Morphological, phenological, and transcriptional analyses provide in sight into the diverse flowering traits of a mutant of the relic woody plant Liriodendron chinense

Flowering is crucial to plant reproduction and controlled by multiple ***,the mechanisms underlying the regulation of flowering in perennial plants are still largely ***,we first report a super long blooming 1(slbl)mutant of the relict tree Liriodendron chinense possessing a prolonged blooming period of more than 5 months,in contrast to the 1 month blooming period in the wild type(WT).Phenotypic characterization showed that earlier maturation of lateral shoots was caused by accelerated axillary bud fate,leading to the phenotype of continuous flowering in slbl *** transcriptional activity of genes related to hormone signaling(auxin,cytokinin,and strigolactone),nutrient availability,and oxidative stress relief further indicated active outgrowth of lateral buds in slbl ***,we discovered a unique FT splicing variant with intron retention specific to slbl mutants,representing a potential causal mutation in the slbl ***,most slbl inbred offspring flowered precociously with shorter juvenility(~4 months)than that(usually 8-10 years)required in WT plants,indicating heritable variation underlying continuous flowering in slbl *** study reports an example of a perennial tree mutant that flowers continuously,providing a rare resource for both breeding and genetic research.

CTAB-silica Method for DNA Extraction and Purification from Castanea mollissima and Ginkgo biloba

A new method CTAB-silica for DNA extraction and purification from the leaves and buds of Castanea mollissima and Ginkgo biloba was tested. The method is based on the silica-based purification protocol developed by Boom et al. (1990). By modifying the protocol, plant genome DNA could be extracted easily from dormant buds, mature leaves, and other parts of plant. Our results showed that the purified DNA was of high purity and could be analyzed by PCR. Furthermore, this CTAB-silica method took much less time for a successful DNA purification process compared to the traditional methods (CTAB and SDS). By our method, the suitable DNA can be extracted and purified from over 10 plant samples by one person in an hour.

Epigenetic mechanisms are integral to plant growth, development, and adaptation to environmental stimuli. Over the past two decades,our comprehension of these complex regulatory processes has expanded remarkably, producing a substantial body of knowledge on both locus-specific mechanisms and genome-wide regulatory patterns. Studies initially grounded in the model plant Arabidopsis have been broadened to encompass a diverse array of crop species, revealing the multifaceted roles of epigenetics in physiological and agronomic traits. With recent technological advancements, epigenetic regulations at the single-cell level and at the large-scale population level are emerging as new focuses. This review offers an in-depth synthesis of the diverse epigenetic regulations, detailing the catalytic machinery and regulatory functions. It delves into the intricate interplay among various epigenetic elements and their collective influence on the modulation of crop traits. Furthermore, it examines recent breakthroughs in technologies for epigenetic modifications and their integration into strategies for crop improvement. The review underscores the transformative potential of epigenetic strategies in bolstering crop performance, advocating for the development of efficient tools to fully exploit the agricultural benefits of epigenetic insights.

Selection of female parents with high fertility and high combining abilities for cross-breeding Populus tomentosa

To select maternal parents with high fertility and high combining ability during cross breeding of Populus tomentosa Carr., the ploidy level of 75 female clones was determined using flow cytometry. In addition, genetic variations and phenotypic correlations of seed traits and early growth traits, as well as the general combining ability(GCA) for seedling height(SH) and basal diameter(BD) of all diploid half-sib families were examined. A total of 26 natural triploid clones were identified. For all diploid families, family effects were significant for inflorescence length(IL), thousand kernel weight(TKW), seedling emergence rate(SER), and for SH and BD. There was a positive correlation between TKW and other traits,excluding IL. The seed-bearing coefficient(NS 9 SER)ranged from zero to 32.4%. Clones 3-10-2, 2-8, 3119,3206, and 3-10-1 had the best performance of the GCA for SH and BD. Based on the fertility and GCA for SH and BD, clone 3-10-2 and clone 3119 could be used as female parents for controlled cross-breeding of P. tomentosa.

Site index for Chinese fir plantations varies with climatic and soil factors in southern China

Chinese fir[Cunninghamia lanceolata(Lamb.)Hook.]has a large native distribution range in southern ***,we tested differences in productivity of Chinese fir plantations in different climatic regions and screened the main environmental factors affecting site productivity in each *** of a Chinese fir site index with climatic factors and the soil physiochemical properties of five soil layers were examined in a long-term positioning observation trial comprising a total of 45 permanent plots in Fujian(eastern region in the middle subtropics),Guangxi(south subtropics)and Sichuan(central region in the middle subtropics)in southern *** mixed effects models were developed to predict the site index for Chinese fir,which was found to vary significantly among different climatic *** P,total N,bulk density and total K were dominant predictors of site index in three climatic *** regional linear mixed models built using these predictors in the three climatic regions fit well(R~2=0.86–0.97).For the whole study area,the available P in the 0–20-cm soil layer and total N in the 80–100-cm soil layer were the most indicative soil *** was the most important climatic variable influencing the site *** model evaluation results showed that the fitting performance and prediction accuracy of the global site index model using the climatic region as the dummy variable and random parameters and the most important soil factors of the three climatic regions as predictors was higher than that of global site index model using the climatic variable and the most indicative soil variables of the whole study *** results will help with further evaluation of site quality of Chinese fir plantations and the selection of its appropriate sites in southern China as the climatic changes.

Seasonal dynamics of foliar δ^(13)C and nutrient concentration of Chinese chastetree and spine jujube in foothill rock outcrop habitat of the southern Taihang Mountains, Central China

In this study, in situ investigation was conducted on Chinese chastetree(Vitex negundo var. heterophylla)and spine jujube(Ziziphus jujuba var. spinosa) during early, developed, and closing succession stages. Monthly data were recorded for foliar δ^(13)C values and foliar N, P,and K concentrations from May to October. Foliar δ^(13)C values declined with time and differences by succession stage varied by species. Older spine jujube displayed higher foliar N concentration while foliar P concentration was inversely related with N. Chinese chastetree had lower foliar K concentration than spine jujube. Foliar C/N ratio declined during the dry season and increased in the wet season. Foliar N/P ratio increased during the dry season and declined to its initial level until October, when it increased again. Foliar δ^(13)C value was positively correlated with foliar N concentration. Foliar P and K concentrations were positively correlated to each other. In conclusion, the two shrubs had contrasting strategies of water use efficiency during their succession, but water use efficiency of both species was promoted by N availability and limited by P availability.

Characterization and comparison of chloroplast genomes from two sympatric Hippophae species(Elaeagnaceae)
