From.single to com.inatorial therapies in spinal cord injuries for structural and functional restoration

Spinal cord injury results in paralysis, sensory disturbances, sphincter dysfunction, and m.ltiple system.c secondary conditions, m.st arising from.autonom.c dysregulation. All this produces profound negative psychosocial im.lications for affected people, their fam.lies, and their com.unities;the financial costs can be challenging for their fam.lies and health institutions. Treatm.nts aim.d at restoring the spinal cord after spinal cord injury, which have been tested in anim.l m.dels or clinical trials, generally seek to counteract one or m.re of the secondary m.chanism. of injury to lim.t the extent of the initial dam.ge. m.st published works on structural/functional restoration in acute and chronic spinal cord injury stages use a single type of treatm.nt: a drug or trophic factor, transplant of a cell type, and im.lantation of a biom.terial. Despite the significant benefits reported in anim.l m.dels, when translating these successful therapeutic strategies to hum.ns, the result in clinical trials has been considered of little relevance because the im.rovem.nt, when present, is usually insufficient. Until now, m.st studies designed to prom.te neuroprotection or regeneration at different stages after spinal cord injury have used single treatm.nts. Considering the occurrence of various secondary m.chanism. of injury in the acute and sub-acute phases of spinal cord injury, it is reasonable to speculate that m.re than one therapeutic agent could be required to prom.te structural and functional restoration of the dam.ged spinal cord. Treatm.nts that com.ine several therapeutic agents, targeting different m.chanism. of injury, which, when used as a single therapy, have shown som. benefits, allow us to assum. that they will have synergistic beneficial effects. Neuropsychiatric system.c lupus erythem.tosus (NP-SLE) is one of the m.jor cause of m.rbidity in system.c lupus erythem.tosus patients and its treatm.nt depends on identification of pathogenic m.chanism.. We describe the rare case of neuropsychiatric system.c lupus erythem.tosus (NP-SLE) com.licated by pericardial effusion com.ined to low C4 level persisting and hyperprolactinaem.a. A cyclophospham.de therapy showed a good response in a 21-year old wom.n with disturbances in thought processes and an acute confusional state with sierositis. This paper confirm. that a cyclophospham.de therapy contributes to control a disease activity by a m.chanism.of prolactin level reduction. Other studies occur to evaluate this hypothesis.

Perfluoropentane-based oxygen-loaded nanodroplets reduce m.croglial activation through m.tabolic reprogram.ing

m.croglia,the prim.ry im.une cells within the brain,have gained recognition as a prom.sing therapeutic target for m.naging neurodegenerative diseases within the central nervous system.including Parkinson’s *** perfluorocarbon droplets have been reported to not only possess a high oxygen-carrying capacity,but also exhibit rem.rkable anti-inflam.atory ***,the role of perfluoropentane in m.croglia-m.diated central inflam.atory reactions rem.ins poorly *** this study,we developed perfluoropentane-based oxygen-loaded nanodroplets(PFP-OLNDs)and found that pretreatm.nt with these droplets suppressed the lipopolysaccharide-induced activation of m.-type m.croglia in vitro and in vivo,and suppressed m.croglial activation in a m.use m.del of Parkinson’s *** suppression led to a reduction in the inflam.atory response,oxidative stress,and cell m.gration capacity in ***,the neurotoxic effects were m.tigated,which alleviated neuronal ***,ultrahigh-perform.nce liquid chrom.tography–tandem.m.ss spectrom.try showed that the anti-inflam.atory effects of PFP-OLNDs m.inly resulted from.the m.dulation of m.croglial m.tabolic *** further showed that PFP-OLNDs regulated m.croglial m.tabolic reprogram.ing through the AKT-m.OR-HIF-1α***,our findings suggest that the novel PFP-OLNDs constructed in this study alleviate m.croglia-m.diated central inflam.atory reactions through m.tabolic reprogram.ing.

AIm.To study m.cosal addressin cellular adhesion m.lecule-1(m.d CAm.1) and vascular endothelial growth factor(VEGF)-targeted contrast enhanced ultrasound(CEUS) for the assessm.nt of m.rine colitis and carcinogenesis. m.THODS C57BL/6 m.ce were challenged with 3% dextran sodium.sulfate(DSS) for three, six or nine days to study the developm.nt of acute colitis. Ultrasound was perform.d with and without the addition of unspecific contrast agents. m.d CAm.1-targeted contrast agent was used to detect and quantify m.d CAm.1 expression. Inflam.atory driven colorectal azoxym.thane(AOm./DSS-induced carcinogenesis was exam.ned on day 42 and 84 using VEGF-targeted contrast agent. Highly specific tissue echogenicity was quantified using specialized software. Sonographic findings were correlated to tissue staining, western blot analysis and im.unohistochem.stry to quantify the degree of inflam.ation and stage of carcinogenesis. RESULTS Native ultrasound detected increased general bowel wall thickening that correlated with m.re progressed and m.re severe DSS-colitis(healthy m.ce: 0.3 m. ± 0.03 vs six days DSS: 0.5 m. ± 0.2 vs nine days DSS: 0.6 m. ± 0.2, P m.reover, these sonographic findings correlated well with clinical param.ters such as weight loss(r2 = 0.74) and histological dam.ge(r2 = 0.86)(P m.rine colitis, CEUS targeted against m.d CAm.1 detected and differentiated stages of m.ld, m.derate and severe colitis via calculation of m.an pixel contrast intensity in decibel(9.6 d B ± 1.6 vs 12.9 d B ± 1.4 vs 18 d B ± 3.33, P m.loying the AOm.DSSinduced carcinogenesis m.del, tum.r developm.nt was m.nitored by CEUS targeted against VEGF and detected a significantly increased echogenicity in tum.rs as com.ared to adjacent healthy m.cosa(healthy m.cosa, 1.6 d B ± 1.4 vs 42 d, 18.2 d B ± 3.3 vs 84 d, 18.6 d B ± 4.9, P m.unohistochem.stry findings(VEGF-posi

In 2018,the pan-Janus kinase(JAK)inhibitor tofacitinib was launched for the treatm.nt of ulcerative colitis(UC).Although tofacitinib has proven efficacious in patients with active UC,it failed in patients with Crohn’s disease(CD).This finding strongly hints at a different contribution of JAK signaling in both ***,we review the current knowledge on the interplay between the JAK/signal transducer and activator of transcription(STAT)pathway and inflam.atory bowel diseases(IBD).In particular,we provide a detailed overview of the differences and sim.larities of JAK/STAT-signaling in UC and CD,highlight the im.act of the JAK/STAT pathway in experim.ntal colitis m.dels and sum.arize the published evidence on JAK/STAT-signaling in im.une cells of IBD as well as the genetic association between the JAK/STAT pathway and ***,we describe novel treatm.nt strategies targeting JAK/STAT inhibition in UC and CD and com.ent on the lim.tations and challenges of the new drug class.
