The switch from methadone to buprenorphine-naloxone for individuals with heroin dependence is associated with several obstacles and *** patients may experience discomfort from discontinuing methadone, precipitated withdrawal symptoms induced by buprenorphine-naloxone,and poor psychosocial adjustments such as anticipatory anxiety regarding severe opioid *** herein describe a 46-year-old man with a history of heroin dependence who underwent traditional chinese medicine(TCM)-facilitated switching from methadone to buprenorphine-naloxone. No precipitated withdrawal was induced by buprenorphine-naloxone. The drug-switching process was successful and smooth. He maintained abstinence from heroin for the following year. In this case, we applied TCM for enhancement of methadone metabolism and detoxification, analgesic effects, and anxiolytic and hypnotic effects during the drug switch. We observed that TCM effectively facilitated the switch from methadone to buprenorphine-naloxone in our case. Further studies regarding TCM-facilitated treatment for heroin dependence should be conducted.

Objective： To approach a better solution for enhancing the therapeutic results of acupuncture therapy in the treatment of Tourette syndrome, by observing the clinical results of combined scalp with body acupuncture and mono-body acupuncture. Methods： Fifty-seven patients were randomized into a treatment group （31 cases） and a control group （26 cases）. The patients in the treatment group all received combined scalp-body acupuncture treatment, while the patients in the control group were given mono-body acupuncture treatment, for I month as a treatment session. At the end of the third treatment session, the Yale Global Tic Severity Scale （YGTSS） would be compared between pre- and post-treatment. Results： In the treatment group, 2 patients were clinically cured, 4 showed markedly effective, 18 showed effective, and 7 failed, making a total therapeutic rate of 77.4%. In the control group, 0 were clinically cured, 3 showed markedly effective, 9 showed effective, 14 failed, making a total therapeutic rate of 46.2%. There was a significant difference between the two total therapeutic rates （P〈0.05）. Conclusion： The combination of scalp and body acupuncture had a better therapeutic result than the mono-body acupuncture therapy in the treatment of Tourette syndrome.

Additive Effect of Zfhx3/Atbf1 and Pten Deletion on Mouse Prostatic Tumorigenesis

The phosphatase and tensin homolog （PTEN） and the zinc finger homeobox 3 （ZFHX3）/AT-motif binding factor 1 （ATBF1） genes have been established as tumor suppressor genes in prostate cancer by their frequent deletions and mutations in human prostate cancer and by the formation of mouse prostatic intraepithelial neoplasia （mPIN） or tumor by their deletions in mouse prostates. However, whether ZFHX3/ATBF1 deletion together with PTEN deletion facilitates prostatic tumorigenesis is unknown. In this study, we simultaneously deleted both genes in mouse prostatic epithelia and performed histological and molecular analyses. While deletion of one Pten allele alone caused low-grade （LG） mPIN as previously reported, concurrent deletion of Zfhx3/Atbfl promoted the progression to high-grade （HG） mPIN or early carcinoma. ZJhx3/Atbfl and Pten deletions together increased cell proliferation, disrupted the smooth muscle layer between epithelium and stroma, and increased the number of apoptotic cells. Deletion of both genes also accelerated the activation of Akt and Erkl/2 oncoproteins. These results suggest an additive effect of ZFHX3/ATBF1 and PTEN deletions on the development and progression of prostate neoplasia.

Gypenosides,structurally analogous to ginsenosides and derived from a sustainable source,are recognized as the principal active compounds found in Gynostemma pentaphyllum,a chinese medicinal plant used in the treatment of the metabolic *** bioactive tracking isolation of the plants collected from different regions across China,we obtained four new gypenosides(1−4),together with nine known gypenosides(5−13),from the methanol extract of the *** structures of new gypenosides were elucidated by one-dimensional(1D)and two-dimensional(2D)nuclear magnetic resonance(NMR)spectra,complemented by chemical degradation *** comprehensive evaluation involving COL1A1 promoter assays and PP2Cαactivity assays,we established a definitive structure-activity relationship for these dammarane-type triterpenoids,affirming the indispensability of the C-3 saccharide chain and C-17 lactone ring in effectively impeding extracellular matrix(ECM)deposition within hepatic stellate *** in vivo study on the CCl4-induced liver damage mouse model corroborated that compound 5 significantly ameliorated the process of hepatic fibrosis by oral *** results underscore the potential of dammarane-type triterpenoids as prospective antifibrotic leads and highlight their prevalence as key molecular frameworks in the therapeutic intervention of chronic hepatic disorders.

Ischemic stroke(IS)is the most prevalent form of brain disease,characterized by high morbidity,disability,and ***,there is still a lack of ideal prevention and treatment measures in clinical ***,the trans-plantation therapy of mesenchymal stem cells(MSCs)has been a hot research topic in ***,there are risks associated with this cell therapy,including tumor formation,coagulation dysfunction,and vascular ***,a growing number of studies suggest that the therapeutic effect after transplantation of MSCs is mainly attributed to MSC-derived exosomes(MSC-Exos).And this cell-free mediated therapy appears to circumvent many risks and difficulties when compared to cell therapy,and it may be the most promising new strategy for treating stroke as stem cell replacement *** suggest that suppressing inflammation via modulation of the immune response is an additional treatment option for ***,MSC-Exos mediates the inflam-matory immune response following IS by modulating the central nervous system,the peripheral immune system,and immunomodulatory molecules,thereby promoting neurofunctional recovery after ***,this paper reviews the role,potential mechanisms,and therapeutic potential of MSC-Exos in post-IS inflammation in order to identify new research targets.

Experimental Research of Electroacupuncture on ERK Phosphorylation in Rats of Hepatofibrosis

Objective： To observe the intervening effect of electroacupuncmre at Ganshu （BL 18） and Zusanli （ST 36） on ERK phosphorylation in rats of hepatofibrosis, and to explore the possible molecular mechanisms of electroacupunctre for hepatofibrosis. Methods： Forty male Wistar rats were randomly divided into normal group, model group, electroacupuncture group, and medicine group. Rats in the latter three groups were hypodermically injected with the mixture of olive oil and 30% carbon tetrachloride and drank tap water containing 10% alcohol to induce hepatic fibrosis model. Rats in the electroacupuncture group were stimulated at Zusanli （ST 36） and Ganshu （BL 18） by electroacupuncture, and rats in the medicine group were treated with Colchicine. The collagen hyperplasia of rats＇ hepatic tissue was observed by Sirius Red staining, and protein expression of ERK and phosphorylated ERK （P-ERK） were observed by Western Blot. Results： Compared with those in the normal group, collagen deposition in hepatic tissue obviously increased in the model group, and P-ERK protein expression was also up-regulated obviously （P〈0.01）. Compared with those in the model group, collagen deposition in hepatic tissue decreased in the electroacupuncmre and medicine groups, and P-ERK protein expressed was down-regulated （P〈0.01）. The difference between the electroacupuncture and medicine groups had no significance. Conclusion： Electroacupuncture can prevent or delay the development of hepatic fibrosis in rats by down-regulating the protein expression of P-ERK to block MAPK/ERK signal pathway and to suppress the synthesis of extracellular matrix.

Objective:The aim of this study was to investigate the protective effects of ginsenoside Rb1 and assess whether these protective effects are related to calcium/calmodulin-dependent protein kinaseⅡ(Ca MKⅡ).Methods:A myocardial ischemia(IS)*** and a myocardial H9 C2 cell hypoxia model were *** was induced by occluding the left anterior descending artery for 120 *** Rb1(10 mg/kg)was administered 30 min before ischemia induction,and the treatment continued for 7 ***:In the rat IS injury model,ginsenoside Rb1 reduced myocardial infarct size,mean left ventricular diastolic pressure,incidence of arrhythmia,and levels of serum creatine kinase,lactate dehydrogenase,and ***,the mean left ventricular systolic pressure,and maximal rising and falling rates of ventricular pressure(±dp/dtmax)*** the myocardial H9 C2 cell hypoxia model,ginsenoside Rb1 reduced intracellular calcium concentrations([Ca2+]i)during hypoxia,and markedly reversed the hypoxia-induced decrease in cell *** Rb1 was involved in the downregulation of CaMKⅡand the ryanodine receptor,as well as hypoxia-induced H9 C2 cell ***:The findings of the present study suggest that ginsenoside Rb1 attenuates MI injury in rats,partially through the downregulation of CaMKⅡexpression.

According to folk usage of Aconitum carmichaelii Debx., the present study was designed to determine the feasibility of the stems and leaves of Aconitum carmichaelii Debx. as a new medicinal resource. Fourteen alkaloids in mother roots, fibrous roots, stems, and leaves of Aconitum carmichaelii Debx. were measured by HPLC-MS/MS. And multivariate analysis methods, such as clustering analysis and principal component analysis, were applied to analyze the difference among various parts. In addition, the acute toxicity, analgesia, and anti-inflammatory tests were carried out. The results suggested that the contents of alkaloids in mother roots and fibrous roots were approximate, but those of leaves and stems were different from mother roots and fibrous roots. The results of the acute toxicity testing demonstrated the toxicity of fibrous root was strongest, and mother roots were slightly less toxic than fibrous roots. The stems and leaves were far less toxic than mother and fibrous roots. In addition, the analgesia and inflammatory tests showed the effects of the various tissues had no difference each other. These results provided a basis for developing new complementary and alternative treatments for rheumatoid arthritis patients. Simultaneously, the approach may also turn wastes into treasure and promote the development of circular economy.

OBJECTIVE: To evaluate the clinical effectiveness and safety of acupoint catgut embedding and acupuncture on simple obesity by Meta-analysis. METHODS: Studies on clinical randomized controlled trials of acupoint catgut embedding for simple obesity which were published from January 2015 to November 2020 were searched in Cochrane Central Register of Control Trials(Central), Pub Med, China Science and Technology Journal Database, China National Knowledge Infrastructure Database and Wanfang databases. And those that met the inclusion criteria were screened. Rev Man5.3 was used for Meta-analysis. The “Risk of Bias” tool was used to evaluate the quality of included studies. R studio software was used for the measurement of publication bias. RESULTS: A total of 33 studies were included for Metaanalysis, including 2685 patients with simple obesity. Meta-analysis results showed the comparison of effectiveness rate was relative risk(RR) = 1.12, 95%CI(1.08, 1.16), body mass index(BMI) was mean difference(MD) =-1.12, 95%CI(-2.09,-0.14), waist circumference was MD =-2.14, 95%CI(-4.22,-0.06), and body mass was MD =-2.36, 95%CI(-3.99,-0.73). On the basis of diet and exercise intervention, the effectiveness rate [RR = 1.12, 95%CI(1.05, 1.19)], BMI [MD =-0.88, 95%CI(-1.35,-0.40)], waist circumference [MD =-1.10, 95%CI(-4.27, 2.07)], and body mass [MD =-0.68, 95%CI(-2.90, 1.54)]. The risk of bias of included literatures was low. CONCLUSIONS: Acupoint catgut embedding therapy was slightly better than acupuncture therapy in most of the outcomes. Moreover, the treatment frequency of acupoint catgut embedding is less with larger stimulation intensity, which is more conducive to clinical promotion.
