Activities of the Deriviatives of Chitin on the Osbeoblast Proliferation and the Effect on Bone Strength in Ovariectomized Rats

This study intends to examine the effects of different concentrations of four kinds of degradations of chitin： glucosamine （GLC）, N-acetyl-D-glucosamine （NAG）, chitooligosaccharide（COS）, CM-chitooligosaccharide （CM-COS）—on the proliferation of MC3T3-E1 cell line cultured in vitro. Results suggest that all of the glucoses mentioned above promoted the proliferation of osteoblast, and various concentrations have different effects： the proliferation was remarkable when the concentration of GLC, NAG, COS, CM-COS was 100 , 100, 500, 500 μg/mL ,respectively. Furthermore we choose the glucosamine as the material and study the effect on the bone strength in ovariectomized rats. The results showed that the middle does of glucosamine can significantly increase the hone strength of femur in ovariectomized rats.

Living cells and active factors are the two core elements of tissue repair,directly affecting the healing efficiency of damaged ***(NF)can release living cells,such as adipose-derived stem cells(ADSCs),as well as active growth factors to promote angiogenesis,thus realizing cell-based wound ***,a novel living electrospun short fibrous sponge is constructed by modifying three-dimensional(3D)bionic short fibers with engineered *** uniform distribution of the polydopamine(PDA)modification endows the living sponges with stable mechanical properties,reversible water absorption and excellent adhesion even after repeated compression by an external force and long-term aqueous ***,the living electrospun short fibrous sponges with uniform NF modification contain living cells such as ADSCs and active growth factors such as vascular endothelial growth factor(VEGF),which can effectively promote the tube formation of human umbilical vein endothelial cells(HUVECs).In vivo,the living sponges can effectively and continuously act on wounds and act as a bionic living skin to prevent the loss of internal nutrients,creating a comfortable and favorable microenvironment for tissue regeneration and promoting the healing of diabetic ***,living electrospun short fibrous sponges via engineered NF are expected to achieve continuous wound healing with in situ living cells and active factors in injured tissues.

Gelatin-based composite hydrogels with biomimetic lubrication and sustained drug release

The occurrence of osteoarthritis is closely related to progressive and irreversible destruction of the articular cartilage,which increases the friction significantly and causes further inflammation of the ***,a scaffold for articular cartilage defects should be developed via lubrication restoration and drug *** this study,we successfully synthesized gelatin-based composite hydrogels,namely GelMA–PAM–PMPC,with the properties of biomimetic lubrication and sustained drug release by photopolymerization of methacrylic anhydride modified gelatin(GelMA),acrylamide(AM),and 2-methacryloyloxyethyl phosphorylcholine(MPC).Tribological test showed that the composite hydrogels remarkably enhanced lubrication due to the hydration lubrication mechanism,where a tenacious hydration shell was formed around the zwitterionic phosphocholine *** addition,drug release test indicated that the composite hydrogels efficiently encapsulated an anti-inflammatory drug(diclofenac sodium)and achieved sustained ***,the in vitro test revealed that the composite hydrogels were biocompatible,and the mRNA expression of both anabolic and catabolic genes of the articular cartilage was suitably *** indicated that the composite hydrogels could effectively protect chondrocytes from inflammatory cytokine-induced *** summary,the composite hydrogels that provide biomimetic hydration lubrication and sustained local drug release represent a promising scaffold for cartilage defects in the treatment of osteoarthritis.
