Integrins and their potential roles in mammalian pregnancy

Integrins are a highly complex family of receptors that, when expressed on the surface of cells, can mediate reciprocal cell-to-cell and cell-to-extracellular matrix(ECM) interactions leading to assembly of integrin adhesion complexes(IACs) that initiate many signaling functions both at the membrane and deeper within the cytoplasm to coordinate processes including cell adhesion, migration, proliferation, survival, differentiation, and metabolism. All metazoan organisms possess integrins, and it is generally agreed that integrins were associated with the evolution of multicellularity, being essential for the association of cells with their neighbors and surroundings, during embryonic development and many aspects of cellular and molecular biology. Integrins have important roles in many aspects of embryonic development, normal physiology, and disease processes with a multitude of functions discovered and elucidated for integrins that directly influence many areas of biology and medicine, including mammalian pregnancy, in particular implantation of the blastocyst to the uterine wall, subsequent placentation and conceptus(embryo/fetus and associated placental membranes) development. This review provides a succinct overview of integrin structure, ligand binding, and signaling followed with a concise overview of embryonic development, implantation, and early placentation in pigs, sheep, humans, and mice as an example for rodents. A brief timeline of the initial localization of integrin subunits to the uterine luminal epithelium(LE) and conceptus trophoblast is then presented, followed by sequential summaries of integrin expression and function during gestation in pigs, sheep, humans, and rodents. As appropriate for this journal, summaries of integrin expression and function during gestation in pigs and sheep are in depth, whereas summaries for humans and rodents are brief. Virus replication relies on complex interactions between viral *** the case of African swine fever virus(ASFV),only a few such interactions have been identified so *** this study,we demonstrate that ASFV protein p72 interacts with p11.5 using co-immunoprecipitation and liquid chromatography-mass spectrometry(LC-MS).It was found that protein p72 interacts specifically with p11.5 at sites amino acids(aa)1–216 of p72 and aa 1–68 of *** assess the importance of p11.5 in ASFV infection,we developed a recombinant virus(ASFVGZΔA137R)by deleting the A137R gene from the ASFVGZ *** with ASFVGZ,the infectious progeny virus titers of ASFVGZΔA137R were reduced by approximately 1.0 *** addition,we demonstrated that the growth defect was partially attributable to a higher genome copies-to-infectious virus titer ratios produced in ASFVGZΔA137R-infected MA104 cells than in those infected with *** finding suggests that MA104 cells infected with ASFVGZΔA137R may generate larger quantities of noninfectious ***,we found that p11.5 did not affect virus-cell binding or ***,we show for the first time the interaction between ASFV p72 and *** results effectively provide the relevant information of the p11.5 *** results extend our understanding of complex interactions between viral proteins,paving the way for further studies of the potential mechanisms and pathogenesis of ASFV infection.

The phenotypic diversity resulting from artificial or natural selection of sheep has made a significant contribution to human *** sheep are a local sheep breed unique to China with high reproductive rates and rapid *** selection signatures have been widely used to investigate the genetic mechanisms underlying phenotypic variation in ***,we conduct whole-genome sequencing of 207 Hu sheep and compare them with the wild ancestors of domestic sheep(Asiatic mouflon)to investigate the genetic characteristics and selection signatures of Hu *** on six signatures of selection approaches,we detect genomic regions containing genes related to reproduction(BMPR1B,BMP2,PGFS,CYP19,CAMK4,GGT5,and GNAQ),vision(ALDH1A2,SAG,and PDE6B),nervous system(NAV1),and immune response(GPR35,SH2B2,PIK3R3,and HRAS).Association analysis with a population of 1299 Hu sheep reveals that those missense mutations in the GPR35(GPR35 g.952651 A>G;GPR35 g.952496 C>T)and NAV1(NAV1 g.84216190 C>T;NAV1 g.84227412 G>A)genes are significantly associated(P<0.05)with immune and growth traits in Hu sheep,*** research offers unique insights into the selection characteristics of Hu sheep and facilitates further genetic improvement and molecular investigations.

Betulinic acid protects against ovarian impairment by decreasing F-2 toxin-induced oxidative stress and inflammation associated with the downregulation of p38 expression in mice

F-2 toxin is an estrogenic mycotoxin that causes reproductive disorders in *** acid(BA)is a natural pentacyclic lupane-structure triterpenoid that has diverse pharmacological *** this study,the antioxidative and anti-inflammatory effects of BA and its underlying mechanism are explored in F-2 toxin-triggered mouse ovarian *** found that BA alleviated the F-2 toxin-induced ovarian impairment by stimulating follicle growth,reducing inflammatory cell infiltration,repairing damaged mitochondria and endoplasmic ***,BA not only reversed F-2 toxin-induced reduction of follicle stimulating hormone(FSH)and luteinizing hormone(LH)levels in the serum,but also restrained the protein expression of the estrogen receptors a(ERa)and ERβ.Moreover,BA restored the balance of F-2 toxin-induced ovarian redox system ***,we found that 0.25 mg/kg BA played an anti-inflammatory role in the F-2 toxin-induced ovarian impairment by decreasing interleukin-1β(IL-1β).IL-6,and tumor necrosis factor-α(TNF-α)mRNA expression,as well as inhibiting p38 protein *** data demonstrated that BA exerts its protective effect on F-2 toxin-induced ovarian oxidative impairment and inflammation by inhibiting p38 expression,which implies a natural product-based medicine to ameliorate F-2 toxin-caused female reproductive toxicity and provides a detoxifying method for food contaminated by mycotoxin.

Epidemiology and genetic characterization of tet(X4)-positive Klebsiella pneumoniae and Klebsiella quasipneumoniae isolated from raw meat in Chengdu City, China

The rapid spread of mobile tigecycline resistance presents a significant public health threat,particularly with the increasing prevalence of tet(X4)-positive Enterobacterales across various *** study aimed to inves-tigate the epidemic features and transmission dynamics of tet(X4)-positive Klebsiella pneumoniae(***-niae)through the analysis of 206 raw meats,including pork(n=182),beef(n=16),duck(n=5),and chicken(n=3).These samples were collected from schools,markets,and restaurants in Chengdu City,China.A total of 25 isolates were obtained from 13 administrative *** isolates exhibited resistance to tetracycline,tigecycline,ampicillin,chloramphenicol,and *** half of the isolates also demon-strated resistance to streptomycin(80%),sulfamethoxazole/trimethoprim(72%),ciprofloxacin(64%),and ampicillin/sulbactam(56%).Among these strains,14 distinct sequence types(STs)were identified,revealing evidence of inter-regional clonal spread,notably among 9 *** *** analysis revealed the presence of two *** ST5 closely resembling hypervirulent *** from ***,12 isolates were capable of transferring tigecycline resistance to Escherichia coli *** plasmid analysis showed that the tet(X4)-harboring plasmids in *** could be classified into four types,primarily belonging to the IncFIA(HI1)/HI1A/HI1B hybrid plasmid(n=16)and IncFII plasmid(n=7),which significantly contributed to the cross-species dissemination of tet(X4).In summary,this study highlights the prevalence of MDR tet(X4)-positive *** in Chengdu,driven predominantly by clonal expansion and plasmid-mediated horizontal gene *** findings emphasize the importance of contin-uous surveillance of tet(X4)-positive *** in raw meat and the implementation of effective measures to control their spread.
