Bacillus sp. JM7, a strain isolated from the deep-sea of the South China Sea, was found to efficiently degrade 79.4% native chicken feather within 30 h. Scanning electron microscopy analysis showed that JM7 strain could gradually degrade feather by modifying the microstructure of feather keratin. A total of 25 protease genes were predicted from the draft genome of JM7 strain, among which a predicted subtilisin-like serine protease(designated as Ker02562) was further characterized for its keratinolytic activity. The recombinant Ker02562 functioned at a wide range of temperatures from 30℃ to 60℃, with an optimum at 40–50℃. Ker02562 was highly active at various pHs ranging from 5.0 to 13.0, with a maximum activity observed at pH 7.0–9.0. Remarkably, recombinant Ker02562 was stable in extreme alkaline environments(pH 10–13), which was much better than most other reported keratinases. Collectively, these favorable properties could make Bacillus sp. JM7 and Ker02562 attractive to be applied in the detergent formulation and feather bioconversion.

Grain shape and size both determine grain weight and therefore crop yield. However, the molecular mechanisms controlling grain shape and size are still largely unknown. Here, we isolated a rice mutant, beak-shaped grain1 （bsgl）, which produced beak-shaped grains of decreased width, thickness and weight with a loosely interlocked lemma and palea that were unable to close tightly. Starch granules were also irregularly packaged in the bsgl grains. Consistent with the lemma and palea shapes, the outer parenchyma cell layers of these bsgl tissues developed fewer cells with decreased size. Map-based cloning revealed that BSG1 encoded a DUF640 domain protein, TRIANGULAR HULL 1, of unknown function. Quantitative PCR and GUS fusion reporter assays showed that BSG1 was expressed mainly in the young panicle and elongating stem. The BSG1 mutation affected the expression of genes potentially involved in the cell cycle and GW2, an important regulator of grain size in rice. Our results suggest that BSG1 determines grain shape and size probably by modifying cell division and expansion in the grain hull.

Invasive species generally possess unique characteristics that allow them to survive the invasion process in order to es- tablish and spread in new habitats. Successful invaders must resist both physical and physiological stresses associated with the changing environment. A common littoral barnacle, Chthamalus challengeri Hoek, 1883 （Crustacea, Cirripedia）, which is native to Japan, South Korea and northern China, has become established in the high-littoral zone adjacent to Yangshan Port, Shanghai, China. A comparison of the morphology of Chthamlus species from Zhoushan archipelago with previous description indicates the occur- rence of C. challengeri. The new immigrant becomes a dominant species in certain high-intertidal habitats of the adjacent area to of Yangshan Port. C. challengeri was found in part of sampling sites in Zhoushan in 2010; however, it dispersed to all the eleven sam- piing sites in 2012. Densities of *** had increased over 10 times in the last 2 years, with the highest mean value reaching 39533 ± 6243 ind. m-2 in the new habitat. The specific ratios of both operculum area （Sa） to base area （SA） and average height of pa- rietal plates （H） to length of base （L） revealed that C. challengeri displays morphological changes to resist stronger currents in the new habitats for invasion.

Optimizational production of phenyllactic acid by a Lactobacillus buchneri strain via uniform design with overlay sampling methodology

A Lactobacillus buchneri GBS3 strain isolated from the traditional Chinese pickles was used for the production of 3-phenyllactic acid(PLA), an important compound with antimicrobial activities against a wide species of grampositive and gram-negative bacteria and some fungi. The growth performance of this strain in the de Man, Rogosa and Sharpe(MRS) medium, the production of metabolites of valuable organic acids, and the biosynthesis of PLA using this strain as the whole-cell biocatalyst and phenylpyruvic acid(PPA) as the precursor, were investigated experimentally. The uniform design method with overlay sampling was developed for the optimization of the biotransformation conditions. The results showed that although it produced naturally lactic acid with the maximum concentration of 1.84 g·L^(-1) and PLA with the concentration of 0.015 g·L^(-1) after 66 to 72 h cultivation in MRS broth by fermentation, the present strain displayed an effective utilization ability by transforming PPA to PLA. By the uniform design method with overlay sampling for the design and optimization of transformation conditions, a maximum yield of 10.93 g·L^(-1) PLA with the mole conversion ratio of 83.07% from PPA to PLA was achieved under the optimized condition, i.e., 20 g·L^(-1) glucose, 270 g·L^(-1) cells, 13 g·L^(-1) PPA, pH 8.0 and the reaction time of 15 h, indicating that Lactobacillus buchneri GBS3 was an interesting strain for the biosynthesis of PLA via the microbial transformation. The prediction of PLA yield under different conditions was achieved successfully based on the limited information of only a small number of experiments by the uniform design with overlay sampling. Therefore, the present methodology is effective and helpful for the optimization of the biosynthesis processes of PLA.

Benzimidazole derivatives have wide-spectrum biological activities and pharmacological effects,but remain challenging to be produced from biomass ***,we report a green hydrogen transfer strategy for the efficient one-pot production of benzimidazoles from a wide range of bio-alcohols and o-nitroanilines enabled by cobalt nitride species on hierarchically porous and recyclable nitrogen-doped carbon catalysts(Co/CN_(x)-T,T denotes the pyrolysis temperature)without using an external hydrogen source and base *** the tested catalysts,Co/CN_(x)-700 exhibited superior catalytic performance,furnishing 2-substituted benzimidazoles in 65%–92%*** mechanistic studies manifest that the coordination between Co^(2+)and N with appropriate electronic state on the porous nitrogen-doped carbon having structural defects,as well as the remarkable synergetic effect of Co/N dual sites contribute to the pronounced activity of Co/CN_(x)-700,while too high pyrolysis temperature may cause the breakage of the catalyst Co-N bond to lower down its ***,it is revealed that the initial dehydrogenation of bio-alcohol and the subsequent cyclodehydrogenation are closely correlated with the hydrogenation of nitro *** catalytic hydrogen transfer-coupling protocol opens a new avenue for the synthesis of N-heterocyclic compounds from biomass.
