This study aimed to investigate the osteo-inductive capacity of a fucoidan polysaccharide network derived from brown algae on human adipose-derived stem cells(HA-MScs)for bone *** physiochemical properties of the scaffold including surface morphology,surface chemistry,hydrophilicity,mechanical stiffness,and porosity were thoroughly *** in vitro and in vivo measurements implied a superior cell viability,proliferation,adhesion,and osteo-inductive performance of obtained scaffolds compared to using specific osteogenic induction medium with increased irregular growth of calcium crystallites,which mimic the structure of natural *** scaffold was highly biocompatible and suitable for cell *** examinations,such as quantification of mineralization,alkaline phosphatase,gene expression,and immunocytochemical staining of pre-osteocyte and bone markers confirmed that HAd-MScs differentiate into osteoblasts,even without an osteogenic induction *** study provides evidence for the positive relationship and synergistic effects between the physical properties of the decellularized seaweed scaffold and the chemical composition of fucoidan in promoting the osteogenic differentiation of ***,the natural matrices derived from brown seaweed offers a sustainable,cost-effective,non-toxic bioinspired scaffold and holds promise for future clinical applications in orthopedics.

Arthropod-borne chikungunya virus(cHIKV)infection can cause a debilitating arthritic disease in ***,there are no specific antiviral drugs and effective licensed vaccines against cHIKV available for clinical ***,we developed an m RNA-lipid nanoparticle(m RNA-LNP)vaccine expressing cHIKV E2-E1 antigen,and compared its immunogenicity with soluble recombinant protein s E2-E1 antigen expressed in S2 *** comparison,we first showed that recombinant protein antigens mixed with aluminum adjuvant elicit strong antigenspecific humoral immune response and a moderate cellular immune response in c57BL/6 ***,s E2-E1vaccine stimulated 12-23 folds more neutralizing antibodies than s E1 vaccine and s E2 ***,when E2-E1 gene was delivered by an m RNA-LNP vaccine,not only the better magnitude of neutralizing antibody responses was induced,but also greater cellular immune responses were generated,especially for cd8+T cell ***,E2-E1-LNP induced cd8~+T cells can perform cytotoxic effect in *** its better immunogenicity and convenience of preparation,we suggest that more attention should be placed to develop cHIKV E2-E1-LNP m RNA vaccine.

The fungus Trichophyton schoenleinii(***)is the causative agent of Trichophytosis and Tinea favosa of the scalp in certain regions of Eurasia and ***-man innate immune system plays an important role in combating with various pathogens including *** inflammasome is one of the most critical arms of host innate immunity,which is a protein complex controlling maturation of IL-1β.To clarify whether *** is able to activate the infl ammasome,we analyzed human monocytic cell line THP-1 for IL-1βproduction upon infec-tion with *** strain isolated from Tinea favosa patients,and rapid IL-1βsecretion from THP-1 cells was ***,applying competitive inhibitors and gene specifi c silencing with shRNA,we found that ***-oenleinii induced IL-1βsecretion,ASc pyroptosome for-mation as well as caspase-1 activation were all dependent on *** B activity,ROS production and K+effl ux were required for the infl ammasome activation by *** data thus reveal that the NLRP3 infl am-masome plays an important role in host defense against ***,and suggest that manipulating NLRP3 signaling can be a novel approach for control of diseases caused by *** infection.

context: The gut microbiota represents a complex ecosystem encompassing all unicellular microorganisms residing in the digestive tract, primarily bacteria, fungi, archaea, and even viruses. The relationship between the host and the microbiota is symbiotic: bacteria benefit from a stable environment, while the host gains numerous capabilities in terms of digestion, metabolism, nutrition, and immunity. However, numerous studies suggest that the gut microbiota plays a crucial role in various non-communicable diseases, including obesity, chronic inflammatory bowel diseases, allergic and immune disorders, behavioral disorders, and even certain cancers. The objective of our study was to characterize the gut microbiota of a group of breast cancer patients by comparing it to that of control subjects in côte d’ivoire, using a metagenomic approach. Method: A case-control study was conducted from May 2020 to September 2023. A total of 85 women (39 cases and 46 controls) were recruited, and stool samples were collected from both breast cancer patients and healthy women. Among these, ten (10) samples from patients and ten (10) samples from healthy women were randomly selected for the study of the gut microbiota. The gut microbiota was characterized by sequencing the V4 region of 16S rRNA using metagenomic NGS technology, and bioinformatic analysis was performed using the mothur pipeline. Results: In women with breast cancer, we observed a reduction in the relative abundance of the phyla Firmicutes and Bacteroidetes, as well as an increase in the phyla Actinobacteria and Verrucomicrobia.
