IL-13/IL-13RA2 signaling promotes colorectal cancer stem cell tumorigenesis by inducing ubiquitinated degradation of p53

Cancer stem cells(CSCs)are considered tumor-initiating cells and the main drivers of disease *** these rare cancer cells,however,remains challenging with respect to therapeutic ***,we report the up-regulation of IL-13RA2 expression in colorectal cancer(CRC)tissues and spheroid *** expression of IL-13RA2 was positively correlated with canonical stemness markers in *** further demonstrated that the level of IL-13 was up-regulated in the serum of CRC ***,recombinant IL-13(rIL13)stimulation promoted the sphere formation,proliferation,and migration of CRC cells in vitro and enhanced tumorigenesis in *** phenotype could be reversed by knocking down ***,IL-13 activated autophagy by inducing LC3I/LC3II transformation in CRC-CSCs,which was crucial for the biological functions of *** further demonstrated that IL-13RA2 acted as a modular link of the E3 ligase UBE3C and the substrate p53 protein,enhancing the interaction of UBE3C and p53,thereby inducing the K48-linked ubiquitination of *** conclusion,the IL-13/IL-13RA2 signaling cascade promotes CRC-CSC self-renewal and tumorigenesis by inducing p53 ubiquitination,adding an important layer to the connection between IL-13 and p53,which can be translated into novel targeted therapies.

Self-aggregating behavior of poly(4-vinyl pyridine)and the potential in mitigating sand production based onπ-πstacking interaction

Unconsolidated sandstone reservoirs are most susceptible to sand production that leads to a dramatic oil production *** this study,the poly(4-vinyl pyridine)(P_(4)VP)incorporated with self-aggregating behavior was proposed for sand migration *** P_(4)VP could aggregate sand grains spontaneously throughπ-πstacking interactions to withstand the drag forces *** influential factors on the self-aggregating behavior of the P_(4)VP were evaluated by adhesion force *** adsorption as well as desorption behavior of P_(4)VP on sand grains was characterized by scanning electron microscopy and adhesion force test at different pH *** result indicated that the pH altered the forms of surface silanol groups on sand grains,which in turn affected the adsorption process of P_(4)*** spontaneous dimerization of P_(4)VP molecules resulting from theπ-πstacking interaction was demonstrated by reduced density gradient analysis,which contributed to the self-aggregating behavior and the thermally reversible characteristic of the P_(4)*** sand stabilization test revealed that the P_(4)VP showed wide pH and temperature ranges of *** production of sands can be mitigated effectively at 20-130℃ within the pH range of 4-8.

Study on the Networks of "Nature-Family-Component" of chinese Medicinal Herbs Based on Association Rules Mining

Objective： To explore appropriate methods for the research of the theory of chinese medicine nature property and find the relationship between Nature-Family-Component of chinese herbs. Methods： From perspective of systems biology, we used Associate Network to identify useful relationships among ＂Nature- Family-Component＂ of Herbs. In this work, Associate Network combines association rules mining method and network construction method to evaluate the complicate relationship among ＂Nature-Family-Component＂ of herbs screened. Results： The results of association rules mining showed that the families had a close relationship with nature properties of herbs. For example, the families of Magnoliaceae, Araceae had a close relationship with hot nature with confidence of 100%, the families of Cucurbitaceae has a close relationship to cold nature with confidence of 90,91%. Moreover, the results of constructed Associate Network implied that herbs belonging to the same families generally had the same natures. In addition, some herbs belonging to different families may also have same natures when they contain the same main components. Conclusion： These results implied that the main components of herbs might affect their natures; the relationships between families and natures were based on the main compounds of herbs.
