Zika virus(ZIKV)has been isolated from mosquitoes such as Aedes,Mansonia uniformis,and Culex perfuscus;However,the isolation of ZIKV from Anopheles sinensis and Culex tritaeniorhynchus has not yet been *** June and July2018,22,985 mosquitoes and 57,500 midges were collected in Jiangxi Province in southeastern *** them,six strains of ZIKV were isolated from mosquitoes:four from *** and two from *** genetic analysis showed that the ZIKV isolated from *** and *** belonged to genotype 2 in the Asian evolutionary branch of *** addition,the ZIKV strains isolated from *** and *** had amino acid substitutions identical to ZIKV strains prevalent in South America since *** study is the first to isolate ZIKV from mosquito specimens collected in the wild of Jiangxi Province,China;This is also the first time that ZIKV has been isolated from *** and *** that *** and *** have a very wide geographical distribution in China and even in eastern and southern Asia,the isolation of several strains of ZIKV from these two mosquitoes poses new challenges for the prevention and control of ZIKV infection in the mainland of China and countries and regions with the same distribution of mosquitoes.

Neutralizing Antibody Titer Test of Ebola Recombinant Protein Vaccine and Gene Vector Vaccine pVR-GP-FC

Objective In previous studies, we immunized mice with Ebola recombinant protein vaccine and gene vector vaccine. Both stimulated high levels of humoral immunity. In this work, we constructed a pseudovirus containing Ebola membrane proteins to verify whether the two immunization strategies can induce neutralizing antibodies in mice. Methods A pseudovirus containing an Ebola virus membrane protein based on the HIV-1 viral gene sequence was constructed and evaluated using a known neutralizing antibody. The titer of the neutralizing antibody in the sera of mice immunized with the recombinant protein and the gene vector vaccine was examined using a neutralization test. Results Ebola pseudovirus was successfully prepared and applied for neutralizing antibody detection. Immunological experiments showed that recombinant protein GP-Fc and gene vaccine pVR-modGP-Fc had good immunogenicity. The titer of the bound antibody in the serum after 8 weeks of immunization in mice was more than 1：105, and the recombinant protein induced greater humoral immunity. The results of the neutralization test based on the Ebola pseudovirus system demonstrated that both vaccines induced production of protective antibodies, while the gene vaccine induced a higher titer of neutralizing antibodies. Conclusion An Ebola pseudovirus detection system was successfully established and used to evaluate two Ebola vaccines. Both produced good immunogenicity. The findings lay the foundation for the development of new Ebola vaccines and screening for neutralizing monoclonal antibodies.

Pollution of HCHs,DDTs and PCBs in tidal flat of Hangzhou Bay 2009–2013

The concentration and distribution of three persistent organic pollutants （hexachlorocyclohexanes （HCHs）, dichlorodiphenyltrichloroethanes （DDTs） and polyehlorinated biphenyls （PCBs）） was assessed in tidal fiat sediments collected from the south bank of Hangzhou Bay, China from 2009 to 2013. Gas chromatography coupled to triple quadrupole mass spectrometry （GC-MS/MS） was used for analysis, based on United States Environmental Protection Agency methods EPAS080A, EPA8081B, and EPA3550B. The results showed that the levels of HCHs, DDTs and PCBs decreased in the order of DDTs 〉 HCHs 〉 PCBs, and their mass fractions ranged from 0.29-32.91, 0.09-13.19 and 0.16-4.10 ktg/kg （dry mass）, respectively. The levels of HCHs, DDTs and PCBs decreased slowly from 2009 to 2013, with considerably greater concentrations in winter than in spring and summer. In this study area, the concentrations of DDTs and HCHs decreased gradually towards the mouth of Hangzhou Bay, while the concentrations of PCBs were related to changes in the local economy. In addition, the sources of HCHs and DDTs were identified as atmospheric precipitation and historical residues, Finally, we predicted that PCBs pollution primarily originated from Aroclor 1254（Lot A4）, which might root in the illegal demolition and stacking of abandoned paint, transformer or electronic equipment in the south bank of Hangzhou Bay.

Re-isolation of Wuxiang Virus from Wild Sandflies Collected from Yangquan County, China

We previously isolated a new species of the genus Phlebovirus from wild sandflies collected from Wuxiang County in central China,which named the Wuxiang virus(WUXV).In this study,we re-isolated the WUXV from wild sandflies collected from two villages in Yangquan County,China in *** virus isolates that caused cytopathic effects in BHK-21 cells were successfully isolated from sandfly specimens collected from chicken pens and sheep *** analyses of the L,M and S gene segments of the viruses revealed that the four virus strains represented the previously isolated *** minimum infection rate(MIR)of the virus isolated from the sheep pen was 3.21,and the MIR of the virus isolated from the chicken pen was *** positive rates of Wuxiang virus neutralizing antibodies in serum samples of local healthy people and domestic chickens were 8.7%(4/46)and 100%(4/4),respectively,suggesting that Wuxiang virus can infect human and *** view of the fact that Wuxiang virus is infectious to humans and animals and has a relatively wide geographical distribution in China,it is of great public health significance to strengthen the investigation and study on the infection status of Wuxiang virus in humans and animals.

Development and application of a real-time polymerase chain reaction method for Campylobacter jejuni detection

AIM:To develop a real-time polymerase chain reaction(PCR) method to detect and quantify Campylobacter jejuni(***) from stool ***:Primers and a probe for real-time PCR were designed based on the specific DNA sequence of the hipO gene in *** specificity of the primers and probe were tested against a set of Campylobacter *** other enteric *** optimal PCR conditions were determined by testing a series of conditions with standard a *** *** detection limits were obtained using purified DNA from bacterial culture and extracted DNA from the stool *** hundred and forty-two specimens were analyzed for the presence of *** by direct bacterial culture and real-time ***:The optimal PCR system was determined using reference DNA templates,1 × uracil-DNA glycosylase,3.5 mmol/L MgCl 2,1.25 U platinum Taq polymerase,0.4 mmol/L PCR nucleotide mix,0.48 μmol/L of each primer,0.2 μmol/L of probe and 2 μL of DNA template in a final volume of 25 μ*** PCR reaction was carried as follows:95 ℃ for 4 min,followed by 45 cycles of 10 s at 95 ℃ and 30 s at 59 ℃.The detection limit was 4.3 CFU/mL using purified DNA from bacterial culture and 10 3 CFU/g using DNA from stool ***(8.3%,20/242) *** strains were isolated from bacterial culture,while 41(16.9%,41/242) samples were found to be positive by realtime *** sequencing of the PCR product indicated the presence of *** in the *** mixed infection of *** and Salmonella was detected in one specimen and the PCR test for this specimen was ***:The sensitivity of detection of *** from stool specimens was much higher using this PCR assay than using the direct culture method.

Fucoxanthin Isolated from Undaria pinnatifida Can Interact with Escherichia coli and lactobacilli in the Intestine and Inhibit the Growth of Pathogenic Bacteria

Fucoxanthin is a xanthophyll-type carotenoid that provides many benefits to human health. However, the mechanism by which fucoxanthin interacts with microbes and inhibits pathogenic bacteria is unknown. In this study, we investigated the effects of fucoxanthin isolated from the edible seaweed Undaria pinnatifida on pathogenic bacteria Escherichia coli and lactobacilli both in vitro and in vivo. Fucoxanthin strongly inhibited the growth of Gram-positive pathogenic bacteria but was less effective against Gram-negative bacteria. Fucoxanthin extracted from the crude mixture had a recovery rate of 93.38% and a purity of 82.70%, which were higher than those of fucoxanthin extracted using a previous method. Fucoxanthin also promoted the growth of intestinal mi- crobes in mice. Fucoxanthinol, a metabolite of fucoxanthin, was generated in the culture media. Fucoxanthin can be deacetylated into fucoxanthinol not only by conventional digestive enzymes in the digestive tract, but also by E. coli and lactobacilli in the intestine. These results indicate that fucoxanthin interacts with and influences E. coli and lactobacilli in the intestine. Therefore, fucoxanthin isolated from Undaria pinnatifida possibly can be applied in human health maintenance.

High Prevalence and Factors Contributing to Hyperhomocysteinemia, Folate Deficiency, and Vitamin B_(12) Deficiency among Healthy Adults in Shanghai, China

Elevated plasma or serum total homocysteine (tHcy) level has been established as a risk factor for cardiovascular disease[1], as well as dementia and cognitive decline[2]. Plasma or serum folate and vitamin B12 influence homocysteine (Hcy) metabolism as a co-substrate and cofactor respectively, so that low concentrations of folate and vitamin B12 are also associated with high Hcy levels[1]. However, not much information is available describing serum tHcy, folate, and vitamin B12 status in Shanghai adults, especially in a healthy population. Therefore, we hypothesize that low serum folate and vitamin B12 is associated with high Hcy in healthy adults in Shanghai. The aim of this study was to determine the status of serum tHcy, folate, and vitamin B12, and the prevalence and factors contributing to HHcy, folate deficiency, and vitamin B12 deficiency among healthy adults in Shanghai, China.
