An accurate and validated liquid chromatography method and a triple quadrupole mass spectrometry method were developed and validated to simultaneously evaluate the cytochrome P450(CYP)enzymes(CYP1A2,CYP2C11,CYP2D6 and CYP3A1)in vivo using the co-administration of these ***(CYP1A2),losartan(CYP2C11),metoprolol(CYP2D4)and midazolam(CYP3A1)were used as the probe *** purpose of the study was to investigate the effect of apatinib on the cytochrome P450 enzymes(CYP1A2,CYP2C11,CYP2D4 and CYP3A1)in vivo with co-administration of these *** samples were prepared by precipitating protein with *** analytes were separated using a reversed-phase BEH C18 column(2.1*100 mm,1.7μm,Waters,USA)maintained at 40°*** mobile phase consisted of acetonitrile and water(containing 0.1%formic acid)with a gradient elution pumped at a flow rate of 0.4 m L/*** analytes were detected with positive electrospray ionization in multiple reaction monitoring(MRM)mode for target fragment ions m/z 180.05→109.94 for phenacetin,m/z 423.1→207.2 for losartan,m/z268.12→115.8 for metoprolol,m/z 326.02→290.99 for midazolam and m/z 285.1→193.1 for diazepam(IS).Good linearity was achieved to quantify the concentration ranges of 10-2000 ng/m L for phenacetin,10-1000 ng/m L for losartan,10-1000 ng/m L for metoprolol and 1-100 ng/m L for midazolam in rat *** mean recoveries of phenacetin,losartan,midazolam and metoprolol from the plasma exceeded 77.07%.The intra-run and inter-run assay precisions were both less than 8.9%.This method was successfully applied to evaluate the effects of apatinib on the cytochrome P450 enzymes in rats.

OBJECTIVE: To determine the therapeutic effect of Ziyin Jianghuo Ningxin Decoction(ZYJHNXD) plus dehydroepiandrosterone(DHEA) and menopausalhormone therapy(MHT) in patients suffering from menopausal symptoms identified as, in terms of Traditional Chinese Medicine, symptom pattern of Yin deficiency with hyperactive ***: Totally 180 postmenopausal women aged 40 to 60 years were assigned into four groups and accepted femoston, femoston with ZYJHNXD,femoston with DHEA, femoston with ZYJHNXD and DHEA therapies, respectively, for three *** questionnaire-based measure instruments included modified Kupperman index(MKI),Hamilton Rating Scale for Anxiety(HAMA), and Hamilton Rating Scale for Depression(HAMD). Follicle-stimulating hormone(FSH), luteinizing hormone(LH), estradiol(E2), 5-hydroxyindole-3-acetic acid(5-HIAA), norepinephrine(NE), dopamine(DA),bone mineral density(BMD), and sleep quality were evaluated before and three months after the ***: In all four groups, the scores of MKI, HAMA, HAMD and the levels of FSH, LH decreased significantly(P < 0.05) after the treatment, while the levels of E2, 5-HIAA, NE, and DA showed obvious elevation(P < 0.05). The group receiving ZYJHNXD and DHEA combined with femoston had superiority in the preservation of bone mineral density and improvement of total sleep time and nighttime sleep time over the other three ***: ZYJHNXD and DHEA combined with MHT therapy have a favorable outcome in managing menopausal symptoms, restoring hormone levels, preventing skeletal rarefaction or osteoporosis,and improving sleep quality for postmenopausal women.

Objective: To explore the mechanism of Chuanxiong in alleviating hypoxic pulmonary hypertension in rats by inhibiting pulmonary vascular remodeling. Methods: Thirty healthy and clean male SD rats weighing (180 - 220) g were randomly divided into three groups (n = 10): normoxia group (n), hypoxia group (H) and Chuanxiong group (L). Group N was fed in normoxic environment, and the other two groups were fed in hypoxic (9% 11% O2) environment for 4 weeks, 8 h/D, 6 days a week. Rats in group L were gavaged with Ligusticum chuanxiong solution diluted with normal saline at the concentration of 300 mg/kg, and rats in group H were gavaged with equal volume of normal saline. After 4 weeks, the mean pulmonary artery pressure was measured. After pulmonary perfusion, the right ventricular free wall and left ventricle plus ventricular septum were taken to measure the right ventricular hypertrophy index. The changes of pulmonary morphology and ultrastructure were observed under light microscope. Results: Compared with group n, the average pulmonary artery pressure and right ventricular hypertrophy index in the other two groups increased, and the thickening of pulmonary vascular wall was obvious under microscope (P Conclusion: Ligusticum chuanxiong can relieve pulmonary artery pressure in rats by inhibiting pulmonary vascular remodeling.

OBJECTIVE: Acephate is a widely used organophosphorus insecticide, but whether it inhibits androgen synthesis and metabolism remains unclear. In the current study, we investigated acephate in the inhibition of androgen synthetic and metabolic pathways in rat immature Leydig cells. METHODS: After 3 h culture in vitro with different concentrations（0, 0.5, 5,and 50 m M） of acephate, immature Leydig cell androgen synthesis related and other genes expression tested by qPCR and Western blot. RESULTS: Acephate inhibited basal androgen output in a dose-dependent manner with the inhibition starting at 0.5 m M. It significantly inhibited luteinizing hormone and 8-Br-cAMP stimulated androgen production at 50 m M. It significantly inhibited progesterone-mediated androgen output at 50 mM. Further study found that acephate down-regulated the expression of Hsd3b1 at 0.5 mM, Lhcgr at 5 mM and Star at 50 mM. Acephate directly blocked rat testicular HSD3B1 activity at 50 m M. Acephate did not affect other androgen synthetic and metabolic enzyme activities as well as ROS production, proliferation, and apoptosis of Leydig cells. CONCLUSION: Acephate targets LHCGR, STAR, and HSD3B1, thus blocking androgen synthesis in rat immature Leydig cells and HSD3B1 is the most sensitive target of acephate.
