Rational design and exploitation of nanomaterials with superior treatment properties for suitable indications is a way out to relieve cost constraint of therapy and solve the unsatisfactory efficacy for cancer *** this work,we propose a greatly facile approach to produce heterogeneous Pd-Au nanorods(Pd-Au NRs)that solve the current bottleneck problems of photothermal thermal therapy(PTT)as well as completely eliminate tumors in animal models without toxic side *** Pd clusters on both tips of Au NRs offers Pd-Au NRs three novel functions,i.e.,the extension of the absorption into NIR-Ⅱ region,the activation of prodrug of 5-fluorouracil(5-Fu)via the bioorthogonal reaction,and the peroxidase-mimic activity to produce·*** heterogeneous nanorods showed a high and stable photothermal conversion efficiency(52.07%)in a safer NIR-Ⅱ irradiation region(1,064 nm),which not only eliminate most of tumor cells at only one dose of the irradiation for 5 min but also improve the in situ conversion of 5-fluoro-1-propargyluracil and H2O2 into active 5-Fu and·OH to eradicate residual tumors for inhibiting tumor *** dual catalytic activity-synergistic mechanism of PTT demonstrates the importance of material design in solving current bottleneck problem of tumor therapy.

To promote the development of extracellular vesicles of herbal medicine especially the establishment of standardization,led by the National Expert Committee on Research and Application of Chinese Herbal Vesicles,research experts in the field of herbal medicine and extracellular vesicles were invited nationwide with the support of the Expert Committee on Research and Application of Chinese Herbal Vesicles,Professional Committee on Extracellular Vesicle Research and Application,Chinese Society of Research Hospitals and the Guangdong Engineering Research Center of Chinese Herbal *** on the collation of relevant literature,we have adopted the Delphi method,the consensus meeting method combined with the nominal group method to form a discussion draft of“Consensus statement on research and application of Chinese herbal medicine derived extracellular vesicles-like particles(2023)”.The first draft was discussed in online and offline meetings on October 12,14,November 2,2022 and April and May 2023 on the current status of research,nomenclature,isolation methods,quality standards and research applications of extracellular vesicles of Chinese herbal medicines,and 13 consensus opinions were finally *** the Third Academic Conference on Research and Application of Chinese Herbal Vesicles,held on May 26,2023,Kewei Zhao,convenor of the consensus,presented and read the consensus to the experts of the Expert Committee on Research and Application of Chinese Herbal *** consensus highlights the characteristics and advantages of Chinese medicine,inherits the essence,and keeps the righteousness and innovation,aiming to provide a reference for colleagues engaged in research and application of Chinese herbal vesicles at home and abroad,decode the mystery behind Chinese herbal vesicles together,establish a safe,effective and controllable accurate Chinese herbal vesicle prevention and treatment system,and build a bridge for Chinese medicine to the world.

Long-wavelength fluorescence carbon dots (CDs) show great importance in multiple fields,especially for the biochemical ***,we proposed one type of CDs doped with nitrogen and sulfur through the hydrothermal method,which exhibited obvious yellow-fluorescence in aqueous ***,the fluorescence intensity of CDs decreased with pH decreasing in the acidic range,thus a linear relationship between pH and fluorescence intensity was established,exhibiting the potential of pH ***,introducing tigecycline into CDs resulted in their decreased fluorescence,thus,we further established a strategy of detecting tigecycline with the concentration range of 200 μM to 7 ***,we elucidated the static quenching as the major mechanism for CDs responding tigecycline,which was induced by the formed new complex between CDs and ***,the practicality of the method was verified by examining the recovery of tigecycline in the actual lake-water samples.

AIM: The aim of this work was to establish a specific and sensitive method to comprehensively investigate and compare chemical constituents of Fuzi-Gancao herb pair(FG), consisting of Aconitum carmichaelii Debeaux(Fuzi, Chinese) and Roast Radix Glycyrrhizae(Glycyrrhiza glabra L., Gancao, in Chinese) and Fuzi alone to explore the underlying interaction mechanism of FG. METHOD: An ultra-fast liquid chromatography-ion trap/time-of-flight mass spectrometry(UFLC/MS-IT-TOF) method using diazepam as internal standard was developed for the identification and semi-quantitative analysis of the phytochemical constituents of Fuzi and FG. Chromatographic separation was achieved on a UFLC column using a gradient program with 40 mmol?L-1 ammonium acetate and acetonitrile as the mobile phase. RESULTS: Fifty-one of the sixty compounds, including forty-five C19-diterpenoid alkaloids and six C20-diterpenoid alkaloids were tentatively identified in the extracts of Fuzi and FG through accurate mass measurements and fragmentation patterns. Comparing the contents of these alkaloids in these two extracts, it was found that the diester-diterpenoid alkaloids(DDAs) and the alkylolamine-diterpenoid alkaloids(ADAs) were increased, while the monoester-diterpenoid alkaloids(MDAs) were decreased in the extracts of FG. CONCLUSION: This work provided comprehensive information for the quality control of Fuzi preparations, and the further investigation on the compatibility mechanisms of FG.

Fatty acids(FAs)are an important class of functional small molecules and participate in a variety of life biological *** experiment proposed a liquid chromatography–quadrupole-Qrbitrap mass spectrometry(LC–Q-Orbitrap MS)method to perform FA profiling under parallel reaction monitoring(PRM)acquisition *** FA was derivatized by 2-dimethylaminoethylamine(DMED)to increase the ionization e fficiency and provide the characterized fragment *** mass spectra obtained from full scan,multiple ion monitoring(MIM),and PRM were *** results showed that the protonated ion of FA+DMED–H2O was detected and the neutral loss of 45.06 Da was observed in the tandem mass *** PRM method provided the highest selectivity and sensitivity for FA detection with the help of accurate mass weight and specific neutral loss-based *** method validation was performed using the FA standards and pooled quality control serum sample,which showed that the established method had good repeatability,stability,and ***,the developed method was successfully applied to analyze the rat serum and brain tissue samples for the drug e ffi-cacy evaluation of gross saponin of Tribulus terrestris ***(GSTTF)against middle cerebral artery occlusion(MCAO)-induced ischemic *** method has extensive practicability and great potential in the detection of FA,especially for the analysis of samples with complex matrices.

Pulmonary fibrosis(PF)is an irreversible lung disease that is characterized by excessive scar tissue with a poor median survival rate of 2-3 *** inhibition of transforming growth factor-β receptor type-I(TGF-β RI)by an appropriate drug may provide a promising strategy for the treatment of this *** cuspidatum(PC)is a well-known traditional Chinese herbal medicine which has an anti-PF ***,a combination of high resolution mass spectrometry with an in silico strategy was developed as a new method to search for potential chemical ingredients of PC that target the TGF-β *** on this strategy,a total of 24 ingredients were ***,absorption,distribution,metabolism,and excretion(ADME)-related properties were subsequently predicted to exclude compounds with potentially undesirable pharmacokinetics *** docking studies on TGF-β RI were adopted to discover new PF ***,a compound that exists in PC known as resveratrol was proven to have excellent biological activity on TGF-β RI,with an IC_(50) of 2.211 μM in ***,the complex formed through molecular docking was tested via molecular dynamics simulations,which revealed that resveratrol had strong interactions with residues of TGF-β *** study revealed that resveratrol has significant potential as a treatment for PF due to its ability to target TGF-β *** addition,this research demonstrated the exploration of natural products with excellent biological activities toward specific targets via high resolution mass spectrometry in combination with in silico technology is a promising strategy for the discovery of novel drugs.

AIM: To study the hepatoprotective effect of methanol extract of Gentiana veitchiorum(MGV) against CCl4-induced oxidative stress and liver injury in mice. METHOD: The acute hepatic model was developed by injection of 20% CCl4 in mice. ICR mice were divided into six groups, including control, CCl4, CCl4+ silymarin, and CCl4+ MGV(100, 200, and 400 mg·kg–1) groups. Hepatic enzymes including AST, ALT and ALP levels in serum, and antioxidant enzymes, including SOD, CAT and GPX activity in liver tissue, were determined. Histopathological examination and Western blot analysis were performed. RESULTS: Oral administration of MGV at 200 and 400 mg·kg–1 for 15 days dose-dependently inhibited the serum elevations of AST, ALT, and ALP, and recovered the reduction of SOD, CAT, and GPX in liver tissue. Hematoxylin and eosin staining examination performed in liver tissues suggested that MGV treatment ameliorated histopathological changes in CCl4-induced mice. Western blotting analysis implied that MGV increased HO-1 expression and recovered TNF-α alternation. CONCLUSION: G. veitchiorum can protect the liver against CCl4-induced damage in mice, and this hepatoprotective effect was due at least in part to its ability through scavenging CCl4-associated free radical activities. The study provided in vivo evidence that G. veitchiorum can be used as a safe, cheap, and effective agent to reduce acute liver damage, supporting its folk medicine use.

Objective:Phellodendron chinense(PC)bark and Phellodendron amurense(PA)bark are two herbal medicines recorded in the Chinese Pharmacopoeia(2020 edition)that are easily mistaken for one *** this study,the chemical constituents of PC and PA were compared using chromatographic fingerprints,and the potential synergistic acetylcholinesterase(ACh E)inhibitor components were screened based on the correlation of fingerprint *** and Methods:Chromatographic fingerprints based on high-performance liquid chromatography were developed for the analysis and comparison of chemical compounds in PC and PA *** ACh E inhibitory activity of PC and PA was determined using the Ellman ***,the contribution of the characterized alkaloids in PC and PA to the overall ACh E inhibition was modeled using partial least squares regression(PLSR).Results:The total alkaloid content in PC was higher than that in PA,which causes PC to have stronger anti-ACh E ***,13 and 20 common peaks were identified in the PC and PA samples,*** them,berberine(BER)was the dominant alkaloid in PC,which covered more than 65%of the total peak area in PC,but only approximately 25%of that in PA,indicating that the chemical composition is different between PC and *** spectrum–effect analysis based on PLSR and the correlation analysis showed that the BER-palmatine(PAL)and BER-jatrorrhizine(JAT)pairs have a synergistic inhibitory effect on ACh E ***:A high-performance chromatographic fingerprint was established to distinguish PC and *** efficacy-associated markers were screened,including the pairs of BER-PAL and BER-JAT with anti-ACh E activity,and the findings may assist with the quality control of PA and PC.

In this study,37 derivatives of phorbol esters were synthesized and their anti-HIV-1 activities evaluated,building upon our previous synthesis of 51 phorbol derivatives.12-Para-electron-acceptor-trans-cinnamoyl-13-decanoyl phorbol derivatives stood out,demonstrating remarkable anti-HIV-1 activities and inhibitory effects on syncytia *** derivatives exhibited a higher safety index compared with the positive control *** them,12-(trans-4-fluorocinnamoyl)-13-decanoyl phorbol,designated as compound 3c,exhibited the most potent anti-HIV-1 activity(EC_(50)2.9 nmol·L^(−1),CC50/EC_(50)11117.24)and significantly inhibited the formation of syncytium(EC_(50)7.0 nmol·L^(−1),CC50/EC_(50)4891.43).Moreover,compound 3c is hypothesized to act both as an HIV-1 entry inhibitor and as an HIV-1 reverse transcriptase *** titration calorimetry and molecular docking studies indicated that compound 3c may also function as a natural activator of protein kinase C(PKC).Therefore,compound 3c emerges as a potential candidate for developing new anti-HIV drugs.
