MicroR-9c-5p and novel-mir50 co-target Akt to regulate Lasioderma serricorne reproduction

High fecundity is a common characteristic of insect pests which increases the difficulty of population control.Serine/threonine kinase Akt is an indispensable component of the insulin signaling pathway.Silencing of LsAkt severely hinders reproduction in Lasioderma serricorne,a stored product insect pest.However,the post-transcriptional pathway of LsAkt in L.serricorne remains unknown.This study identified 2 binding sites of miR-9c-5p and novel-mir50 in the coding sequences of LsAkt.The expression profiles of 2 microRNAs(miRNAs)and LsAkt displayed an opposite pattern during the adult stages.Luciferase reporter assay showed that novel-mir50 and miR-9c-5p could downregulate the expression of LsAkt.Overexpression of miR-9c-5p and novel-mir50 by injection of mimics inhibited the expression of LsAkt and reduced oviposition,decreased egg hatchability,and blocked ovarian development.It also decreased the expression of genes involved in ovarian development(LsVg and LsVgR)and the nutritional signaling pathway(LsTOR,LsS6K,and Ls4EBP),and reduced the phosphorylation of Akt.Conversely,injection of miR-9c-5p and novel-mir50 inhibitors induced the expressions of LsAkt,LsVg,LsVgR,LsTOR,LsS6K,and Ls4EBP,enhanced Akt phosphorylation level,and accelerated ovarian development.Injection of bovine insulin downregulated the expression of miR-9c-5p and novel-mir50 and upregulated the LsAkt expression.It also rescued the reproductive development defects associated with miR-9c-5p/novel-mir50 overexpression,forming a positive regulatory loop of insulin signaling.These results indicate that miR-9c-5p/novel-mir50 regulates the female reproduction of L.serricorne by targeting Akt in response to insulin signaling.The data also demonstrate the effects of the insulin/miRNA/Akt regulatory axis in insect reproduction.

The Effects of Fertilizers on Rabbiteye Blueberry(Vaccinium ashei Reade.)Root Distribution and Fruit Yield

The root system plays an important role in the growth and development of blueberry.The aim of this study was to assess the impacts of different fertilizers on the root growth and root–yield relationship of blueberry to provide insight into the regulation of root growth and fruit yield by fertilizing from the perspective of the root system.Rabbiteye blueberry variety‘Britewell’as the test material,and six fertilizers,including BF,OR,CF,SF,HF,and RT were used in single-factor fertilization experiments to analyze the effects of different fertilizer treatments on the root morphology,root distribution,and fruit yield of blueberry.Fertilization overall increased the root length density and root surface area in most soil layers,and the RT treatment significantly increased the total root length density and total root surface area 98.6%and 98.5%,respectively,compared with a control lacking fertilizer.In addition,the effect of fertilization on the blueberry root system was mainly observed in the 0–20 cm layer.Fruit yield was positively correlated with total root length density and total root surface area,and negatively correlated with average root diameter.In summary,the SF and RT treatments increased the morphological indexes of the root system,particularly in the shallow soil layers,leading to an increase in blueberry fruit yield.

Molting process revealed by the detailed expression profiles of RXR1/RXR2 and mining the associated genes in a spider mite, Panonychus citri

Spider mites have one ecdysone receptor(EcR)and multiple retinoid X receptors(RXRs).However,the function of these RXRs in spider mite development is unknown.Here,we screened the expression dynamics of two PcRXR isoforms at 4 h intervals in the deutonymphal stage of Panonychus citri.The results showed that PcEcR had an expression pattern similar to that of PcRXR2.For PcRXR1,its expression remained at a certain high level,when there was a decrease of both PcEcR and PcRXR2.In situ hybridization showed that PcRXR2 was detected in the central nervous mass,while the ecdysteroid biosynthesis gene PcSpo was mainly expressed at the edge of the central nervous mass.RNAi-based silencing of PcRXR1 or PcRXR2 showed the same phenotype as in mites with that of silencing PcEcR.Furthermore,RNA-seq was used to mine the genes associated with the expression dynamics of PcRXR1 or PcRXR2,which revealed that the heterodimer of EcR-RXR2 in spider mites might be linked with the cell autophagy and tissue remodeling during apolysis,and RXR1 might be linked with new epicuticle and exocuticle secretion during ecdysis.Taken together,these results increase our understanding of the regulation mechanism of ecdysteroid signal pathway in spider mite development.

Smurf participates in Helicoverpa armigera diapause by regulating the transforming growth factor-βsignaling pathway

Diapause,an important strategy used by insects to avoid adverse environments,is regulated by various cell signaling pathways.The results of our previous studies demonstrated that the transforming growth factor-β(TGF-β)signaling pathway regulated pupal diapause in Helicoverpa armigera,which was accompanied by downregulation of proteins in TGF-βsignaling.However,to date the mechanism underlying this phenomenon remains unknown.Here,we cloned the E3 ubiquitin ligases gene Smurf.In vitro experiments showed that Smurf directly bound to TGF-βreceptor type I(TGFβRI)and Smad2.Overexpressing Smurf promoted ubiquitination of TGFβRI and Smad2,thereby downregulating their protein levels.Conversely,silencing of the Smurf gene suppressed ubiquitination of TGFβRI and Smad2 thereby increasing their protein levels.Results from in vivo co-immunoprecipitation assays revealed that the binding of Smurf to TGFβRI or Smad2 was stronger in diapause pupae than in nondiapause pupae.Injection of Smurf inhibitor A01 into diapause pupae markedly upregulated expression of TGFβRI and Smad2 proteins,leading to resumption of development in diapause pupae.Taken together,these findings suggested that ubiquitin ligase E3 Smurf participated in H.armigera diapause by regulating TGF-βsignaling,and thus could be playing a crucial role in insect diapause.

Characterization and functional analysis of chitinase family genes involved in nymph-adult transition of Sogatella furcifera

Chitinase degrades chitin in the old epidermis or peritrophic matrix of insects,which ensures normal development and metamorphosis.In our previous work,we comprehensively studied the function of SfCht7 in Sogatella furcifera.However,the number and function of chitinase genes in S.furcifera remain unknown.Here,we identified 12 full-length chitinase transcripts from S.furcifera,which included nine chitinase(Cht),two imaginal disc growth factor(IDGF),and one endo-β-N-acetylglucosaminidase(ENGase)genes.Expression analysis results revealed that the expression levels of eight genes(SfCht3,SfCht5,SfCht6-1,SfCht6-2,SfCht7,SfCht8,SfCht10,and SfIDGF2)with similar transcript levels peaked prior to molting of each nymph and were highly expressed in the integument.Based on RNA interference(RNAi),description of the functions of each chitinase gene indicated that the silencing of SfCht5,SfCht10,and SfIDGF2 led to molting defects and lethality.RNAi inhibited the expressions of SfCht5,SfCht7,SfCht10,and SfIDGF2,which led to downregulated expressions of chitin synthase 1(SfCHS1,SfCHS1a,and SfCHS1b)and four chitin deacetylase genes(SfCDA1,SfCDA2,SfCDA3,and SfCDA4),and caused a change in the expression level of two trehalase genes(TRE1 and TRE2).Furthermore,silencing of SfCht7 induced a significant decrease in the expression levels of three wing development-related genes(SfWG,SfDpp,and SfHh).In conclusion,SfCht5,SfCht7,SfCht10,and SfIDGF2 play vital roles in nymph–adult transition and are involved in the regulation of chitin metabolism,and SfCht7 is also involved in wing development;therefore,these genes are potential targets for control of S.furcifera.

A water mite species is described as new to science, Atractides （Tympanomegapus） aprojectus sp. nov. The male ofAtractides （Tympanomegapus） huaxiensis Yi ＆ Jin, 2010 is described for the first time from Guizhou Province, China. The materials were collected during a survey in 2011 from the rivers in Guizhou. The specimens are deposited in the Institute of Entomology, Guizhou University （GUGC）.

In vivo and in vitro antiproliferative and antimetastatic effects of hemolymph of Aspongopus chinensis Dallas on breast cancer cells

OBJECTIVE:To investigate the effects of Aspongopus(A.)chinensis hemolymph on the proliferation and metastasis of breast cancer cells.METHODS:The in vitro effects of A.chinensis hemolymph were investigated in murine(4T1)and human(HCC1937)breast cancer cell lines.Cytotoxicity,cell apoptosis,and cell migration were evaluated by using the cell counting kit-8 assay,Hoechst staining,and wound healing experiments,respectively.A syngeneic mouse model was established to evaluate the in vivo effects of the hemolymph extract on tumor growth and metastasis.Mouse body weight,tumor size,blood levels of function-related enzymes,and pathological features of the liver and kidney tissues were evaluated.RESULTS:The hemolymph of A.chinensis significantly inhibited in vitro tumor cell migration and viability while inducing apoptosis.Furthermore,it inhibited in vivo tumor growth and metastasis with a minimal effect on mouse body weight,and did not induce liver or kidney damage.CONCLUSION:Our results suggested that the A.chinensis hemolymph has antitumorigenic properties,suggesting it has potential as a novel therapeutic option for the treatment of metastatic breast cancer.

First record of Cubachipteria(Acari:Oribatida:Achipteriidae) in China,with descriptions of two new species

The genus Cubachipteria of Achipteriidae is firstly reported from China. Two new species of C. pianmaensis sp. nov. and C. gaoligongensis sp. nov., are described here, which are collected on moss and leaf litter from Yunnan Province, China. These two species can be distinguished from all known species of Cubachipteria by the absence of interlamellar setae. A key to the known species of Cubachipteria is given. All type materials are deposited in Guizhou University, Guiyang City, China （GUGC）.
