Whole-exome mutational landscape of metastasis in patient-derived hepatocellular carcinoma cells

In order to explore the genomic basis for liver cancer metastasis,whole-exome sequencing(WES)was performed on patient-derived hepatocellular carcinoma(HCC)cell lines with differential metastatic potentials and analyzed their clonal evolution *** evolutionary tree based on genomic single nucleotide polymorphism(SNP)was constructed in MegaX *** WES data showed that the average percentage of heterogeneous mutations in each HCC cell lines was 16.55%(range,15.38%e18.17%).C:G>T:A and T:A>C:G somatic transitions were the two most frequent *** these metastatic HCC cell lines,non-silent gene mutations were found in 21.88%of known driver genes and 10 classical signaling *** protein interaction network was constructed by STRING,and hub genes were found in the shared trunk mutation genes and the heterogeneous branch mutations *** cBioPortal database,some of the selected hub genes were found to be associated with poor overall survival(OS)of HCC *** the mutated HCC driver genes,a novel KEAP1 mutation with a homozygous frameshift truncation at the c-terminal Nrf2 binding region was detected and verified in MHCC97-H and HCC97LM3 *** conclusion,WES data demonstrate that HCC cell lines from tumor biopsy specimens of the same patient have obtained different metastatic potentials through repeated selection in rodents in vivo,and they do indeed have a genetic relationship at the genomic level.

Forecasting the number of zoonotic cutaneous leishmaniasis cases in south of Fars province, Iran using seasonal ARIMA time series method

Objective: To predict the trend of cutaneous leishmaniasis and assess the relationship between the disease trend and weather variables in south of Fars province using Seasonal Autoregressive Integrated Moving Average(SARIMA) model,Methods: The trend of cutaneous leishmaniasis was predicted using Mini tab software and SARIMA model,Besides,information about the disease and weather conditions was collected monthly based on time series design during January 2010 to March 2016,Moreover,various SARIMA models were assessed and the best one was selected,Then,the model's fitness was evaluated based on normality of the residuals' distribution,correspondence between the fitted and real amounts,and calculation of Akaike Information Criteria(AIC) and Bayesian Information Criteria(BIC),Results: The study results indicated that SARIMA model(4,1,4)(0,1,0)(12) in general and SARIMA model(4,1,4)(0,1,1)(12) in below and above 15 years age groups could appropriately predict the disease trend in the study area,Moreover,temperature with a three-month delay(lag3) increased the disease trend,rainfall with a four-month delay(lag4) decreased the disease trend,and rainfall with a nine-month delay(lag9) increased the disease trend,Conclusions: Based on the results,leishmaniasis follows a descending trend in the study area in case drought condition continues,SARIMA models can suitably measure the disease trend,and the disease follows a seasonal trend.

1 Summary The 2015 Nobel Prize in Physiology or Medicine was awarded to professor Tu YouYou for her key contributions to the discovery of artemisinin. Artemisinin has saved mil- lions of lives and represents one of the significant contribu- tions of China to global health. Many scientists were in- volved in the previously unknown 523 Project, and the No- bel Prize given to a single person has not been without con- troversy. Here we summarize some key events in the 523 Project and present our views on the Award to help the pub- lic better understand the rationale of the Nobel Committee＇ s decision, the significance of the discovery, and current is- sues related to artimisinin in treating malaria.

Prevalence of depression and anxiety among children with type 1 and type 2 diabetes:a systematic review and meta-analysis

Background Evaluation of psychiatric disorders in children is essential in timely *** individual studies,there is no information on the exact status of psychiatric disorders in *** present study was conducted to determine the prevalence of depression among children with type 1 and type 2 *** This meta-analysis is registered with PROSPERO(CRD42021231491).Several databases(Web of Science,Scopus,and PubMed)were searched from January 1,2000 to December 15,*** key words included:depression,anxiety,children,and *** steps of search,comprising screening,risk of bias,and extraction of study data,were performed separately by two *** It was found that a total of 109 studies had been conducted involving 52,493 children with type 1 and type 2 *** studies were included in the present review *** prevalence of depression,anxiety among children with type 1 diabetes were 22.2%[95%confidence interval(CI)19.2–25.2],and 17.7%(95%CI 15.2–20.1),*** prevalence of depression was higher among girls(29.7%)than boys(19.7%).The prevalence of depression was higher in lower-middle-income countries at 29.3%(95%CI 18.6–40.0).The prevalence of depression among children with type 2 diabetes was 22.7%(95%CI 17.3–28.0).Conclusion The results of the present study indicate the importance of paying attention to extensive periodic screening and appropriate activities to reduce pediatric depression.

INTRODUCTIONIn China, liver fibrosis in most patients resulted from the viruses of hepatitis B. Both anti-virus and anti-fibrosis should be considered in designing a program for the treatment of liver fibrosis. Therefore, 40 cases of liver fibrosis due to hepatitis B were treated by using IFN-α1 and traditional medicinal preparations from February 1994 to April 1996. Good curative effect was achieved.

The cephalosporinsare the most widely used antibiotics in theUnited States and account for almost$ 1. 6 billion in U. S. health-care dollarsin 1987, cephalosporins commanded 55%of the inpatient/parenteral market and40% of the outpatient/oral ***, gram-negative organisms suchaspseudomonas aeruginosa and Serratiaspp. may be resistant to the older cepha-losporins, and Enterococcus, methicillin-resistant Staphylococcus *** Candidaalbicans are resistant to all *** troublesome pathogens now causean increasing numbers of nosocomialinfections and superinfections in patientswho have received prolonged antibiotictreatment Unfortunately, nosocomial infe-ctions often affect the growing number ofimmunocompromised patients. In the future,nosocomial infections will be more frequentlyassociated with the emergnce of more resistantbacterial strains. Oral monotherapy alternativesto parenteral cephalosporin therapy arebeing tested, and if proven useful mayhelp reduce the upward trend in antibioticcosts.

Pleiotropic Effect of tatC Mutation on Metabolism of Pathogen Yersinia enterocolitica

Objective To analyze the impact of depletion of the twin arginine translocation （TAT） system on virulence and physiology of Yersinia enterocolitica for a better understanding of its pathogenicity. Methods We constructed a △tatC：：Sp^R mutant of Yersinia enterocolitica by P1 phage mediated transduction using Escherichia coli K-12 △tatC：：Sp^R strain as a donor. Results A Pl-mediated genetic material transfer was found between the two species of enterobacteria, indicating a great potential of acquisition of antibiotic resistance in emergency of a new threatening pathogen by genetic material exchanges. Periplasmic trimethylamine N-oxidase reductase activity was detected in the wild type E enterocolitica strain and translocation of this enzyme was completely abolished by the △tatC：：Sp^R mutation. In addition, the △tatC：：Sp^R mutation showed a pleiotropic effect on the metabolism of E enterocolitica. However, the tat mutation did not seem to affect the mobility and virulence of Y. enterocolitica under the conditions used. Conclusion Unlike other pathogenic bacteria studied, the TAT system of E enterocolitica might play an important role in the pathogenic process, which is distinct from other pathogens, such as Pseudomonas aeruginosa and enterohemorrhagic E. coli O 157：H7.

Objectives： To review the calendar, mortality and morbidity of Iraq-Iran chemical war among Iranians based on researchers＇ reports. Methods： We used national and international databanks such as PubMed, ISI, Scopus, Irandoc and Iranmedex and studied 350 articles related to chemical agents and their effects on different organs. The main criteria for qualification of articles were relevancy orientation and being published in approved medical journals. Results： The Iraqi army invaded to west and southwest Iran using chemical weapons such as nerve agents （NAs） and sulfur mustard （SM）. Most victims were civilians including women and children. These attacks had imposed more than 150 types of diseases and complications on Iranians and the frequency of death was 2%-3%. Most reports were about respiratory problems and a few were in the domain of socio-economic damages Conclusion： At present, 25 years after me end of war, the victims are faced with different complications induced by chemical agents and it is estimated that they will be continuously troubled by these problems in future.

Prevalence and genotype distribution of hepatitis B virus among migrant workers in Lombok Island, Indonesia

Objective:To examine the potential risk of hepatitis B virus(HBV)spread in Indonesia by migrant workers,based on the molecular characteristics of HBV ***:Sera collected from migrant workers traveling to their destination countries(pre-migrant workers)and those returning to Indonesia(post-migrant workers)were screened for HBsAg by ELISA,followed by HBV DNA detection by PCR and(sub)genotype/subtype determination according to surface region and whole genome ***:Of 87 pre-migrant workers,15(17.24%)were HBsAgpositive,whereas 15(12.10%)of 124 post-migrant workers were HBs Ag *** genotype analysis based on the S region showed that HBV-B3/adw2 was predominant(96.15%,25/26)whereas 3.85%(1/26)of isolates were HBV-C3/adrq+.Whole genome sequencing of selected strains and phylogenetic tree analysis identified subgenotype B7 in three samples previously categorized as subgenotype B3 based on S region analysis,supporting a recent argument that subgenotypes B5/B7/B8/B9 could be considered as a quasi-subgenotype of ***:A high prevalence of HBsAg carriers was detected among migrant workers from Lombok Island,with no significant difference in prevalence between before and after returning to *** strains were classified into genotypes common in Indonesia,and the results suggested that migrant workers are not a risk factor for HBV transmission into Indonesia.
