Apocytochrome c (Apocyt. c) is the precursor of cytochrome c. It is synthesized in the cytosol and posttranslationally imported into mitochondria. In order to determine the crucial se-quence in apocyt. c translocation, deleted mutant and chemically synthesized peptides with differ-ent length were used. Obtained results showed that sequence 68-88 of apocyt. c plays a critical role in its insertion into membrane and binding to mitochondria.

The structural information of functionally important mac- romolecular assemblies is a key to molecular biology and is of great interest to structural biologists. Cryo-electron mi- croscopy （cryo-EM）, particularly the single particle analysis （SPA）, has been regarded as one of the three primary tech- niques for structure determination, together with X-ray crystallography and nuclear magnetic resonance （NMR）. For single particle cryo-EM, a tiny amount （~3 μL） of puri- fied biological samples at a concentration around 0.1-2 mg mL-1 are placed onto an EM grid, blotted with a filter paper and rapidly frozen into a thin layer of vitreous ice. The EM grid with frozen samples is then transferred to a transmis- sion electron microscope for imaging.

The unfolding of bovine serum albumin, lysozyme and ribonuclease A denatured in 6 mol/L GuHCl with their disulfide bridges intact and reduced have been compared by FTIR studies. The peak positions and heights in the deconvolved spectra of amide I bands of the above denatured proteins with native disulfide bonds show marked differences whereas those for the denatured proteins without disulfide linkages are closely similar. The above and other evidence suggest that denatured proteins with intact disulfides still have considerable ordered conformation even in 6 mol/L GuHCl.

For a long time phenolic additions have been widely used for the preservation of insulin preparation, but its action mechanism has not been elucidated. In 1958 Schlichkrull first obtained monoclinic insulin crystals in a system with 1.0% phenol which differs remarkably from those crystal growing systems of rhombohedral 2Zn and 4Zn insulin. In contrast to the horse heart apocytochrome c,the chicken heart apocytochrome c under-went a conformational change from random coil to partial folding during a renaturation *** theapocytochrome horse heart and that of chicken heart c were subjected to a translocation assay in vitro usinglarge trypsin-enclosed unilamellar vesicles from soybean phospholipids,the ability of the chicken heart apoc-ytochrome c to penetrate into the liposomes was found to decrease markedly with the renaturation procedure,while that of horse heart apocytochrome c remained relatively *** from circular dichroismmeasurement on the induction of secondary folding of these two species of apocytochrome c upon interactionwith soybean phospholipid vesicles suggested that a more flexible structure of apocytochrome c embedded inthe lipid matrix be required for its efficient translocation across the bilayer.
