Background:Insomnia is a prevalent clinical condition and Shangxia Liangji formula(SXLJF)is a well-established method of ***,the specific mechanism of action of SXLJF remains ***:The mouse model of insomnia was established by intraperitoneal injection of ***-two mice were randomly divided into a negative control group,model group,SXLJF group(18.72 g/kg/day),and positive control group(diazepam,2 mg/kg)and treated with the corresponding drugs for 7 consecutive *** open field test and pentobarbital-induced sleeping test were ***-MS-based untargeted metabolomics and network pharmacology were applied to explore the potential targets of SXLJF for treating ***,key targets were validated using ***:Behavioral tests demonstrated that SXLJF reduced the total distance,average velocity,central distance,and sleep latency,and prolonged sleep *** and network pharmacology revealed potential targets,signaling pathways,metabolic pathways,and metabolites associated with the anti-insomnia effects of ***,tyrosine hydroxylase(TH)and tyrosine metabolism emerged as crucial metabolic pathways and targets,***-qPCR results supported the role of TH in the mechanism of SXLJF in treating ***:In conclusion,TH and tyrosine metabolism may represent significant targets and pathways for SXLJF in treating insomnia.

Identification of Peptides Inhibiting Adhesion of Monocytes to the Injured Vascular Endothelial Cells through Phage-displaying Screening

Onychin inhibits proliferation of vascular smooth muscle cells by regulating cell cycle

Aim:To investigate the effects of onychin on the proliferation of cultured ratartery vascular smooth muscle cells(VSMCs)in the presence of 10% new-borncalf serum(NCS).Methods:Rat VSMCs were incubated with onychin 1-50μmol/L or genistein 10μmol/L in the presence of 10% NCS for 24 *** prolif-eration of VSMCs was measured by cell counting and MTS/PMS *** cycle progression was evaluated by flow ***(Rb)phosphorylation,and expression of cyclin D1and cyclin E were measured byWestern blot *** tyrosine phosphorylation of ERK1/2 was examined byimmunoprecipitation techniques using anti-phospho-tyrosine ***:The proliferation of VSMCs was accelerated significantly in the presence of 10%*** reduced the metabolic rate of MTS and the cell number of VSMCsin the presence of 10% NCS in a dose-dependent *** cytometry analy-sis revealed that the G1-phase fraction ratio in the onychin group was higher thanthat in the 10% NCS group(85.2% vs 70.0%,P<0.01),while the S-phase fractionratio in the onychin group was lower than that in 10% NCS group(4.3% vs16.4%,P<0.01).Western blot analysis showed that onychin inhibited Rb phos-phorylation and reduced the expression of cyclin D1and cyclin *** effects ofonychin on proliferation,the cell cycle and the expression of cyclins in VSMCswere similar to those of genistein,an inhibitor of tyrosine *** studies showed that both onychin and genistein markedlyinhibited the tyrosine phosphorylation of ERK1/2 induced by 10% ***:Onychin inhibits the proliferation of VSMCs through G1phase cellcycle arrest by decreasing the tyrosine phosphorylation of ERK1/2,and the ex-pression of cyclin D1and cyclin E,and sequentially inhibiting Rb phosphorylation.

Silencing of Bcl-XL Expression in Human MGC-803 Gastric Cancer Cells by siRNA

To investigate the inhibitory effect of the Bcl-XL small interfering RNA(siRNA)on BcI-XLgene expression in the human gastric cancer cell line MGC-803,green fluorescent protein(GFP)siRNAwas constructed and transfected into MGC-803 ceils,together with GFP expression vector pTrace *** expression levels were observed using fluorescence ***-XL siRNA and negative siRNAwere then constructed and stably transfected into MGC-803 ***-PCR and immunofluorescence wereused to detect the expression of *** apoptosis was detected by acridine orange(AO)andflow *** were as follows:(1)48 h after GFP expression vector and GFP siRNA co-transfection,the expression level of GFP in the GFP siRNA group was much lower than the negative siRNA group,according to fluorescence microscopy *** mRNA and protein levels of Bcl-XL in Bcl-XL siRNAstable transfectants were reduced to almost background level compared with negative siRNA transfectantsor untreated cells.(2)Changes in nucleus morphology was observed by AO staining nucleic and flowcytometry analysis,which showed that stable Bcl-XL siRNA transfectants have an increased spontaneousapoptosis (21.17%+1.26% vs.1.19%+0.18% and 1.56%+0.15% respectively,P<0.05 *** siRNAor untreated control),siRNA targeting GFP or Bcl-XL genes can specifically suppress GFP or BcI-XLexpression in MGC-803 cells,and Bcl-XL siRNA can increase spontaneous ***-XL siRNA maybe a beneficial agent against human gastric adenocarcinoma.

Bilateral Breast Cancer with a Unilateral Lipid-rich Carcinoma:A Case Report

Case ReportA 38-year old female patient was hospitalized on the 10th of December, 2006 as she had noted a lump in her right breast for 3 months. Her past medi. cal history indicated that, because of a left-breast carcinoma complex, she had undergone a radical operation for a left-sided mastocarcinoma on the 20th, October, 2002, in Xiangya hospital, Chasha. She received 5-course postoperative chemotherapy （i.e. CAF regimen）, and one course of radiotherpy on the left supraclavicular and left parasternal region. There was no medical record of breast cancer in her familv.

Relation Between Cellular Senescence and Liver Diseases

Cellular senescence refers to a process that cellular proliferation and differentiation modulated by the multiple stimulating factors gradually decline. Aging cells present the irreversible stop of proliferation and differentiation and change in secretory function because the cell cycle of aging cells is steadily blocked at some point. It has have been shown that cellular senescence plays an important role in the occurrence and development of liver diseases. In this paper, we review the advances in relations between cellular senescence and liver diseases.

Investigation of transcriptional gene silencing and mechanism induced by shRNAs targeted to RUNX3 in vitro

AIM:To investigate transcriptional gene silencing induced by short hairpin RNAs (shRNAs) that target gene prompter regions of RUNX3 gene, and whether shRNAs homologous to DNA sequences may serve as initiators for methylation. METHODS:According to the principle of RNAi design, pSilencer3.1-H1-shRNA/RUNX3 expression vector was constructed, The recombinant plasmid shRNA was transfected into human stomach carcinoma cell line SGC7901 with Lipofectamine 2000. Then, the positive cell clones were screened by G418. The mRNA and protein expression level of RUNX3 in the stable transfected cell line SGC7901 were determined by RT-PCR, Western blotting and immunocytochemistry. Characteristics of the cell lines including SGC7901, pSilencer3.1-H1/SGC7901 and pSilencer3.1-H1-shRNA/RUNX3/SGC7901 were analyzed with growth curves, clone formation rate and cell-cycle distribution. The activated level of RUNX3 was examined after treatment with the different density of 5’-aza-2’-deoxycytidine (5-Aza-CdR) by using semi- quantitative RT-PCR and Western blotting. RESULTS:In the cell line SGC7901 transfected with pSilencer3.1-H1-shRNA/RUNX3, mRNA and proteinexpression of the RUNX3 gene was lost identified by RT- PCR, Western blotting and immunocytochemistry assay. The growth of pSilencer3.1-H1-shRNA/ RUNX3/SGC7901 cells without expression of RUNX3 was the fastest (P < 0.05), its rate of clone formation was the highest (P < 0.01), and the cell distribution in G0/G1 and S/M phases was lowest and highest, respectively (P < 0.05), compared with that of the transfected pSilencer3.1-H1 and non-transfected cells. Through RT-PCR and Western blot assay, inactivated RUNX3 could not be reactivated by 5-Aza-CdR. CONCLUSION:We found that, although shRNAs targeted to gene prompter regions of RUNX3 could effectively induce transcriptional repression with chromatic changes characteristic of inaction promoters, this was independent of DNA methylation, and the presence of RNA-dependent transcriptional silencin

Progress of Targeting Transforming Growth Factor-β1 Small Interfering RNA in Liver Fibrosis

Liver fibrosis is a common pathological consequence of a variety of chronic stimuli, including viral, autoimmune, drug-induced, cholestatic and metabolic diseases. Fibrosis is driven by a dynamic process involving increased synthesis of matrix components and a failure of physiological mechanisms of matrix turnover. Activation of hepatic stellate cells(HSCs) remains a central event in fibrosis. HSCs are the main source of extracellular matrix(ECM). Transforming growth factor-beta(TGF-β), which is the fibrogenic master cytokine, can induce the activation of HSCs to produce a large amount of ECM, and is capable of inducing apoptosis of liver cells. RNA interference(RNAi) is a novel gene disruption technology. Studies have shown that small interfering RNA(si RNA) targeting TGF-β1 may inhibit the activation and proliferation of HSCs, suppress ECM synthesis and block liver fibrosis. TGF-β1 si RNA-mediated gene silencing therapy provides a new avenue for liver fibrosis. This review summarizes recent progresses in research on HSCs, TGF-β1 and TGF-β1 si RNA in liver fibrosis.
