Bioinformatics and in vitro study reveal the roles of microRNA-346 in high glucose-induced human retinal pigment epithelial cell damage

AIM:To study microRNAs(miRNAs)and their potential effects in high glucose-induced human retinal pigment epithelial cell ***:We screened the GSE52233 miRNA expression dataset for differentially expressed miRNAs(DEMs).The target genes of the top 10 DEMs were predicted using miRWalk 2.0 database,followed by function enrichment and protein-protein interaction *** expression was determined in the human retinal pigment epithelial cell line ARPE-19 treated with high glucose(HG)by quantitative reverse transcription-polymerase chain reaction(qRT-PCR).Cell proliferation was determined using cell counting kit(CCK)-8 *** cycle,apoptosis,and reactive oxygen species(ROS)levels were determined by flow *** direct interaction between miRNA and targets was validated using dual-luciferase reporter ***:Thirty-nine DEMs were screened,and we predicted 125 miRNA-mRNA pairs for the top 10 DEMs,including 119 target genes of seven DEMs such as miR-346,which was upregulated in diabetic retinopathy(DR).miR-346 target genes were substantially enriched in the regulation of intracellular transport and retinoic acidinducible gene I(RIG-I)-like receptor signaling *** of three upregulated and downregulated miRNAs were verified by qRT-PCR in HG-treated ARPE-19 *** of miR-346 was elevated in HG treated ARPE-19 cells in a dose-dependent *** inhibited cell proliferation and induced apoptosis,which were partly reversed by transfecting an miR-346 inhibitor,which even decreased the ROS levels elevated due to *** 2(AGO2)was a target of ***:miR-346 is a key miRNA and plays an important role in HG-induced damage in human retinal pigment epithelial cells.

Cells are the fundamental units of biological systems and exhibit unique development trajectories and molecular features. Our exploration of how the genomes orchestrate the formation and maintenance of each cell, and control the cellular phenotypes of various organismsis, is both captivating and intricate. Since the inception of the first single-cell RNA technology, technologies related to single-cell sequencing have experienced rapid advancements in recent years. These technologies have expanded horizontally to include single-cell genome, epigenome,proteome, and metabolome, while vertically, they have progressed to integrate multiple omics data and incorporate additional information such as spatial sc RNA-seq and CRISPR screening. Single-cell omics represent a groundbreaking advancement in the biomedical field,offering profound insights into the understanding of complex diseases, including cancers. Here, we comprehensively summarize recent advances in single-cell omics technologies, with a specific focus on the methodology section. This overview aims to guide researchers in selecting appropriate methods for single-cell sequencing and related data analysis.

Clinical research of EX-PRESS drainage device and modified trabeculectomy combined with intravitreal conbercept treatment for neovascular glaucoma

AIM:To evaluate the efficacy and safety of modified trabeculectomy(experimental group) and implantation of EX-PRESS drainage device(control group),combined with intravitreal conbercept injection for neovascular glaucoma(NVG).METHODS:Totally 30 patients with NVG were selected from June 2014 to June 2017,and randomly divided into experimental group and control *** patients were underwent intravitreal conbercept(0.5 mg/0.05 mL) treatment before *** trabeculectomy was performed in MT group,while EX-PRESS drainage device implantation was performed in EX *** success rates,best corrected visual acuity(BCVA),intraocular pressure(IOP),filtering bleb and complications were observed and ***:The differences of success rate,BCVA and filtering bleb were not statistically significant 12mo after the surgery(P>0.05),however,the difference of IOP at 1d,1wk,1,3,and 6mo after surgery was statistically significant(F_(time)=390.64,P_(time)0.05),while the results at other time point indicate statistical differences(P<0.05).CONCLUSION:The modified trabeculectomy and the implantation of EX-PRESS drainage device have clinical application value in reducing IOP and postoperative complications of refractory NVG.

Effects of“Moxibustion Serum”on Proliferation and Phenotypes of Tumor Infiltrating Lymphocytes

Tumor infiltrating lymphocytes (TIL) were cultured with “moxibustion serum”(MS), and the results were examined by flow cytometry. The results indicated that MS could enhance the proliferation of TIL,accelerate it to reach the exponential growth phase, and assist recombinant interleukin 2 (rIL-2) to enhance successively the percentage of CD3^+ positive cells, maintain the number of CD4^+ positive T cells, promote greatly the percentage of CD8^+ positive T cells among TILs, and reverse the CD4^+/CD8^+ ratio. Such cooperative effects rely on relative specificity of acupoints. It is suggested that MS is beneficial to the growth of TIL both in the aspects of proliferation and phenotypes.

Design of tunable surface mode waveguide based on photonic crystal composite structure using organic liquid

With the method of replacing the surface layer of photonic crystal with tubes, a novel photonic crystal composite structure used as a tunable surface mode waveguide is designed. The tubes support tunable surface states. The tunable propagation capabilities of the structure are investigated by using the finite-difference time-domain. Simulation results show that the beam transmission distributions of the composite structure are sensitive to the frequency range of incident light and the surface morphology which can be modified by filling the tubes with different organic liquids. By adjusting the filler in tubes, the T-shaped, Y-shaped, and L-shaped propagations can be realized. The property can be applied to the tunable surface mode waveguide. Compared with a traditional single function photonic crystal waveguide, our designed structure not only has a small size, but also is a tunable device.

Effect of Nd^(3+) on spectroscopic properties of lithium borate glasses

The spectroscopic properties of lithium borate glasses as a function of Nd3＋ ions concentration were *** absorption spectra of these glasses showed a number of absorption bands in ultra violet and visible *** absorption edge was found to shift towards the longer wavelength（red shift） with increase in *** spectra revealed three major bands at 902, 1063 and 1334 nm which was due to 4F3/2→4I9/2, 11/2 ＆13/2 transitions of Nd3＋ *** intensity was maximum for 1 mol.% Nd2O3 and further increase in Nd2O3 resulted in luminescence *** luminescence quenching behavior at higher concentration of Nd2O3 was attributed to the Nd3＋-Nd3＋ interaction in the glass *** absorption and emission property of these glasses suggested that these glasses could be useful for 1.06 μm infrared laser applications.

Effects of In-situ Generated Coinage Nanometals on Crystallization and Microstructure of Fluorophlogopite Mica Containing Glass-Ceramics

The effects of in-situ generated coinage nanometals（Cu,Ag and Au） on crystallization behavior,microstructure,thermal and mechanical properties of SiO2-MgO-Al2O3-B2O3-K2O-MgF2（BMAPS） glassceramics were systematically *** addition of coinage nanometai,the glass transition temperature（Tg） is increased by 20-30 ℃,crystallization temperature（Tc） by 30-50 ℃ and dilatometric softening temperature（Td） by 10-25 ℃.It was found that the density of Cu-containing glass was 2.59 g cm-3and for other glasses it was in the range of 2.56-2.57 g *** the non-isothermal differential scanning calorimetry study,the activation energy of crystallization for BMAPS base glass was calculated as 344 kJ/mol,and changed to 406,334 and 274 kJ/mol on addition of Cu,Ag and Au-nanometals,*** evolved in the opaque BMAPS glass-ceramics derived by controlled heat treatment,were identified as fluorophlogopite mica(KMg3(AlSi3O10)F2) by X-ray diffraction（XRD） technique and confirmed by Fourier transformed infrared *** of copper,silver and gold nanometals were also identified by XRD *** is found from field emission scanning electron microscopy that the interlocked grain like microstructure developed in BMAPS glass-ceramics（being heat-treated at 1050 ℃ for 4 h） changed to denser house-of-cards like microstructure（containing smaller sized mica crystals） on addition of coinage *** of BMAPS base glass-ceramic was 2.60 g cm-3and marginally changed to 2.61-2.62 g cm-3on addition of Cu,Ag and *** change in microstructure resulted in the decrease of Vickers micro hardness value from 5.37 to 4.12,4.20 and 4.58 GPa on addition of Cu,Ag and Au,*** nanometai doped mica glass-ceramics containing interlocked microstructure with higher thermal expansion coefficient,hence,is suitable for high temperature sealing application（like solid oxide fuel cell）.
