Effect of Qilongtoutong granule on calcitonin gene-related peptide,beta-endorphin, serotonin, dopamine, and noradrenalin in migraine model rats and mice

OBJECTIVE： To study the effect of Qilongtoutong granule （QLTT） on plasma calcitonin gene-related peptide （CGRP）, beta-endorphin （I[3-EP）, 5-HT, dopa- mine （DA）, noradrenalin （NE）, and blood viscosity in migraine model rats and mice. METHODS： Both the acute blood stasis model group and nitroglycerin-induced migraine model group included 60 Sprague-Dawley rats. The reser- pine-reduced model group had 60 Kunming mice. Rats from each test were grouped into normal con- trol group, model group, Zhengtian pill （ZTP） group, and high, moderate, or low-dose QLTT groups. In the acute blood stasis model test, after gavage for 7 days, rats were given 0.8 mL/kg adren- aline hydrochloride subcutaneously twice, and kept in ice water for 5 min. After fasting for 12 h, rats were anesthetized and blood samples were collected for detection of blood viscosity. In the nitro- glycerin-induced migraine group, after gavage for 7 days, rats were intraperitoneally injected nitro- glycerin （10 mg/kg）, and 4 h later, blood samples were collected from postcava for measuring the plasma CGRP and 13-EP levels. In the reserpine-re- duced model test, except the normal control group, mice were administered reserpine （0.25 mg/ kg, i.h.） for 9 days. Mice received intragastric admin- istration from the third day to the ninth day. One hour after the last gavage, blood and brain tissue samples were obtained. Then, blood clotting time and the contents of neurotransmitters were deter- mined. RESULTS： QLTT- （3.6, 1.8, and 0.9 g/kg） and ZTP-treated rats had lower blood viscosity than that in model rats under different shear rates （P〈 0.01）. QLTT- （3.6, 1.8 g/kg） and ZTP-treated rats had significantly lower plasma CGRP levels and higher plasma 13-EP levels than those in model rats （P〈 0.01）. QLTT treatment at dose of 0.9 g/kg had lower plasma CGRP levels as well （P〈0.05）. QLTT- （5.2, 2.6 g/kg） and ZTP-treated mice had longer blood clotting time than that in model mice （P〈0.01）. Tendon ruptures remain a significant musculoskeletal injury. Despite advances in surgical techniques and procedures, traditional repair techniques maintain a high incidence of rerupture or tendon elongation. Mechanical loading and biochemical signaling both control tissue healing. This has led some researchers to consider using a technique based on tension regulation at the suture line for obtaining good healing. However, it is unknown how they interact and to what extent mechanics control biochemistry. This review will open the way for understanding the interplay between mechanical loading and the process of tendon healing.

Genetic and epigenetic predispositions underlying cardiovascular outcomes among patients treated with clopidogrel and aspirin

Coronary artery disease(CAD) is a major cause of death and disability worldwide,and consumes a considerable amount of medical resources every *** is a first-line *** therapy for CHD,butit is associated with substantial variability in PK and pharmacodynamics *** date,gene variants explain only a smallproportion of the *** study aimed to identify new genetic loci-modifying antiplatelet response to clopidogrel in Chinese patients with CAD by a systematic analysis combining antiplatelet effects and PK,and further to investigate the PON1 gene promoter DNA methylation and genetic variations possibly influencing clinical outcomes in *** undergoing *** identified novel variants in two transporter genes(SLC14 A2 rs12456693,ATP-binding cassette [ABC]A1 rs2487032) and in N6 AMT1(rs2254638) associated with P2Y12 reaction unit(PRU) and plasma active metabolite(H4) *** new variants dramatically *** the predictability of PRU variability to 37.7%.The associations between these loci and PK *** of clopidogrel and H4 were observed in additional patients,and its function on the activation of clopidogrel was validated in liver S9 fractions(P<0.05).Rs2254638 was further identified to exert a marginal risk effect formajor adverse cardiac events in an independent *** logistic regression analysis indicated that PON1 methylation level at CpG site-161(OR=0.95;95% CI=0.92–0.98;P<0.01) and the use of angiotensin converting enzyme inhibitors(OR=0.48;95% CI=0.26–0.89;P<0.01) were associated with decreased risk of bleeding *** conclusion,new genetic variants were systematically identified as risk factors for the reduced efficacy of clopidogrel *** *** expression of DNA methylation-regulating key genes in the pharmacokinetic and *** pathways of clopidogrel and aspirin may modify clinical outcomes in dual antiplatelet-treated patients undergoing PCI.

Structural-visual functional relationships detected by optical coherence tomography in varying age-cohorts' patients with optic neuritis

AIM: To assess the relationships of final best-corrected visual acuity(BCVA) and the optic nerve structural loss in varying age-cohorts of optic neuritis(ON) ***: This is a retrospective, cross-sectional *** 130 ON subjects(200 eyes) without ON onset within 6mo were included, who underwent BCVA assessment,peripapillary retinal nerve fibre layer(pRNFL) and macular segmented layers evaluation by optical coherence tomography(OCT).RESULTS: For the 0-18y cohort, the final BCVA(logMAR)was significantly better and less frequent recurrences than adult cohorts(P=0.000). The final BCVA(logMAR) in all age-cohorts of the ON patients had negative and linear correlations to the pRNFL thicknesses and macular retinal ganglion cell layer(mRGCL) volumes, when the pRNFL thicknesses were reduced to the thresholds of 57.2-67.5 μm or 0.691-0.737 mm;in mRGCL volumes, respectively, with the strongest interdependence in the 19-40y cohort. The ON patients from varying age cohorts would be threatened by blindness when their pRNFL thicknesses dropped 36.7-48.3 μm or the mRGCL volumes dropped to 0.495-0.613 mm;.CONCLUSION: The paediatric ON has best prognosis and young adult ON exhibits perfectly linear correlations of final vision and structural loss. The pRNFL and the mRGCL could be potential structural markers to predict the vision prognosis for varying-age ON patients.

Over the latest years,both randomized controlled trials(RCTs)and observational studies have provided solid basis for percutaneous coronary intervention as a treatment option for unprotected left main(LM)bifurcation coronary artery disease.A recent meta-analysis comparing the long-term outcome between LM bifurcation disease one-stent technique versus twostent technique,the study has demonstrated that one-stent strategy resulted associated with both major adverse cardiac events and target lesion revascularizations(TLRs)lower rates but resulted equivalent to two-stent strategy in terms of cardiovascular mortality,target lesion failure and stent thrombosis(ST)even at one-year and three-year follow-up.[1]

Neoplastic polyps in gallbladder: a retrospective study to determine risk factors and treatment strategy for gallbladder polyps

Background:Preoperative differentiation of malignant and premalignant gallbladder polyps(GBPs)from benign lesions is a key imperative to guide treatment *** aimed to characterize the various types of GBPs and sought to identify the risk factors for neoplastic *** findings may help optimize treatment ***:Retrospective analysis of 686 patients with post-cholecystectomy pathologically-proven GBPs between January 2003 and December *** patients were classified into non-neoplastic polyp group,benign neoplastic polyp group,and adenoma canceration *** features,ultrasound findings,and results of laboratory investigations and histopathological examination were reviewed and compared between the ***:Out of 686 patients,542(79.0%)had non-neoplastic polyps,134(19.5%)had neoplastic polyps,and 10(1.5%)had adenoma *** mean age was 46.06±12.12 years;383(55.8%)patients were *** median(25th percentile,75th percentile)time between diagnosis and surgery in the cholesterol polyp group[24(3.5,60)months]was significantly longer than that in adenoma[12(2,60)months]and adenoma canceration[5(0.475,12)months]*** mean diameter was 1.14±0.61 cm(range,0.5–8.4 cm).Three hundred twelve(45.5%)patients had solitary polyps and intralesional blood flow was observed in 41(6.0%)*** univariate analysis,age>49.5 years,polyp size>1.15 cm,solitary polyp,intralesional blood flow,absence of symptoms,and lack of cholecystitis showed a significant association with *** multivariate analysis,polyp size(>1.15 cm),intralesional blood flow,and lack of cholecystitis were independent predictors of ***:Polyp size>1.15 cm,intralesional blood flow,and lack of cholecystitis were predictors of neoplastic *** transformation of adenoma may occur over a relatively short time.

DNA damage stress induces the dissociation of Smurf1/2 from MDM2 in a slow manner

The tumor suppressor p53 locates at the key point of cell growth or apoptosis balance, and the expression level of p53 is tightly controlled by ubiquitin ligases including MDM2. Upon DNA damage stresses, p53 was accumulated and activated, leading to cell cycle arrest or apoptosis. We previously showed that Smad ubiquitylation regulatory factor 1/2 (Smurf1/2) promotes p53 degradation by interacting with and stabilizing MDM2, and consequently enhancing MDM2-mediated ubiquitylation of p53. However, it is unclear how the Smurf1-MDM2 interaction is regulated in response to DNA damage stress. Here, we show that in response to etoposide treatment Smurf1 dissociates from MDM2, resulting in MDM2 destabilization and p53 accumulation. The negative regulation of Smurf1 on apoptosis is released. Notably, this dissociation is a slow process rather than a rapid response, implicating high expression of Smurf1 might confer the resistance against p53 activation. Consistent with this notion, we observed that Smurf1/2 ligases are highly expressed in colon cancer, esophageal squamous cell carcinoma and pancreatic cancer tissues, suggesting the oncogenic tendency of Smurf1/2.
