The aim is to develop a liquid chip technique to detect Taura syndrome virus( TSV) and yellow head disease virus( YHDV) on Penaeus orientalis simultaneously. The CP2 gene of TSV and N gene of YHDV in Gen Bank was analysed by using the software DNAStar 7. 0 to design the TSV-and YHDV-specific primers. The primers were labeled with biotin and subjected to amination modification. They were then coupled with fluorescence-coded microspheres and then used for hybridization with RT- PCR products of TSV and YHDV. The liquid chip detection technique for detection of TSV and YHDV was established by using BD FACSArray to detect fluorescence signal in the reaction system. This assay system had a high sensitivity to TSV and YHDV,with the detection of limit of 100 pg. Moreover,the assay was specific for the detection of TSV,YHDV and was not susceptible to cross with other viruses,including white spot syndrome virus( WSSV),spring viremia of carp virus( SVCV),infectious haematopoietic necrosis virus( IHNV). In conclusion,the liquid chip assay technique established in this study is highly sensitive and specific to TSV and YHDV detection. Moreover,it provides a novel,convenient and rapid approach for the detection of TSV and YHDV.

PCR Detection of the Three <i>Neofabraea</i>Pathogenic Species Responsible for Apple Bull’s Eye Rot

Apple bull’s eye rot is caused by pathogenic Neofabraea species including N. malicorticis, N. perennans and N. alba. Fruits carrying this fungal quarantine disease are prohibited from entering China. The host plants for the pathogens include several Rosaceae fruits including apple and pear. Disease symptoms and pathogen morphology are often insufficient to determine the identity of the pathogen, particularly at the species level. In the current study, we analyzed the inter-species sequence variations in the β-tublin gene, and designed specific primers to allow PCR amplification of 554 bp fragments from pathogenic Neofabraea species. The PCR products were recovered and sequenced, and Blast search was conducted using the DNA sequences in the Genbank database. The results indicated precise PCR amplification of the target sequences from the host pathogen, which allowed unambiguous identification of the species.

Obesity is a worldwide epidemic and results from excessive energy intake or inefficient energy *** is promising to utilize the thermogenic function of brown adipose tissue for obesity ***,the mechanisms controlling the efficacy of norepinephrine-induced thermogenesis in brown adipocytes remain *** we demonstrate that norepinephrine(NE)induces low-efficacy thermogenesis,evoking both heterogeneous changes(ΔΨm andΔpH)and homogenous responses,one of which is that NE stimulation causes large amounts of ATP consumption in brown *** reveal that the proton-ATPase activity of mitochondrial complex V is a key factor that antagonizes proton leakage by UCP1 and determines the efficacy of NE-induced thermogenesis in brown ***,to avoid unnecessary and undesired heat production,we reveal that ATP is a necessary sympathetic cotransmitter for the high efficacy and specificity of NE-induced thermogenesis in brown adipocytes as it increases intracellular calcium concentrations and upregulates the ATP synthase activity of complex ***,we demonstrate the modulation mechanism of thermogenic efficacy in brown *** findings imply new strategies to partially or fully utilize the thermogenic capacity of brown adipocytes to identify therapeutic targets for the treatment of obesity and diabetes.

The study was conducted to investigate the effects of follicle-stimulating hormone (FSH) on embryonic chicken ovarian germ cell proliferation and its possible involvements of protein kinases A (PKA) and C (PKC) *** cells were treated with FSH alone or in the presence of forskolin (FRSK),PKA inhibitor (H89),PKC activator (PMA) or inhibitor (H7).The germ cell number was counted from *** immunocytochemistry of proliferating cell nuclear antigen (PCNA) was applied to identify the proliferating *** germ cell labeling index (LI) was determined for cell *** FSH treatment increased the germ cell number,and this stimulating effect was enhanced by FRSK or PMA,but inhibited by H89 or H7 in a dose-dependent ***,the PCNA-LI showed parallel changes with germ cell *** study suggests that FSH may stimulate proliferation of cultured chicken ovarian germ cells by activation of both the PKA and PKC signaling pathways.

We cultured different-sized fractions of dominant phytoplankton species, Skeletonema costatum, Chaetoceros curvisetus, and Thalassiosira nordenskioldii, collected in different sea areas in various seasons, and measured and compared their C, N, P, Si contents. The N content of these species is similar, while the C, P, and Si contents of S. costatum from eutrophic Changjiang （Yangtze River） estuary are higher than those from Jiaozhou Bay （JZB）, particularly the content of Si. The C, N, P, and Si contents of cultured phytoplankton in JZB increase with size fraction augmentation, and the percentages of C, N, and P follow the same trend, while the percentage of Si remain constant. Moreover, S. costatum from small-sized fraction assimilated Si more easily than C. curvisetus and T. nordenskirldii, which is explained by the dominance of S. costatum under the conditions of low SiO3-Si concentration in JZB. The C, N, P, and Si contents of cultured S. costatum collected during summer and winter are higher, which is consistent with the phytoplankton blooming seasons in JZB. The SiO3-Si concentration of seawater during spring restrain the growth of phytoplankton, supported by the fact that the N, P, and Si contents and their ratios in cells of cultured S. costatum are low in spring season.

Evaluation of Uncertainty for Detecting the Content of Banned Aromatic Amines in Textile Products by GC-MS

The article evaluated the uncertainty of the content of banned aromatic amines in textile products in terms of GB/ T17592. 1 - 1998 Textiles-test Method of Banned Azo Coloumnts-gas Chromatography/Mass Spectrometric Method, analyzed and quantified the uncertainty components which affected the measurement results. Through calculation, the uncertainty of the sample mainly came from Co standard uncertainty, weighing, uncertainty of volmne, effect on the recovery rate of sample during the course of extraction and purification and ucertainty from the random effect during the course of measurement. It was obtained that the expanded uncertainties of the content of 2 - naphthylamine, benzidine and 2, 4 - diaminotoluenc with higher frequency in 24 kinds of forbidden aromatic amines were respectively 2. 128, 2.302 and 4.778 mg/kg.

Speciation Analysis of Arsenic Compounds in Seafood by Ion Chromatography-Atomic Fluorescence Spectrometry

Ion chromatography-ultra violet-hydride generation-Atomic Florescence Spectrometry was applied to detect 5 arsenic species in seafoods. The arsenic species studied include arsenobetaine(As B), arsenite(As(III)), dimethylarsinic acid(DMA), monomethylarsonic acid(MMA), and arsenate(As(V)), which were extracted from samples using 2% formic acid. Gradient elution using 33 mmol L^(-1) CH_3COONH_4 and 15 mmol L^(-1) Na_2CO_3 with 10 mL CH_3CH_2OH at pH 8.4 allowed the chromatographic separation of all the species on a Hamilton PRP-X100 anion-exchange column in less than 8 min. In this study, an ultrasound extraction method was used to extract arsenic species from seafood. The extraction efficiency was good and the recoveries from spiked samples were in the range of 72.6%–109%; the precision between sample replicates was higher than 3.6% for all determinations. The detection limits were 3.543 μg L^(-1) for As B, 0.4261 μg L^(-1) for As(III), 0.216 μg L^(-1) for DMA, 0.211 μg L^(-1) for MMA, and 0.709 μg L^(-1) for As(V), and the linear coefficients were greater than 0.999. We also developed an application of this method for the determination of arsenic species in bonito, Euphausia superba, and Enteromorpha with satisfactory results. Therefore, it was confirmed that this method was appropriate for the detection of arsenic species in seafood.

Two species of the genus Boysidia Ancey, 1881 were reported as new to science, Boysidia（Boysidia） xianfengensis sp. nov. and B.（B.） xiaoguanensis sp. nov., while the distribution of the 23 species of Boysidia from China was discussed. The materials of the new species were collected from Hubei Province, China by authors. The shell of B.（B.） xianfengensis sp. nov. looks small（height 4.90 mm, diameter 3.10 mm）, and has five apertural teeth： one large parietal tooth, one long columellar tooth, one small nipple-shaped upper palatal tooth, one large flake-shaped lower palatal tooth, one small infrapalatal tooth. The shell of B.（B.） xiaoguanensis sp. nov. looks small（height 3.55 mm, diameter 2.20 mm）, and has four apertural teeth： one large parietal tooth, one long columellar tooth, one large flake-shaped upper palatal tooth, one slightly small flake-shaped lower palatal tooth. The type specimens of the new species are deposited in the Institute of Zoology, Chinese Academy of Sciences, China.
