, Chablais Geopark （France）, North Pennines Area of Outstanding Natural Beauty （England） and Geopark de Hondsrug （The Netherlands）. Each partner has developed a pilot project to investigate the practicalities of interpreting and co.municating heritage, especially geological heritage, using a particular technology. An overview of each of the four pilot projects is given. The overall aim of the pilot projects is to develop best practice in using new technologies for heritage interpretation and to share information and ideas within the project partnership and with geoparks and heritage organizations around the world. This paper, developed by the HINT partners, aims to give an insight into the practical process of developing interpretive tools using new technologies. Some co.mon problems are highlighted and some ways in which these can be addressed are shown.
