Induced neural stem cells regulate microglial activation through Akt-mediated upregulation of CXCR4 and Crry in a mouse model of closed head injury

Microglial activation that occurs rapidly after closed head injury may play important and complex roles in neuroinflammation-associated neuronal damage and *** previously reported that induced neural stem cells can modulate the behavior of activated microglia via CXCL12/CXCR4 signaling,influencing their activation such that they can promote neurological ***,the mechanism of CXCR4 upregulation in induced neural stem cells remains *** this study,we found that nuclear factor-κB activation induced by closed head injury mouse serum in microglia promoted CXCL12 and tumor necrosis factor-αexpression but suppressed insulin-like growth factor-1 ***,recombinant complement receptor 2-conjugated Crry(CR2-Crry)reduced the effects of closed head injury mouse serum-induced nuclear factor-κB activation in microglia and the levels of activated microglia,CXCL12,and tumor necrosis factor-α.Additionally,we observed that,in response to stimulation(including stimulation by CXCL12 secreted by activated microglia),CXCR4 and Crry levels can be upregulated in induced neural stem cells via the interplay among CXCL12/CXCR4,Crry,and Akt signaling to modulate microglial *** agreement with these in vitro experimental results,we found that Akt activation enhanced the immunoregulatory effects of induced neural stem cell grafts on microglial activation,leading to the promotion of neurological recovery via insulin-like growth factor-1 secretion and the neuroprotective effects of induced neural stem cell grafts through CXCR4 and Crry upregulation in the injured cortices of closed head injury ***,these beneficial effects of Akt activation in induced neural stem cells were positively correlated with the therapeutic effects of induced neural stem cells on neuronal injury,cerebral edema,and neurological disorders post–closed head *** conclusion,our findings reveal that Akt activation may enhance the immunoregulatory effects of induced neura

OBJECTIVE: To investigate the effect of Sanhuangyilong decoction plus methotrexate(MTX) on Interferon gamma(IFN-γ) and tumor necrosis factor alpha(TNF-α) in the serum and synovial fluid of rheumatoid arthritis(RA) patients with damp-heat-obstruction symptom pattern, Sanhuangyilong decoction and the role of TNF-α and IFN-γ in the development of ***: RA inpatients with damp-heat-obstruction symptom pattern(partly with knee joint effusion) were selected as the research subjects. Before the treatment, healthy subjects and osteoarthritis(OA) patients with knee joint effusion were assigned to the serum control group and the synovial fluid control group, respectively; during the treatment, RA patients with damp-heat-obstruction symptom pattern were divided into two groups:one is combined group that was administered Sanhuangyilong decoction plus MTX; the other group was MTX group that received MTX only. Polypyrrole （PPy）, the earliest prepared conducting polymer, has good biocompatibility, easy synthesis and flexibility in processing. Compared with metal and inorganic materials, doped PPy has better mechanical match with live tissue, resulting in its many applications in biomedical field. This mini-review presents some information on specific PPy properties for tissue engineering applications, including its synthesis, doping, bio-modiflcation. Although some challenges and unanswered problems still remain, PPy as novel biomaterial has promoted the development tissue engineering for its clinical application in the future.

Stages based molecular mechanisms for generating cholangiocytes from liver stem/progenitor cells

Except for the most organized mature hepatocytes,liver stem/progenitor cells(LSPCs)can differentiate into many other types of cells in the liver including *** addition,LSPCs are demonstrated to be able to give birth to other kinds of extra-hepatic cell types such as insulin-producing *** more,under some bad conditions,these LSPCs could generate liver cancer stem like cells(LCSCs)through malignant *** this review,we mainly concentrate on the molecular mechanisms for controlling cell fates of LSPCs,especially differentiation of cholangiocytes,insulin-producing cells and *** of all,to certificate the cell fates of LSPCs,the following three features need to be taken into account to perform accurate phenotyping:(1)morphological properties;(2)specific markers;and(3)functional assessment including in vivo ***,to promote LSPCs differentiation,systematical attention should be paid to inductive materials(such as growth factors and chemical stimulators),progressive materials including intracellular and extracellular signaling pathways,and implementary materials(such as liver enriched transcriptive factors).Accordingly,some recommendations were proposed to standardize,optimize,and enrich the effective production of cholangiocyte-like cells out of *** the end,the potential regulating mechanisms for generation of cholangiocytes by LSPCs were carefully *** differentiation of LSPCs is a gradually progressing process,which consists of three main steps:initiation,progression and ***’s the unbalanced distribution of affecting materials in each step decides the cell fates of LSPCs.

Effect of methotrexate combined with Sanhuang Yilong decoction on serum and synovial fluid aquaporin levels in rheumatoid arthritis dampness-heat blockage syndrome

OBJECTIVE:To study the effect of methotrexate(MTX) combined with Sanhuang Yilong decoction(SYD) on aquaporin(AQP) expression,and to explore the role of AQPs in the pathogenesis of rheumatoid arthritis(RA).METHODS:A total of 118 dampness-heat blockage type RA patients who were hospitalized in the general chengdu military hospital between January2014 and December 2016 were selected as subjects in this study(30 patient of these patients with knee joint effusion were assigned to the RA synovial fluid group).For the pre-treatment controlgroups,30 healthy volunteers were recruited as the healthy control group and 30 osteoarthritis(OA) patients with knee joint effusion were included as OA synovial fluid control *** RA dampnessheat blockage syndrome treatment groups were divided into 45 cases in the combined group and 45 cases in the MTX *** combined group received MTX combined with SYD treatment while the MTX group received MTX ***1,AQP2 and AQP3 expressions were detected in the serum and synovial ***:AQP1 had the highest expression,followed by AQP3,and *** serum levels of AQP1,AQP2 and AQP3 were all significantly lower than those in the healthy volunteers(P 0.05).After treatment for2 weeks,serum AQP1,AQP2,AQP3 were significantly increased and erythrocyte sedimentation rate,C-reactive protein,disease activity score of 28 joints were decreased in the combined group(P play an important role in the pathogenesis of *** combined with SYD for the treatment of RA can effectively increase AQP expression.

Characteristics of fecal microbiota in non-alcoholic fatty liver disease patients

This study was designed to investigate the gut microbiota of patients with non-alcoholic fatty liver disease. The inclusive and exclusive criteria for NAFLD patients and healthy subjects were formulated, and detailed clinical data were collected. The genomic DNA of stool samples were extracted for 16 S r DNA sequencing, and the amplified V4-region was sequenced on the Illumina Miseq platform. Metastats analysis was performed to identify the differential taxa between the groups. Redundancy analysis was used to evaluate the association between gut microbial structure and clinical variables. Thirty NAFLD patients and37 healthy controls were involved. The 16 S r DNA sequencing showed that there was a dramatic variability of the fecal microbiota among all the individuals. Metastats analysis identified eight families and 12 genera with significant differences between the two groups. When some clinical parameters, such as waist-to-hip ratio(WHR) and homeostasis model assessment of insulin resistance(HOMA-IR), were enrolled in Redundancy analysis, the distribution of the two group of samples was obviously changed. The compositional shifts in fecal bacterial communities of NAFLD patients from the healthy controls were mainly at family or genus levels. According to our Redundancy analysis, insulin resistance and obesity might be closely related to both NAFLD phenotype and intestinal microecology.
