Fungal endophytes have been isolated from almost every plant, infecting their hosts without causing visible disease symptoms, and yet have still proved to be involved in plant secondary metabolites accumulation. To decipher the possible physiological mechanisms of the endophytic fungus-host interaction, the role of protein phosphorylation and the relationship between endophytic fungus-induced kinase activity and nitric oxide （NO） and brassinolide （BL） in endophyte-enhanced volatile oil accumulation in Atractylodes lancea plantlets were investigated using pharmacological and biochemical approaches. Inoculation with the endophytic fungus Gilmaniella sp. ALl2 enhanced the activities of total protein phosphorylation, Ca2^-dependent protein kinase, and volatile oil accumulation in A. lancea plantlets. The upregulation of protein kinase activity could be blocked by the BL inhibitor brassinazole. Furthermore, pretreatments with the NO-specific scavenger cPTIO significantly reduced the increased activities of protein kinases in A. lancea plantlets inoculated with endophytic fungus. Pretreatments with different protein kinase inhibitors also reduced fungus-induced NO production and volatile oil accumulation, but had barely no effect on the BL level. These data suggest that protein phosphorylation is required for endophyte- induced volatile oil production in A. lancea plantlets, and that crosstalk between protein phosphorylation and the NO pathway may occur and act as a downstream signaling event of the BL pathway.

In order to improve the quality of oil peony transplanting,based on the characteristics of peony seedlings and the transplanting effect,the transplanting process was analyzed.A peony seedling transplanting machine was designed,which was mainly composed of a chain-clamp type planting mechanism(clamp,transmission chain,sprocket),slideway,profiling wheels,trencher,the power transmission system and *** the movement analysis of the planting mechanism and its operation process,its structural parameters were *** effective length of the clamp was determined as L_(1)=235 mm,the adjustment range was 235-285 mm,and the height of the planting trajectory was H=340 *** on the physical characteristics and mechanical characteristics of the peony seedlings,the parameters of the slideway and the putter were *** effective length of the slideway was determined as L_(2)=420 cm,the width T_(2)=105 mm,the distance between the slideways S=45 *** initial angle difference of the putter wasθ=13°,and the putter rotation angleβ=37°,putter height ratioε=*** on the above parameters and design requirements,the radius of the planting sprocket R_(1)=42 mm,R_(2)=80 mm.“Feng Dan”peony seedlings were used as transplantation test *** results showed that:when the planting frequency was 60-75 plants/min,the upright rate was 89.2%-92.8%,the leakage rate was 0-0.28%,the injury rate of seedlings was 0,and the qualified rate of planting depth was 91.7%-95.3%,the variation coefficient of plant spacing was 0.25%-0.54%.This machine can effectively improve the quality and efficiency of transplanting,meeting the requirements of mechanical design and production needs.

Techno-economic Appraisal of Biodiesel from Jatropha curcas： An Egyptian Case Study

Biodiesel （fatty acid methyl ester） has been identified as a non-toxic biodegradable alternative fuel that is obtained from renewable sources. Over the last decade, there has been increasing interest in producing biodiesel from conventional sources such as soybean, canola, sunflower, and coconut oils. Current efforts are directed towards the development of new non-edible resources. Among these Jatropha Curcas comes at the forefront. In Egypt, Jatropha Curcas has grown successfully using primary treated wastewater. Also, extensive R＆D efforts identified the optimum conditions for the various processing stages namely crushing, extraction, transesterification and purification. Based on the research findings, the techno-economic appraisal of biodiesel production from Jatropha Curcas is conducted. Two nominal capacities namely 8,000 and 50,000 metric tons/yr have been proposed. Several scenarios have been formulated to take into consideration varying productivity （3.4 to 5.8 ton fruits per 4,000 m^2 （acre）） and varying recovery rates of oil from seeds. Economic indicators including capital and production costs for the various processing stages and revenues according to current prices of oil and cake have been obtained. The price of biodiesel that provides a simple rate of return （SRR） on investments of 10% was in the range of $0.3-0.7/liter for the different assumed scenarios which is lower than the prevailing price of biodiesel （about $1/liter） in the US. Thus, in view of experimental results and economic assumptions, there are positive prospects for the production of biodiesel from Jatropha Curcas under Egyptian conditions.

Modifying plant oils for Use as Fuel in Rural Contexts Tanzania: Techno-Economic Analysis

Techno-economic analysis of a small-scale Modified plant oil (MPO) production plant that has an annual production capacity of 15,072,741 kg of MPO (batch process) was carried out to estimate the capital and operating costs of a plant. The analysis was done by using a computer model that was designed and simulated with an aid of SuperPro Designer (Version 4.32) software. The specified feedstock was crude Jatropha oil (JO) and the main product was MPO. The major processes involved were degumming, neutralisation and blending. Degumming involved the removal of gums or phospholipids, and two methods were used: water degumming and acid degumming, whereas blending involved mixing of degummed or purified JO with natural gas condensate (NGC) modifier to lower the viscosity of JO. From techno-economic analysis of the process, it was found that the total capital investment of a plant was about US $ 10,222,000 and the predicted unit production cost of MPO was US $ 1.315/kg at a value of US $ 1.0/kg of JO. The economic feasibility of MPO production was found to be highly influenced by the price of feedstock, which contributed about 95% of the total annual production cost. The relationship between plant throughput and unit cost of producing MPO showed that unit production cost was very sensitive to production rate at low annual throughputs. The MPO cost showed a direct linear relationship with the cost of JO, with a change of US $ 0.50/kg of MPO in MPO cost in every change of US $ 0.50/kg of JO in JO price. The process technology simulated was found to be economically viable and can be implemented in rural setting, taking into consideration Tanzania’s rural situation.

Dynamic changes of nutrient content in herbaceous peony seed

Herbaceous peony is a promising oilseed crop.‘Hangshao’and‘Fenghuang Niepan’are two cultivars with high yield and oil content In this study,seed reserved nutrient of these two genotypes were investigated during seed development *** showed that‘Hangshao’and‘Fenghuang Niepan’matured approximately 85 d after flowering(DAF).The development p rocess could be divided into four stages:seed formation and enlargement stage(S1),seed inclusion and enrichment stage(S2),crude fat rapid accumulation stage(S3)and dehydration and color transition stage(S4).During this process,100-seed fresh weight(HFW)and 100-seed dry weight(HDW)increased,whereas water content(WC)***,HDW and WC for‘Hangshao’seed at 85 DAF were28.56 g,20.78 g and 37.50%respectively,whereas those for‘Fenghuang Niepan’seed were 27.2 g,19.78 g,and30.85%*** terms of the primary metabolites measuring,soluble protein content was consistently low(only 2.24%-3.15%),while starch content was relatively high(5.60%-24.81%).They displayed a trend of gradual increasing in early stage,followed by rapid *** fat tended to accumulate continuously with a linear upward trend starting at 40 DAF,and its maximum level was reached at 85 DAF(27.57%for‘Hangshao’and29.57%for‘Fenghuang Niepan’).For the secondary metabolites determination,total phenols content was relatively steady,maintaining at 107.7-129.9 mg/g for both cultivars,while total flavonoids content varied within25.33-65.33 mg/g for‘Hangshao’and 24.33-52.37 mg/g for Tenghuang Niepan’.The maximum level for both cultivars occurred at 55 ***,crude fat content was found to be highly correlated with WC by correlation analysis and linear regression model definition(P≤0.01 and r=0.954).Thus,WC was recommended for evaluating crude fat in peony *** findings could provide scientific reference for high-yielding production practice for oleaginous peony plant.
