Genes and proteins expressed at different life cycle stages in the model protist Euplotes vannus revealed by both transcriptomic and proteomic approaches

Sexual reproduction first appeared in unicellular protists and has continued to be an essential biological process in almost all *** protists, which contain both germline and somatic genomes within a single cell, have evolved a special form of sexual reproduction called conjugation that involves mitosis, meiosis, fertilization, nuclear differentiation, genome rearrangement, and the development of unique cellular structures. The molecular basis and mechanisms of conjugation vary dramatically among ciliates, and many details of the process and its regulation are still largely unknown. In order to better comprehend these processes and mechanisms from an evolutionary perspective, this study provides the first comprehensive overview of the transcriptome and proteome profiles during the entire life cycle of the newly-established marine model ciliate Euplotes vannus. Transcriptome analyses from 14 life cycle stages(three vegetative stages and 11 sexual stages) revealed over 26,000 genes that are specifically expressed at different stages, many of which are related to DNA replication,transcription, translation, mitosis, meiosis, nuclear differentiation, and/or genome rearrangement. Quantitative proteomic analyses identified 338 proteins with homologs associated with conjugation and/or somatic nuclear development in other ciliates, including dicer-like proteins, Hsp90 proteins, RNA polymerase Ⅱ and transcription elongation factors, ribosomal-associated proteins, and ubiquitin-related proteins. Four of these homologs belong to the PIWI family, each with different expression patterns identified and confirmed by RT-qPCR,which may function in small RNA-mediated genome rearrangement. Proteins involved in the nonhomologous end-joining pathway are induced early during meiosis and accumulate in the developing new somatic nucleus, where more than 80% of the germline sequences are eliminated from the somatic genome. Antioxidative capacity and enzyme activity in Haematococcus pluvialis cells exposed to superoxide free radicals

The antioxidative capacity of astaxanthin and enzyme activity of reactive oxygen eliminating enzymes such as superoxide dismutase (SOD),peroxidase (POD),catalase (CAT) and ascorbate peroxidase (APX) were studied in three cell types of Haematococcus pluvialis exposed to high concentrations of a superoxide anion radical (O2ˉ).The results show that defensive enzymes and astaxanthin-related mechanisms were both active in *** during exposure to reactive oxygen species (ROS) such as Oˉ*** reacted with ROS much faster than did the protective enzymes,and had the strongest antioxidative capacity to protect against lipid *** defensive mechanisms varied significantly between the three cell types and were related to the level of astaxanthin that had accumulated in those ***-enriched red cells had the strongest antioxidative capacity,followed by brown cells,and astaxanthin-deficient green *** there was no significant increase in expression of protective enzymes,the malondialdehyde (MDA) content in red cells was sustained at a low level because of the antioxidative effect of astaxanthin,which quenched Oˉ2 before the protective enzymes could *** green cells,astaxanthin is very low or absent;therefore,scavenging of ROS is inevitably reliant on antioxidative ***,in green cells,these enzymes play the leading role in scavenging ROS,and the expression of these enzymes is rapidly increased to reduce excessive ***,because ROS were constantly increased in this study,the enhance enzyme activity in the green cells was not able to repair the ROS damage,leading to elevated MDA *** the four defensive enzymes measured in astaxanthin-deficient green cells,SOD eliminates Oˉ2,POD eliminates H2O2,which is a by-product of SOD activity,and APX and CAT are then initiated to scavenge excessive ROS.

Optimization and evaluation of a bottom substrate denitrification tank for nitrate removal from a recirculating aquaculture system

A bottom substrate denitrification tank for a recirculating aquaculture system was developed. The laboratory scale denitrification tank was an 8 L tank （0.04 m2 tank surface area）, packed to a depth of 5 cm with a bottom substrate for natural denitrifying bacteria. An aquarium pump was used for gentle water mixing in the tank; the dissolved oxygen in the water was maintained in aerobic conditions （e.g. 〉 2 mg/L） while anoxic conditions predominated only at the bottom substrate layer. The results showed that, among the four substrates tested （soil, sand, pumice stone and vermiculite）, pumice was the most preferable material. Comparing carbon supplementation using methanol and molasses, methanol was chosen as the carbon source because it provided a higher denitrification rate than molasses. When methanol was applied at the optimal COD：N ratio of 5：1, a nitrate removal rate of 4591 ± 133 mg-N/m2 tank bottom area/day was achieved. Finally, nitrate removal using an 80 L denitrification tank was evaluated with a 610 L recirculating tilapia culture system. Nitrate treatment was performed by batch transferring high nitrate water from the nitrification tank into the denitrification tank and mixing with methanol at a COD：N ratio of 5：1. The results from five batches of nitrate treatment revealed that nitrate was successfully removed from water without the accumulation of nitrite and ammonia. The average nitrate removal efficiency was 85.17% and the average denitrification rate of the denitrification tank was 6311 ± 945 mg-N/m2 tank bottom area/day or 126 ± 18 mg-N/L of pumice packing volume/day.

Immobilization of nitrite oxidizing bacteria using biopolymeric chitosan media

The effects of chitosan characteristics including the degree of deacetylation, molecular weight, particle size, pH pretreatment and immobilization time on the immobilization of nitrite-oxidizing bacteria （NOB） on biopolymeric chitosan were investigated. Nitrite removal efficiency of immobilized NOB depended on the degree of deacetylation, particle size, pH pretreatment on the surface of chitosan and immobilization time. Scanning electron microscope characterization illustrated that the number of NOB cells attached to the surface of chitosan increased with an increment of immobilization time. The optimal condition for NOB immobilization on chitosan was achieved during a 24-hr immobilization period using chitosan with the degree of deacetylation larger than 80% and various particle size ranges between 1-5 mm at pH 6.5. In general, the NOB immobilized on chitosan flakes has a high potential to remove excess nitrite from wastewater and aquaculture systems.

Differences in nitrite-oxidizing communities and kinetics in a brackish environment after enrichment at low and high nitrite concentrations

Nitrite accumulation in shrimp ponds can pose serious adverse effects to shrimp production and the *** study aims to develop an effective process for the enrichment of ready-to-use nitrite-oxidizing bacteria（NOB）inocula that would be appropriate for nitrite removal in brackish shrimp *** achieve this objective,the effects of nitrite concentrations on NOB communities and nitrite oxidation kinetics in a brackish environment were ***-bed biofilm sequencing batch reactors and continuous moving-bed biofilm reactors were used for the enrichment of NOB at various nitrite concentrations,using sediment from brackish shrimp ponds as seed *** results from NOB population analysis with quantitative polymerase chain reaction（q PCR）show that only Nitrospira were detected in the sediment from the shrimp *** the enrichment,both Nitrospira and Nitrobacter coexisted in the reactors controlling effluent nitrite at 0.1 and 0.5 mg-NO2^--N/*** the other hand,in the reactors controlling effluent nitrite at 3,20,and 100 mg-NO2^--N/L,Nitrobacter outcompeted Nitrospira in many orders of *** half saturation coefficients（Ks）for nitrite oxidation of the enrichments at low nitrite concentrations（0.1 and 0.5 mg-NO2^--N/L）were in the range of 0.71–0.98 mg-NO2^--N/*** contrast,the Ksvalues of NOB enriched at high nitrite concentrations（3,20,and 100 mg-NO2^--N/L）were much higher（8.36–12.20 mg-NO2^--N/L）.The results suggest that the selection of nitrite concentrations for the enrichment of NOB inocula can significantly influence NOB populations and kinetics,which could affect the effectiveness of their applications in brackish shrimp ponds.

Multi-year assessment of paralytic shellfish toxins in hard clam species along the coastline of Jiangsu Province, China

Paralytic shellfish toxins(PSTs) are notorious neurotoxins that threaten public health and food safety *** PST monitoring programs have recently been established throughout China, the profiles and variation of PSTs in important commercial clams(e.g., Mactra veneriformis, Ruditapes philippinarum, and Meretrix meretrix) along the Jiangsu Province coastline remain largely unexplored. In this study, a validated hydrophilic interaction liquid chromatography–tandem mass spectrometry(HILIC-MS/MS) method was used to examine PST profiles and levels in 540 clam samples from natural production areas along Jiangsu Province coastline during2014–2016. Although the PST levels(≤6.38 μg saxitotoxin equivalents(eq)/kg) were consistently below European Union regulatory limits(≤800 μg saxitotoxin eq/kg) during this time period, saxitotoxin, decarbamoylsaxitotoxin,and gonyautoxins 1 and 4 were detected, and nearly 40% of the samples were saxitotoxin-positive. The PST levels also varied significantly by seasons, with peak values observed in May during 2014–2016. This is the first systematic report of PSTs in clams from Jiangsu Province, and additional research and protective measures are needed to ensure the safety of clams harvested in this area.

Antitumor and Antifungal Activities of Organic Extracts of Seacucumber Holothuria atra from the Southeast Coast of India

In phylum Echinodermata, the family Holothuridae is distinguished by its capacity of bioactive compounds. Sea cu- cumber Holothuria atra is commonly known as the lollyfish. The antifimgal activity was detected using agar well diffusion method against the various fungal strains such as Trichoderma viride, Aspergillus niger, Aspergillus flavis, Candida albicans and Penicillium chrysogenum. Relatively high antifungal activity was seen against Candida albicans at 100 μL-1 concentration of extracts. Zone of inhibition was measured at 18 mm of diameter. The anti-tumor activities were detected against the Vero and Hep2 cell lines using MTT assay. The cells were treated with H. atra extract at concentrations 0.078-10mg mL-1. The extract showed high proliferative activity against the Hep2 cells. The body wall extracts of sea cucumber （H. atra） showed effective antifungal and antitumor activities All these findings suggest that the extracts could be used for the development of drugs.

Design of ionic liquids as liquid desiccant for an air conditioning system

Suitable control of the humidity can contribute to electric energy savings. However, the present dehumidification system has many weak points. The liquid desiccant air-conditioning system has recently gained growing interest from the stand point of reducing energy consumption during dehumidification. In order to find the appropriate ionic liquids(ILs) as a desiccant for the liquid desiccant air-conditioner system, we conducted a systematic evaluation of the humidification capability of 16 types of ILs. Among the tested ILs, tributyl(methyl)phosphonium dimethyl phosphate([P4441][DMPO_4]) exhibited the best dehumidification capacity and had a less corrosive effect on four types of metals as possible piping materials. It should be noted that this [P_(4441)][DMPO_4] has a very stable nature and produced no odor while conducting the experiment and storing for over 1 year at room temperature under ambient conditions. Furthermore, it was revealed that a 77%(w/w) aqueous solution of [P4441][DMPO_4] worked as an efficient desiccant liquid for the liquid desiccant air-conditioner system.

Symbiotic State of Chinese Land-marine Economy

The simultaneous development of both the land and marine-based economy is required to achieve the economic development of coastal regions in China. Based on symbiosis theory, this study discusses land and marine-based economic symbiosis mechanisms and uses a logistic symbiotic function to construct a symbiotic evolution model of the land-marine economy. We conduct a division and feature analysis of the interactive model between the land and marine-based economies of ll provinces （or cities） along the coast of China between 1996 and 2013, and discuss their economic development. The results show that, during the study period, the coordinated development model of the national land-marine economy is a parabiosis model. Fujian Province exhibits mutualism, Jiangsu, Hebei, Shandong, and Guangdong show commensalism, while Tianjin, Zhejiang, and Shanghai display parasitism, Guangxi and Hainan exhibit an antibiosis model, while Liaoning displays a parabiosis model. The land economic development model of Jiangsu and Fujian and the marine economic development model of Guangdong are improving the quality and efficiency of the factors of production. All other provinces＇ development models improve the productivity of the production factors to expand the scale of production.
