Migration mechanism and risk assessment of perand polyfluoroalkyl substances in the Ya'Er Lake oxidation pond area,China

The migration mechanisms,sources,and environmental risks of 29 legacy and emerging perfluorinated and polyfluoroalkyl species present in an oxidation pond(Ya'Er Lake)were investigated for treating sewage based on the analysis of their occurrence and distribution.The concentration of per-and polyfluoroalkyl substances(PFAS)in pond area was between0.30 and 63.2 ng/g dw(dry weight),with the overall average concentration of 8.00 ng/g dw.Notably,the PFAS concentrations in the surface sediments near the sewage outlet in Pond-1(50.2 ng/g dw)and Pond-5(average 15.1 ng/g dw)were 1–2 orders of magnitude higher than those in other areas.In general,the legacy PFAS,i.e.,perfluorooctane sulfonic acid was considered to be the major pollutant in the polluted area,on average,accounting for 73.0%of the total concentration of PFAS pollutants.By evaluating the regional distribution of different PFAS homologs,the short-chain PFAS pollutants with lower K_(ow)were found to migrate farther in both horizontal and vertical directions.The sewage outlets in Pond-1 and Pond-5are the main pollution sources in polluted area and the emerging PFAS pollutants in Pond-5have replaced the legacy PFAS pollutants as the main pollutants.Based on positive matrix factorization analysis,three main industrial sources of PFAS pollutants in the study area were identified:protective coating,fire-fighting,and food packaging sources.Moreover,the environmental risk assessment results showed that most study areas exhibited medium environmental risk(0.01≤Risk quotient(RQ)<1),indicating that the ecological environment risks in this area need further attention.

Metabolic puzzle: Exploring liver fibrosis differences in Asian metabolic-associated fatty liver disease subtypes

BACKGROUND Metabolic-associated fatty liver disease(MAFLD)is a liver condition marked by excessive fat buildup in the absence of heavy alcohol use.It is primarily linked with metabolic issues like insulin resistance,obesity,and abnormal lipid levels,and is often observed with other conditions such as type 2 diabetes and cardiovascular disease.However,whether the subtypes of MAFLD based on the metabolic disorder differentially impact liver fibrosis is not well explicated,especially in the Asian population.AIM To compare the severity of liver fibrosis among different MAFLD subtypes.METHODS A total of 322 adult patients of either gender with fatty liver on ultrasound were enrolled between January to December 2021.MAFLD was defined as per the Asian Pacific Association for the Study of the Liver guidelines.Fibrosis-4 index(Fib-4)and nonalcoholic fatty liver disease fibrosis score(NFS)were employed to evaluate liver fibrosis.RESULTS The mean age was 44.84±11 years.Seventy-two percent of the patients were female.Two hundred and seventy-three patients were classified as having MAFLD,of which 110(40.3%)carried a single,129(47.3%)had two,and 34(12.5%)had all three metabolic conditions.The cumulative number of metabolic conditions was related to elevated body mass index,triglyceride(TG)levels,and glycated hemoglobin,lower high-density lipoprotein(HDL)levels,higher liver inflammation(by aspartate aminotransferase andγ-glutamyl transferase),and higher likelihood of fibrosis(by NFS and Fib-4 scores)(P<0.05 for all).The proportion of advanced fibrosis also increased with an increase in the number of metabolic conditions(4.1%,25.5%,35.6%,and 44.1%by NFS and 6.1%,10.9%,17%,and 26.5%by Fib-4 for no MAFLD and MAFLD with 1,2,and 3 conditions,respectively).Among MAFLD patients,those with diabetes alone were the eldest and had the highest mean value of NFS score and Fib-4 score(P<0.05),while MAFLD patients diagnosed with lean metabolic dysfunction exhibited the highest levels of TG and alanine aminotransferase but the lowest HDL level

Long non-coding RNAs with essential roles in neurodegenerative disorders

Recently, with the advent of high-resolution and high-throughput sequencing technologies, an increasing number of long non-coding RNAs(lncRNAs) have been found to be involved in the regulation of neuronal function in the central nervous system with specific spatiotemporal patterns, across different neurodegenerative diseases. However, the underlying mechanisms of lncRNAs during neurodegeneration remain poorly understood. This review provides an overview of the current knowledge of the biology of lncRNAs and focuses on introducing the latest identified roles, regulatory mechanisms, and research status of lncRNAs in Alzheimer’s disease, Parkinson’s disease, Huntington’s disease, and amyotrophic lateral sclerosis. Finally, this review discusses the potential values of lncRNAs as diagnostic biomarkers and therapeutic targets for neurodegenerative diseases, hoping to provide broader implications for developing effective treatments.

What is already known about this topic?Vitamin A deficiency(VAD)is a leading global nutritional concern,ranking among the top four major nutritional deficiencies worldwide.The prevalence of VAD is unevenly distributed across various regions,both within China and globally.What is added by this report?The report adds valuable insights into the vitamin A nutritional status of rural students aged 6–17 years who participated in the Nutrition Improvement Programme for Rural Compulsory Education Students(NIPRCES).Over the decade from 2012 to 2021,there was a modest improvement in vitamin A status.The prevalence of VAD and sub-clinical VAD(SVAD)declined as the students aged.Throughout the majority of the survey years,the incidence of VAD was higher among males and western regions compared to females and central regions,respectively.What are the implications for public health practice?A comprehensive approach,incorporating dietary diversification,nutrition education,and food fortification,should be implemented to prevent VAD and SVAD especially in males,younger children and children in western areas.

To investigate the circuit-level neural mechanisms of behavior, simultaneous imaging of neuronal activity in multiple cortical and subcortical regions is highly desired. Miniature head-mounted microscopes offer the capability of calcium imaging in freely behaving animals. However, implanting multiple microscopes on a mouse brain remains challenging due to space constraints and the cumbersome weight of the equipment.Here, we present TINIscope, a Tightly Integrated Neuronal Imaging microscope optimized for electronic and opto-mechanical design. With its compact and lightweight design of 0.43 g, TINIscope enables unprecedented simultaneous imaging of behavior-relevant activity in up to four brain regions in mice.Proof-of-concept experiments with TINIscope recorded over 1000 neurons in four hippocampal subregions and revealed concurrent activity patterns spanning across these regions. Moreover, we explored potential multi-modal experimental designs by integrating additional modules for optogenetics, electrical stimulation or local field potential recordings. Overall, TINIscope represents a timely and indispensable tool for studying the brain-wide interregional coordination that underlies unrestrained behaviors.

What is already known about this topic?Mushroom poisoning poses a significant food safety concern in China,with a total of 196 species identified in poisoning incidents by the end of 2022.What is added by this report?In 2023,the China CDC conducted an investigation into 505 cases of mushroom poisoning spanning 24 provincial-level administrative divisions.This investigation resulted in 1,303 patients and 16 deaths,yielding a case fatality rate of 1.23%.A total of 97 mushrooms were identified as the cause of 6 distinct clinical disease types,with 12 species newly documented as poisonous mushrooms in China.What are the implications for public health practice?Close collaboration among CDC staff,physicians,and mycologists remains crucial for the control and prevention of mushroom poisoning in the future.

Accuracy and precision of dried urine spot method for the detection of Schistosoma mansoni circulating cathodic antigens in resource-limited settings

Background The World health Organization recommends the use of Schisto point-of-care circulating cathodic anti-gens(Schisto POC-CCA)for screening of Schistosoma mansoni as it offers better sensitivity than microscopy.However,there are limitation facing the use of this method including timely availability of the test cassettes.The aim of this study was to determine the reliability of dried urine spot(DUS)method for collection of urine and detection of S.mansoni using Schisto POC-CCA cassettes in a resource-limited settings.Methods A cross-sectional study was conducted between October and November 2022 among 250 primary school children in Sengerema District,northwestern Tanzania.S.mansoni CCA was detected in filter paper-based DUSs,liquid urine using DUS Schisto POC-CCA(index),and direct urine Schisto POC-CCA(comparator)methods respectively.S.mansoni eggs in stool were detected using duplicate Kato-Katz(KK)method.The measures of accuracy were com-puted and compared between the index and comparator methods.The strength of agreement between inter-raters precisions was tested using Cohen's kappa(k).Results This study revealed S.mansoni prevalence rates of 28.8%,54.0%and 50.8%by duplicate KK,direct urine Schisto POC-CCA and DUS Schisto POC-CCA methods respectively.The mean intensity of infection among infected participants Was 86.3 eggs per gram of stool(EPG)ranging from 12.0 EPG to 824.0 EPG.The sensitivity of DUS Schisto POC-CCA and direct urine Schisto POC-CCA Was 94.44%(95%CI:89.15-99.74%)and 97.22%(95%CI:93.43-100.00%)respectively.The DUS Schisto POC-CCA method had slightly higher specificity(66.85%)than direct urine Schisto POC-CCA method(63.48%).The accuracy of the DUS Schisto POC-CCA Was found to be slightly high(74.80%,95%CI:68.94-79.06%)compared to that of direct urine Schisto POC-CCA(73.20%,95%CI:67.25-78.59%).There was good agreement between two laboratory technologists who performed the DUS Schisto POC-CCA method on similar samples(k=0.80,95%CI:0.59-0.95).Conclusions The DUS Schisto POC-CCA method had comp

Anemia,a global public health problem,has significant adverse consequences on the cognitive development of children and the work capacity of adults;affecting social and economic development.Globally,roughly 43%of children under five years of age,38%of pregnant women,and 29%of nonpregnant women had anemia.Anemia during pregnancy significantly increases the risk of low birth weight and preterm birth.A 10 g/L increase in hemoglobin has been estimated to decrease the risk of maternal and perinatal mortality by 29%and 28%.

For sessile plants, gene expression plays a pivotal role in responding to salinity stress by activating or suppressing specific genes. However, our knowledge of genetic variations governing gene expression in response to salt stress remains limited in natural germplasm. Through transcriptome analysis of the Global Mini-Core Rice Collection consisting of a panel of 202 accessions, we identified 22 345 and 27 610expression quantitative trait loci associated with the expression of 7787 and 9361 e Genes under normal and salt-stress conditions, respectively, leveraging the super pan-genome map. Notably, combined with genome-wide association studies, we swiftly pinpointed the potential candidate gene STG5—a major salt-tolerant locus known as q STS5. Intriguingly, STG5 is required for maintaining Na+/K+homeostasis by directly regulating the transcription of multiple members of the Os HKT gene family. Our study sheds light on how genetic variants influence the dynamic changes in gene expression responding to salinity stress and provides a valuable resource for the mining of salt-tolerant genes in the future.
