In our previous study,we investigated the dynamic expression of cytokines in the distal nerve stumps after peripheral nerve injury using microarray analysis,which can characterize the dynamic expression of *** the present study,we used a rat model of right sciatic nerve transection to examine changes in the expression of cytokines at 1,7,14 and 28 days after injury using protein microarray *** were increased in the distal nerve stumps at 1–14 days post nerve ***,growth factors and growth factor-related proteins were mainly upregulated in the proximal nerve *** P-values of the inflammatory response,apoptotic response and cell-cell adhesion in the distal stumps were higher than those in the proximal nerve stumps,but the opposite was observed for *** number of cytokines related to axons in the distal stumps was greater than that in the proximal stumps,while the percentage of cytokines related to axons in the distal stumps was lower than that in the proximal nerve *** of the results revealed the specific expression patterns and differences in cytokines in and between the proximal and distal nerve *** findings offer potential therapeutic targets and should help advance the development of clinical treatments for peripheral nerve *** for animal use in this study was obtained from the Animal Ethics Committee of the Chinese pla General hospital on September 7,2016(approval no.2016-x9-07).

AIM： To study the loss of heterozygosity （LOH） on 12p12-13 in Chinese colon carcino.a patients.METHODS： DNA was extracted from 10 specimens of cancer tissue, 10 specimens of adjacent tissue and 10 specimens of no.mal tissue, respectively. LOH of Kras2 gene was analyzed by polymerase chain reaction （PCR） and denaturing polyacrylamide gel electrophoresis using 11 microsatellite markers on 12p-12-13.RESULTS： LOH of Kras gene was detected at least on one marker of 12p-12-13 in 30% （3/10） of adjacent tissue specimens. The highest frequency of LOH was identified on D12S1034 in 28.57% （2]7） of adjacent tissue specimens. LOH was detected at least on one marker of 12p12-13 in 60% （6/10） of carcino.a tissue specimens, the most frequent LOH was found on D12S1034 and D12S1591 in 42.86% （3/7） of carcino.a tissue specimens. LOH was detected in 30% （3/10） of carcino.a tissue specimens, 30% （3/10） of adjacent tissue specimens, and no.signal in 1% （1/0） carcino.a tissue specimen. The occurrence of LOH did no. correlate with sex, age, tumor size and lymph no.e metastasis.CONCLUSION： Geno.ic instability may occur on 12p-12-13 of Kras2 gene in the development and progression of colon carcino.a. The high LOH of Kras2 gene may directly influence the transcription and translation of wild type Kras2 gene.

BACKGROUND Coronary artery disease(CAD)is associated with *** role of inflammation in the association of CAD with cancer remains *** study investigated whether inflammation could impact the relationship between CAD and lung *** The study involved 96 newly diagno.ed lung cancer patients without receiving anti-cancer therapy and 288 matched no.-cancer *** the patients underwent coronary angiography and were free from previous percutaneous coronary inter-vention or coronary artery bypass *** score(SXscore)were used to assess severity of *** SXscore(SXhigh)grade was defined as SXscore>16(highest quartile).Neutrophil-to-lymphocyte ratio(NLR)served as an inflammatory ***-high grade referred to NLR>2.221(median).RESULTS Among 384 study patients,380 patients(98.96%)had NLR value(median:2.221,interquartile range:1.637-3.040).Com-pared to no.-cancer patients,lung cancer patients had higher rate of SXhigh among total study patients(P=0.014)and among pat-ients with NLR-high(P=0.006),but had no. significantly higher rate of SXhigh among patients with NLR-low(P=0.839).Multiv-ariate logistic regression analysis showed that SXhigh was associated with lung cancer[odds ratio(OR)=1.834,95%CI:1.063-3.162,P=0.029].Subgroup analysis showed that SXhigh was associated with lung cancer among patients with NLR-high(OR=2.801,95%CI:1.355-5.794,P=0.005),however,the association between SXhigh and lung cancer was no. significant among pati-ents with NLR-low(OR=0.897,95%CI:0.346-2.232,P=0.823).CONCLUSIONS Inflammation could lead different association between anatomical severity of CAD and lung *** of CAD was significantly associated with increased risk of lung cancer among patients with high inflammation rather than am-ong patients with low inflammation.

AIM: To evaluate the efficacy and safety of corneal collagen crosslinking (01) to prevent the progression of post-laser in situ keratomileusis (LASIK) corneal ectasia. METHODS: In a prospective, no.randomized, single-centre study, CXL was performed in 20 eyes of 11 patients who had LASIK for myopic astigmatism and subsequently developed keratectasia. The procedure included instillation of 0.1% riboflavin-20% dextrane solution 30 minutes before UVA irradiation and every 5 minutes for an additional 30 minutes during irradiation. The eyes were evaluated preoperatively and at 1-, 3-, 6-, and 12-month intervals. The complete ophthalmologic examination comprised uncorrected visual acuity, best spectacle-corrected visual acuity, endothelial cell count, ultrasound pachymetry, corneal topography, and in vivo confocal microscopy. RESULTS: CXL appeared to stabilise or partially reverse the progression of post-LASIK corneal ectasia without apparent complication in our cohort. UCVA and BCVA improvements were statistically significant (Pno. deteriorate. Mean thinnest cornea pachymetry increased significantly. CONCLUSION: The results of the study showed a long-term stability of post-LASIK corneal ectasia after crosslinking without relevant side effects. It seems to be a safe and promising procedure to stop the progression of post-LASIK keratectasia, thereby avoiding or delaying keratoplasty.

Regenerative medicine has rapidly developed over the past decade and created new opportunities to repair or replace tissue or organ function lost because of congenital defects, age, diseases, or serious damage （Cheng et al., 2016a; Cheng et al., 2016b）. Regenerative medicine strategies in- clude the transplantation of bioactive factors, stem cells, or biomaterials, even the induced regeneration in a de no.o, depending on the application （Fu, 2014a; Huang and Fu, 2014）. However, there are several limitations to the use of regenerative medicine in the clinic with respect to using stem cells and biomaterials.

Previous studies have shown that vagus nerve stimulation can improve the progno.is of trau- matic brain injury. The aim of this study was to elucidate the mechanism of the neuroprotective effects of vagus nerve stimulation in rabbits with brain explosive injury. Rabbits with brain ex- plosive injury received continuous stimulation （10 V, 5 Hz, 5 ms, 20 minutes） of the right cervical vagus nerve. Tumor necrosis factor-a, interleukin-l~ and interleukin-10 concentrations were detected in serum and brain tissues, and water content in brain tissues was measured. Results showed that vagus nerve stimulation could reduce the degree of brain edema, decrease tumor necrosis factor-a and interleukin-1β concentrations, and increase interleukin-10 concentration after brain explosive injury in rabbits. These data suggest that vagus nerve stimulation may exert neuroprotective effects against explosive injury via regulating the expression of tumor necrosis factor-a, interleukin-1 β and interleukin-10 in the serum and brain tissue.

AIMTo analyze the low weight gain (WG) from birth to 4 and 6wk of life to predict the development of retino.athy of prematurity (ROP) among very low birth weight (VLBW) preterm *** hundred and three newborns with VLBW were analyzed. Body weight measurements were recorded weekly. In all patients, the proportion of the WG was defined as the preterm weight measured at the 4th and 6th weeks of life minus the birth weight (BW) divided by the BW. Other risk factors for ROP were also *** gestational age and mean BW of the whole cohort were 29.56±1.44wk and 1270.58±176.18g respectively. WG proportion at 4wk postnatal age (18.89%±13.58%) were significantly lower in infants with ROP (P=0.003). WG proportion at 6wk was no. different between ROP and no.ROP group (42.48%±20.36% vs 46.43%±15.65% P=0.118). When all the other risk factors signiﬁcant for ROP were included in the logistic regression poor WG did no. arise as an independent risk factor. Area under the ROC curve was 0.591 (95%CI: 0.515-0.666; P=0.016). For ROP, the best discriminative cutoff of 18.06% of the proportional WG at the 4th week over the BW, sensitivity and specificity values were 67.3% and 50.0% *** WG proportion in the first 4wk of life is maybe an additional predictor of ROP in very low BW infants. Preterm babies with low BW and low WG proportion should be followed closely for ROP.

Dental implantation is an effective standard treatment modality to restore missing teeth and maxillofacial defects. However, in diabetics there is an increased risk for implant failure due to impaired peri-implant osseous healing. Early topical insulin treat- ment was recently shown to no.malize diabetic bone healing by rectifying impairments in osteoblastic activities. In this study, insulin/poly(lactic-co-glycolic acid) (PLGA) microspheres were prepared by a double-emulsion solvent evaporation method. Microspheres were then incorporated in fibrin gel to develop a local drug delivery system for diabetic patients requiring im- plant treatment. In vitro release of insulin from fibrin gel loaded with these microspheres was assessed, and sustained prolonged insulin release over 21 days ascertained. To assess the bioactivity of released insulin and determine whether slow release might improve impaired diabetic bone formation, 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT), alkaline phosphatase (ALP) activity, mineralized no.ule formation, and ELISA (enzyme-linked immuno.orbent assay) assays were performed. The insulin released from the drug delivery system stimulated cell growth in previously inhibited cells, and ameliorated the impaired bone-forming ability of human MG-63 cells under high glucose conditions. Fibrin gel loaded with insulin/PLGA microspheres shows potential for improving peri-implant bone formation in diabetic patients.

AIM To investigate the association between serum human epidermal growth factor receptor 2(HER2) extracellular domain(ECD) and tissue HER2 status, and the progno.tic value of serum HER2 ECD in patients with gastric *** A total of 239 patients with gastric cancer were enrolled from December 2012 to June 2013. Serum HER2 ECD was determined by chemiluminescent assay, and tissue HER2 status was evaluated by immuno.istochemistry and fluorescence in situ hybridization assay. A receiver operating characteristic(ROC) curve was plotted to identify the optimal cut-off value for serum HER2 ECD assay for predicting survival in gastric cancer *** Serum HER2 ECD was significantly correlated with tissue HER2 status(P < 0.001), tumor size(P < 0.001), and intestinal type of gastric cancer(P =0.021). Serum HER2 ECD levels differed significantly between patients with HER2-positive tissue expression and those with HER2-negative tissue expression. ROC analysis yielded an area under the curve value of 0.79(95%CI: 0.71-0.87, P < 0.001), with a sensitivity and specificity of 0.54(95%CI: 0.37-0.70) and 0.93(95%CI: 0.88-0.96), respectively. With a cut-off value of 24.75 ng/m L, high serum HER2 ECD had a negative impact on overall survival of the patients(HR: 1.93, 95%CI: 1.32-4.38, P = 0.006). CONCLUSION Serum HER2 ECD could be a highly specific surrogate biomarker for tissue HER2 status in gastric cancer. Optimal cut-off criteria for predicting survival should be established.
