New perspectives on the genetic structure of dotted gizzard shad(Konosirus punctatus)based on RAD-seq

To maintain,develop and rationally utilize marine organisms,understanding their genetic structure and habitat adaptation pattern is necessary.Konosirus punctatus,which is a commercial fish species inhabiting the Indo-west Pacific Ocean,has shown an obvious annual global capture and aquaculture production decline due to climate changes and human activities.In the present study,restriction-site associated DNA sequencing(RAD-seq)was used to describe its genome-wide single nucleotide polymorphisms panel(SNPs).Among 146 individuals collected at nine locations scattered in China,Korea and Japan,a set of 632,090 SNPs were identified.Population genetic analysis showed that K.punctatus individuals were divided into two significant genetic clusters.Meanwhile,potential genetic differentiation between northern and southern population of K.punctatus was found.Treemix results indicated that gene flow existed among sampling locations of K.punctatus,especially from southern Japan to others.Moreover,candidate genes associated with habitat adaptations of K.punctatus were identified,which are involved in diverse physiological processes of K.punctatus including growth and development(e.g.,KIDINS220,PAN3),substance metabolism(e.g.,PGM5)and immune response(e.g.,VAV3,CCT7,HSPA12B).Our findings may aid in understanding the possible mechanisms for the population genetic structure and local adaptation of K.punctatus,which is beneficial to establish the management and conservation units of K.punctatus,guiding the rational use of resources,with reference significance for a profound understanding of the adaptative mechanisms of other marine organisms to the environment.

Application of the finite analytic numerical method to a flowdependent variational data assimilation

An anisotropic diffusion filter can be used to model a flow-dependent background error covariance matrix,which can be achieved by solving the advection-diffusion equation.Because of the directionality of the advection term,the discrete method needs to be chosen very carefully.The finite analytic method is an alternative scheme to solve the advection-diffusion equation.As a combination of analytical and numerical methods,it not only has high calculation accuracy but also holds the characteristic of the auto upwind.To demonstrate its ability,the one-dimensional steady and unsteady advection-diffusion equation numerical examples are respectively solved by the finite analytic method.The more widely used upwind difference method is used as a control approach.The result indicates that the finite analytic method has higher accuracy than the upwind difference method.For the two-dimensional case,the finite analytic method still has a better performance.In the three-dimensional variational assimilation experiment,the finite analytic method can effectively improve analysis field accuracy,and its effect is significantly better than the upwind difference and the central difference method.Moreover,it is still a more effective solution method in the strong flow region where the advective-diffusion filter performs most prominently.

The application of protease in aquaculture: Prospects for enhancing the aquafeed industry

Low-fishmeal and protein-saving diets are two prominent nutritional strategies utilized to address challenges related to the scarcity and sustainability of protein sources in aquaculture. However, these diets have been associated with adverse effects on the growth performance, feed utilization, and disease resistance of aquatic animals. To mitigate these challenges, exogenous protease has been applied to enhance the quality of diets with lower protein contents or fishmeal alternatives, thereby improving the bioavailability of nutritional ingredients. Additionally, protease preparations were also used to enzymatically hydrolyze fishmeal alternatives, thus enhancing their nutritional utilization. The present review aims to consolidate recent research progress on the use of protease in aquaculture and conclude the benefits and limitations of its application, thereby providing a comprehensive understanding of the subject and identifying opportunities for future research.

Identification and Expression Profile of a Neuropeptide LFRFamide-Like Gene During Different Stages of Gonadal Development in the Cephalopod Sepia pharaonis

Neuropeptides are widely distributed in vertebrates and invertebrates,regulating a variety of physiological activities in the organisms,such as metabolism,feeding and reproduction.In this study,to explore the function of neuropeptide LFRFamide in Sepia pharaonis,the full-length cDNA of LFRFamide-like gene(named SpLFRFL,MG869822.1)was identified with rapid amplification of cDNA ends(RACE)method.The sequence of SpLFRFL was 860 bp in length and encoded 188 amino acids containing 4 different mature peptides:1 copy of PHTPFRFamide,NSLFRFamide,TIFRFamide,and 3 copies of GNLFRFamide.Multiple alignment and phylogenetic analysis results showed that SpLFRFL shared high identity with LFRFamides of Sepia officinalis and Sepiella japonica and had the closest relationship with them.Through quantitative Real-time PCR(qRT-PCR),it was found that the SpLFRFL gene was highly expressed in the optic lobe and brain at three different stages during gonad development in both genders.Moreover,the four mature peptides at a concentration of 0.01μmol L^(−1) could inhibit the protein synthesis in the Chinese hamster ovary cell strain-K1(CHOK1)induced by SpGnRH.These data suggest that SpLFRFL might be involved in the development and reproduction of S.pharaonis.The results can contribute to future studies on neuropeptide evolution and function and benefit the cuttlefish farming.

Characteristics,origins,and significance of pyrites in deep-water shales

As important metal sulfides in the geochemical cycle of sulfur,the characteristics and formation processes of pyrites can provide useful clues regarding their environment.Based on previous findings,shale pyrites were divided into three major classes(euhedral pyrites,framboidal pyrites(framboids)and metasomatic pyrites)and six sub-classes in this study.At the microscopic scale,each type of pyrite is associated with a different formation process.Framboids are formed by burst nucleation in environments with a homogeneous distribution of nutrients while euhedral pyrites are usually formed on pre-existing sites(such as=FeS on the minerals surface)in the heterogeneous system.Metasomatic pyrites formed by the replacement of other ions in accountable material by iron ions and hydrogen sulfide ions in hydrothermal events.The morphology and isotope value of pyrite provide information to track the origins of their nutrient and characteristics of sulfur and iron pools.In addition,the trace element content of pyrite can serve as a proxy for paleo-ocean trace element abundance,indicating changes in atmospheric oxygen content.Additionally,pyrite can also serves as an indicator of shale gas reservoirs.

Full T-cell activation and function in teleosts require collaboration of first and co-stimulatory signals

Mammalian T-cell responses require synergism between the first signal and co-stimulatory signal.However,whether and how dual signaling regulates the T-cell response in early vertebrates remains unknown.In the present study,we discovered that the Nile tilapia(Oreochromis niloticus)encodes key components of the LAT signalosome,namely,LAT,ITK,GRB2,VAV1,SLP-76,GADS,and PLC-γ1.These components are evolutionarily conserved,and CD3εmAb-induced T-cell activation markedly increased their expression.Additionally,at least ITK,GRB2,and VAV1 were found to interact with LAT for signalosome formation.Downstream of the first signal,the NF-κB,MAPK/ERK,and PI3K-AKT pathways were activated upon CD3εmAb stimulation.Furthermore,treatment of lymphocytes with CD28 mAbs triggered the AKT-mTORC1 pathway downstream of the co-stimulatory signal.Combined CD3εand CD28 mAb stimulation enhanced ERK1/2 and S6 phosphorylation and elevated NFAT1,c-Fos,IL-2,CD122,and CD44 expression,thereby signifying T-cell activation.Moreover,rather than relying on the first or co-stimulatory signal alone,both signals were required for T-cell proliferation.Full T-cell activation was accompanied by marked apoptosis and cytotoxic responses.These findings suggest that tilapia relies on dual signaling to maintain an optimal T-cell response,providing a novel perspective for understanding the evolution of the adaptive immune system.

Auto-Detection Method Using Convolution Neural Network for Bottom-Simulating Reflectors

In studies on gas hydrate,bottom-simulating reflectors(BSR)are used to determine the potential hydrate-bearing sedimentary layers.Usually,BSR detection is performed manually by experienced interpreters.Therefore,a method for implementing an auto-matic BSR detection process should be established.In this study,we develop a novel architecture for BSR characterization using the convolutional neural network(CNN)technique.We propose the use of Stokes’transform(ST)to obtain a time-frequency spectrum for the input of CNN.ST fully uses the frequency content of the seismic data,and a part of the 3D seismic data collected from the Blake Ridge is utilized to train the CNN.Synthetic seismic records with variable signal-to-noise ratios(SNR),as well as Blake Ridge seismic data,were used to validate the detection effect of the CNN.Results show that the CNN trained by this method exhibits excellent performance in noise-resistant testing and achieves an accuracy of more than 89% in field seismic data detection.

Numerical Simulation on Production Trials by Using Depressurization for Typical marine Hydrate Reservoirs:Well Type and Formation Dip

Natural gas hydrate has huge reserves and is widely distributed in marine environment.Its commercial development is of great significance for alleviating the contradiction between energy supply and demand.As an efficient research method,numerical simulation can provide valuable insights for the design and optimization of hydrate development.However,most of the current production models simplify the reservoir as a two-dimensional(2D)horizontal layered model,often ignoring the impact of formation dip angle.To improve the accuracy of production prediction and provide theoretical support for the optimization of production well design,two three-dimensional(3D)geological models with different dip angles based on the geological data from two typical sites are constructed.The vertical well,horizontal well and multilateral wells are deployed in these reservoirs with different permeabilities to perform production trial,and the sensitivity analysis of dip angles is also carried out.The short-term production behaviors in high and low permeability reservoirs with different dip angles are exhibited.The simulation results show that 1)the gas and water production behaviors for different well types in the two typical reservoirs show obviously different variation laws when the short-term depressurization is conducted in the inclined formation;2)the inclined formation will reduce the gas production and increase the water extraction,and the phenomena becomes pronounced as the dip angle increases,particularly in the low-permeability reservoirs;3)and the impact of formation dip on hydrate recovery does not change significantly with the variation of well type.

Current observation and analysis based on mooring systems in the Andaman Sea

The basic structure and intraseasonal evolution of currents in the southeastern Andaman Sea was analyzed based on data collected in 2017 from two subsurface moorings(C1 and C5).Periodic variation in the upper ocean currents of the Andaman Sea was investigated by combining observational and satellite data.Mooring observations show that rapid changes of current speed and direction occurred in May and June,with a significant increase in current velocity at the C1 mooring.In the second half of the year,southward flow dominated at the C1 mooring,and alternating northward and southward flows were evident at the C5 mooring during the same period but the northward flow prevailed in boreal winter.In addition,analysis of the power spectra of the upper currents revealed that the tidal period at both moorings is primarily semidiurnal with weaker energy than that of the low-frequency currents.The upper ocean currents at the C1 and C5 moorings exhibited intraseasonal variation of 30-60 d and 120 d,while the zonal current at the C1 mooring exhibited a notable period of approximately 180 d.Further analysis indicated that the variability of currents in the Andaman Sea is influenced primarily by equatorial Kelvin waves and Rossby wave packets.Moreover,our results suggest that equatorial Kelvin waves from the eastern Indian Ocean entered the Andaman Sea in the form of Wyrtki Jets and propagated primarily along two distinct pathways during the observation period.In addition to coastal boundary Kelvin waves,it was found that a branch of the Wyrtki Jet that directly enters the Andaman Sea and flows northward along the slope of the continental shelf,and reflected Rossby wave packets by topography.

Effects of alcohol on digestion,absorption and metabolism of sea cucumber saponins in healthy mice

Sea cucumber saponins have attracted more attention in recent years due to biological activities.It is a popular practice to soak sea cucumber in Baijiu at home and being applied to industrial manufacturing in China.However,knowledge of the effect of alcohol on the absorption and metabolism of sea cucumber saponins is limited.The effects of alcohol on digestion,absorption and metabolism of sea cucumber saponins in BALB/c mice were investigated after gavage and tail intravenous injection.The results showed that the content of saponins in serum and liver was significantly higher under the influence of alcohol than that in the control group after oral administration.Alcohol promoted the absorption of sea cucumber saponins prototype as well as inhibited the process of saponins being transformed into deglycositic metabolites in the small intestine.Moreover,sea cucumber saponins remained in circulation for a long time and alcohol slowed down the clearance of sea cucumber saponins under the influence of alcohol after intravenous injection.This confirmed the feasibility of marinating sea cucumber in Baijiu to improve the efficacy of saponins and provides an important theoretical basis for the utilization of sea cucumber and the development of sea cucumber liquor.
