Mungbean （Vigna radiata （L.） Wilczek） is a unique species in its ability to fix atmospheric nitrogen, with early maturity, and relatively good drought resistance. We used 454 sequencing technology for transcriptome sequencing. A total of 150 159 and 142 993 reads produced 5 254 and 6 374 large contigs （〉_500 bp） with an average length of 833 and 853 for Sunhwa and Jangan, respectively. Functional annotation to known sequences yielded 41.34% and 41.74% unigenes for Jangan and Sunhwa. A higher number of simple sequence repeat （SSR） motifs was identified in Jangan （1 630） compared with that of Sunhwa （1 334）. A similar SSR distribution pattern was observed in both varieties. A total of 8 249 single nucleotide polymorphisms （SNPs） and indels with 2 098 high-confidence candidates were identified in the two mungbean varieties. The average distance between individual SNPs was -860 bp. Our report demonstrates the utility of transcriptomic data for implementing a functional annotation and development of genetic markers. We also provide large resource sequence data for mungbean improvement programs.

So far,ascoviruses have only been identified from Lepidoptera host insects and their transmission vectors-endoparasitic ***,we reported the first finding of a complete novel ascovirus genome from a Diptera insect,Dasineura ***,sequence fragments with homology to ascoviruses were incidentally identified during metagenomic sequencing of the mitochondria of ***(Cecidomyiidae,Diptera)which is a major pest on Ziziphus *** a full circular viral genome was assembled from the metagenomic data,which has an A+T percentage of 74%and contains 142,600 bp with 141 open reading frames(ORFs).Among the 141 ORFs,37 were conserved in all sequenced ascoviruses(core genes)including proteins predicted to participate in DNA replication,gene transcription,protein modification,virus assembly,lipid metabolism and ***-gene families including those encode for baculovirus repeated open reading frames(BROs),myristylated membrane proteins,RING/U-box E3 ubiquitin ligases,and ATP-binding cassette(ABC)transporters were found in the virus *** analysis showed that the newly identified virus belongs to genus Toursvirus of Ascoviridae,and is therefore named as Dasineura jujubifolia toursvirus 2(DjTV-2a).The virus becomes the second reported species of the genus after Diadromus pulchellus toursvirus 1(DpTV-1a).The genome arrangement of DjTV-2a is quite different from that of DpTV-1a,suggesting these two viruses separated in an early time of *** results suggest that the ascoviruses may infect a much broader range of hosts than our previous knowledge,and shed lights on the evolution of ascoviruses and particularly on that of the toursviruses.

Molecular Cloning and Characterization of a Novel Gene Involved in Fatty Acid Synthesis in Brassica napus L.

Based on the sequence of a novel expressed sequence tag （EST）, the full-length cDNA of 1 017 nucleotides was cloned from Brassica napus cv. Xiangyou 15 through rapid amplification of cDNA ends （RACE）. The gene was designated as Bnhol34 （HQ585980）, encoding a protein of 338 amino acids. BLAST analysis showed no high degree of sequence identity to any known gene. The calculated molecular weight of the Bnhol34 protein was 36.23 kDa, and the theoretical isoelectric point was 8.74. The Bnhol34 was also cloned from a high oleic acid mutant 854-1 through homologous cloning. There was no difference between the two Bnhol34 genes. Bnhol34 was localized in a tissue-specific manner in B. napus, and its expression level was about eight-fold greater in Xiangyou 15 seeds than in 854-1. The promoter region sequences of Bnhol34 were then isolated from Xiangyou 15 and 854-1, and a 93-bp deletion was found to occur in the Bnhol34 promoter region of 854-1. Three abscisic acid-responsive cis-elements （ABRE） were identified in the promoter region of Xiangyou 15. Real-time PCR analyses revealed that exogenous abscisic acid increased Bnhol34 expression by about four-fold in Xiangyou 15 seeds, yet did not change Bnhol34 expression in 854-1. It appeared that Bnhol34 might be abscisic acid insensitive in 854-1.

Cultivated land is one of the most important types of land cover in the global mapping of land cover, and its variation influences economic development, food security, and ecological environment protection. Existing products of global cultivated land mapping have a low resolution, and high spatial resolution products are in demand. This study uses global remote sensing image datasets in 2000/2010 with a spatial resolution of 30 m(Landsat TM/ETM+, HJ-1), MODIS 250 m NDVI time-serial data, and many types of reference data. An three-layer extraction method based on pixels, objects, and knowledge(POK) was adopted to ease cultivated land extraction in global-scale 30 m images, i.e., cultivated land classification based on pixel-scale multi-feature optimization, cultivated land automatic identification based on objects, and interactive object processing based on information service and priori knowledge. Global 30 m cultivated land mapping was accomplished for the two reference years(2000 and 2010), and statistical analysis was conducted on the data. Results showed that the total cultivated land area was 1.903 billion ha and 1.960 billion ha, respectively. Accuracy assessments showed that overall accuracy of global cultivated land mapping are higher than 92% for both the two reference years. The global cultivated land products in 2000/2010 developed in this research are superior to their international counterparts in terms of spatial resolution and classification accuracy. They also provide significant basic data on global food security, ecological environment supervision, and global change.

An estimation of nutrient fluxes to the East China Sea continental shelf from the Taiwan Strait and Kuroshio subsurface waters in summer

According to historical mean ocean current data through the field observations of the Taiwan Ocean Research Institute during 1991–2005 and survey data of nutrients on the continental shelf of the East China Sea（ECS） in the summer of 2006, nutrient fluxes from the Taiwan Strait and Kuroshio subsurface waters are estimated using a grid interpolation method, which both are the sources of the Taiwan Warm Current. The nutrient fluxes of the two water masses are also compared. The results show that phosphate（PO4-P）, silicate（SiO3-Si） and nitrate（NO3-N） fluxes to the ECS continental shelf from the Kuroshio upwelling water are slightly higher than those from the Taiwan Strait water in the summer of 2006. In contrast, owing to its lower velocity, the nutrient flux density（i.e., nutrient fluxes divided by the area of the specific section） of the Kuroshio subsurface water is lower than that of the Taiwan Strait water. In addition, the Taiwan Warm Current deep water, which is mainly constituted by the Kuroshio subsurface water, might directly reach the areas of high-frequency harmful alga blooms in the ECS.

Vascular development is essential for the establishment of the circulatory system during embryonic development and requires the proliferation of endothelial ***,the underpinning regulatory mechanisms are not well ***,we report that geranylgeranyl pyrophosphate(GGPP),a metabolite involved in protein geranylgeranylation,plays an indispensable role in embryonic vascular *** is synthesized by geranylgeranyl pyrophosphate synthase(GGPPS)in the mevalonate *** selective knockout of Ggpps in endothelial cells led to aberrant vascular development and embryonic lethality,resulting from the decreased proliferation and enhanced apoptosis of endothelial cells during *** defect in protein geranylgeranylation induced by GGPP depletion inhibited the membrane localization of Rho A and enhanced yes-associated protein(YAP)phosphorylation,thereby prohibiting the entry of YAP into the nucleus and the expression of YAP target genes related to cell proliferation and the antiapoptosis ***,inhibition of the mevalonate pathway by simvastatin induced endothelial cell proliferation defects and apoptosis,which were ameliorated by ***(GGOH),a precursor of GGPP,ameliorated the harmful effects of simvastatin on vascular development of developing fetuses in pregnant *** results indicate that GGPP-mediated protein geranylgeranylation is essential for endothelial cell proliferation and the antiapoptosis process during embryonic vascular development.
