In vitro activity of natural honey alone and in combination with curcuma starch against Rhodotorula mucilaginosa in correlation with bioactive compounds and diastase activity

Objective:To evaluate the in vitro activity and synergism of the combinations of natural honey and curcuma starch against Rhodotorula mucilaginosa in correlation with total phenolic,flavonoid contents,and diastase ***:The Folin-Ciocalteu test was used to determine the total polyphenols content and the flavonoid content was analyzed using by the aluminum chloride *** antifungal activity of the natural honey,determined by an agar well diffusion assay and agar incorporation ***:Total phenolic content varied from(63.93±0.11)to(95.36±6.08)mg GAE/100 g honey as gallic acid *** flavonoids content varied from(5.41±0.04)to(9.94±0.54)mg CE/*** activity values were between(7.3±2.8)and(26±2.8).The zone inhibition diameter for the six honey samples without starch ranged between 6 and 20 *** starch was mixed with honey and then added to well,a zone inhibition increase diameter 7 and 21 *** percentage increase was noticed with each variety and it ranged between 5%and 62.5%.The minimal inhibitory concentrations for the six varieties of honey without starch against Rhodotorula mucilaginosa ranged between 28%and 36%(v/v).When starch was incubated with honey and then added to media,a minimal inhibitory concentration drop has been noticed with each *** ranged between 6.66%and 20%(w/v).No significant correlation was established between diastase activity and bioactive ***:The mixture of curcuma starch and honey could lead to the development of new combination antibiotics against Rhodotorula

Effect of heat treatment on antimycotic activity of Sahara honey

Objective:To evaluate the influence of the temperature on honey colour,polyphenol contents and antimycotic capacity and to evaluate the correlation between these ***:Sahara honey were heated up to 25,50,75 and 100℃for 15,30 and 60 min,and their colour intensity,polyphenol contents and antimycotic *** Folin-Ciocalteu test was used to determine the total polyphenol contents(TPC).The antimycotic activity was evaluated both by agar diffusion method and micro wells dilution method against the Candida albicans(***)and Candida glabrata(***).Results:Initial values for TPC in Sahara honey ranged from 0.55 to 1.14 mg of gallic acid per kg of honey,with the average value of 0.78 mg of gallic acid per kg of *** TPC values after heat-treatment were 0.54 to 1.54 with the average value of 1.49 *** minimal inhibitory concentrations before heat-treatment of Sahara honey against *** and *** ranged from 3.06%-12.5%and 50%*** heat-treatment the minimal inhibitory concentrations between 12.5%and 50%for *** and ***,*** diameters of inhibition zones of Sahara honey with 50%concentration varied from(12.67-15.00)mm by *** to(14.33-15.67)mm by *** diameters of inhibition zones after heat-treatment at 25 and 50°C for 15.30 and 60 min ranged from(2.00-18.67)mm by *** to(8.00-16.67)mm by *** significant relations between the TPC and the colour intensity of Sahara honey(r=0.99,P<0.05).Furthermore,the results showed that the TPC and colour is not correlated with the antimycotic ***:To our knowledge this is the first report on the antimycotic capacity of Sahara honey.

Preliminary comparative study of anti-inlfammatory effect of unheated and heat-treated Sahara honey:In vivo approach

Objective:To investigate the effect of unheated and heat-treated of Sahara ***:A total of 24 Swiss albino mice weighing 25-35 g were divided into four groups(n=6).Anti-inflammatory effect was assessed at 1,2,3,4,5 and 6 h after subplantar injection of carrageenan(0.5 mL of a 1%solution in normal saline).In addition,total phenolic content was determined by modified Folin-Ciocalteu ***:The total phenolic content capacity of the Sahara honey before and after heat treatment was between 72 and 97.9 mg of gallic acid equivalents/100 g of honey *** of unheated honey(oral administration)reduced significantly(P<0.05).The carrageenan induced mice paw edema model at 1,3 and 6 h for 21.85%,5.43%and 80.43%,*** of heat-treated honey showed insignificant inhibition of carrageenan and induced paw edema at 1 h(31.16%),3 h(0.25%)and 6 h(34.19%).The 50 mg/kg diclofenac exhibited percent reduction in paw volume 16.12%,8.90%and 15.32%after 1 h,3 h and 6 h,respectively,when compared with control *** toxicity was ***:Our results suggest that unheated Sahara honey has anti-inflammatory effects by reducing the mice paw edema size while heat-treated Sahara honey decreases the antiinflammatory activity.

Objective:To search for further synergistic combinations of gentamicin and raw honey that might have potential in treating ***:The antibacterial activity and synergistic interaction of raw honey and gentamicin was assessed by using agar well diffusion *** Gram-negative(Escherichia coli ATCC 25922 and Pseudomonas aeruginosa ATCC 2154)bacteria were selected for antibacterial activity *** cultures of bacteria were maintained in their appropriate agar slants at 4°C throughout the study and used as stock ***:Raw honey and gentamicin interacted synergistically to inhibit Escherichia coli and Pseudomonas ***:These results suggest that combinations of raw honey and gentamicin have therapeutic benefits in prophylaxis of infections caused by multidrug-resistant Gram-negative bacilli.

Pharmacologically tested aldose reductase inhibitors isolated from plant sources—A concise report

Aldose reductase (AR), a cytosolic, monomeric oxidoreductase, is a key enzyme in the polyol pathway which controls the conversion of glucose to sorbitol. The accumulation of sorbitol by the activation of AR enzymes in lens, retina, and sciatic nerves leads to the cause of diabetic defects resulting in various secondary complications, viz. retinopathy, neuropathy, nephropathy and Alzheimer's disease. Thus, reduction of the polyol pathway flux by AR inhibitors could be a potential therapeutic opening in the treatment and prevention of diabetic complications. At present, the AR inhibitors belong to two different chemical classes. One is the hydantoin derivatives, such as Sorbinil, Dilantin, and Minalrestat, and the other is the carboxylic acid derivatives, such as Epalrestat, Alrestatin, and Tolrestat. However, it is known that most of these synthethic compounds have unacceptable side-effects. Well known medicinal plants like Chrysanthemum indicum, Chrysanthemum morifolium, Prunus mume, Myrcia multiflora, Centella asiatica, and Salacia reticulata, Salacia oblonga, and Salacia chinensis exhibited potent AR inhibitory activity. The present review summarizes the list of plant material, and their isolated phytoconstituents which have been tested for their AR inhibitory activity. This litreature review covers the period to 2011, and a total of 72 plants are listed.

OBJECTIVE:To explore the mechanistic effects of Yajieshaba(YJSB) on enhanced liver ***:The effects of YJSB on alanine aminotransferase(ALT) and aspartate aminotransferase(AST) were assayed in five acute chemical liver injury models[carbon tetrachloride(CCU),D-galactosamine(D-Glan),4-acetamidophenol(AAP),thioacetamide(TAA) and 1-naphthyl isothiocyanate(ANIT)].Sleep latency and sleep time of pentobarbital sodium were tested in control mice and CCL model miceafter oral YJSB *** effects of YJSB on drug metabolism enzymes of liver microsomes were tested in control rats and CCI_4model *** levels of cytochrome P450(CYP450) and Cyt b5 in liver microsomes were assayed using the method by Omura and Sato,and activities of erythromycin N-demethylase(ERD)and aminopyrine N-demethyl(ADM) were evaluated by Nash *** substrate-based high performance liquid chromatography(HPLC)methods were established for CYP3A4 and ***:The level of serum ALT was reduced by YJSB at 3.51 g/kg in the five models as follows:CCl_4 > D-Glan,AAP,ANIT > *** treatment did not reduce the level of serum *** at 3.51 g/kg prolonged the sleep latency in control mice and shortened the sleep time of control mice and CCl_4 model *** control rats,YJSB at 2.43 g/kg increased the levels of CYP450 and Cyt b5 and induced the activities of ERD and ADM;for liver injuries induced by CCI_4 in rats,YJSB at 2.43 g/kg increasedthe levels of CYP450 and Cyt *** results suggest that YJSB at 2.43 g/kg induces CYP3A4 and ***:These results suggest that YJSB enhanced liver detoxification and the mechanisms may be partially related to CYP3A4 and CYP1A2 induction.

Chenopodium ambrosioides induces an endothelium-dependent relaxation of rat isolated aorta

Objective: This study aims to evaluate the vasodilatory effect of Chenopodium ambrosioides on the isolated rat aorta, and to explore its mechanism of ***: The vasorelaxant effect and the mode of action of various extracts from the leaves of C. ambrosioides were evaluated on thoracic aortic rings isolated from Wistar rats. In addition, ethyl acetate and methanol fractions were analyzed, using thin-layer chromatography and high-performance liquid chromatography techniques, for their polyphenolic ***: The various active extracts of C. ambrosioides at four concentrations(10^(-3), 10^(-2), 10^(-1) and 1 mg/mL) relaxed the contraction elicited by phenylephrine, in a concentration-dependent *** effect seems to be endothelium-dependent, since the vasodilatory effect was entirely absent in denuded aortic rings. The vasorelaxant effect of the methanol fraction(MF) of C. ambrosioides at 1 mg/mL was also inhibited by atropine and tetraethylammonium. This effect remained unchanged by Nx-nitro-L-arginine methyl ester hydrochloride and glibenclamide. The preliminary phytochemical analysis showed that the leaves of C. ambrosioides are rich in phenolic and flavonoid ***: These results suggest that the MF of C. ambrosioides produces an endothelium-dependent relaxation of the isolated rat aorta, which is thought to be mediated mainly through stimulation of the muscarinic receptors, and probably involving the opening of Ca^(2+)-activated potassium channels.
