Comparative Study of Gut Microbiota in Breast Cancer Patients versus Controls in Abidjan, Côte d’Ivoire

Context: The gut microbiota represents a complex ecosystem encompassing all unicellular microorganisms residing in the digestive tract, primarily bacteria, fungi, archaea, and even viruses. The relationship between the host and the microbiota is symbiotic: bacteria benefit from a stable environment, while the host gains numerous capabilities in terms of digestion, metabolism, nutrition, and immunity. However, numerous studies suggest that the gut microbiota plays a crucial role in various non-communicable diseases, including obesity, chronic inflammatory bowel diseases, allergic and immune disorders, behavioral disorders, and even certain cancers. The objective of our study was to characterize the gut microbiota of a group of breast cancer patients by comparing it to that of control subjects in Côte d’Ivoire, using a metagenomic approach. Method: A case-control study was conducted from May 2020 to September 2023. A total of 85 women (39 cases and 46 controls) were recruited, and stool samples were collected from both breast cancer patients and healthy women. Among these, ten (10) samples from patients and ten (10) samples from healthy women were randomly selected for the study of the gut microbiota. The gut microbiota was characterized by sequencing the V4 region of 16S rRNA using metagenomic NGS technology, and bioinformatic analysis was performed using the mothur pipeline. Results: In women with breast cancer, we observed a reduction in the relative abundance of the phyla Firmicutes and Bacteroidetes, as well as an increase in the phyla Actinobacteria and Verrucomicrobia. Additionally, their microbiota exhibited lower Chao1 and Sobs diversities compared to the control women (p < 0.05). Molecular variance analysis (AMOVA) revealed a significant difference between the case and control groups (p < 0.001). Indigenous fermented foods are known for their nutritional and functional properties but they are often spoiled by pathogenic bacteria that can constitute a food safety problem. “Gappal” is a no-thermal treat food based on millet dough and milk and its production conditions can constitute a food safety problem. The aim of this study was to screen and identify LAB producing Bacteriocin-like inhibitory substances using a matrix similar to “Gappal”. The detection of potential BLIS was first performed using overlaid method after enrichment of samples in whey and millet dough. The isolates demonstrating inhibiting area were preselected, purified and tested for the presence of antibacterial properties using their neutralized cell-free culture supernatant and subsequently treated with catalase in combination with protease, pepsin or trypsin. The antimicrobial effect of two isolates (Gbf48 and Gbf50) after growth on MRS broth over 12 h at 30?C were active against E. faecalis ATCC 19433, M. luteus ATCC 49732, S. aureus ATCC 2523, L. monocytogenes, B. megaterium, B. sphaericus and B. cereus with an activity of 2560 AU/mL. The 16S RNA gene sequencing identification indicated that these isolates are Pediococcus acidilactici. Gbf 48 and Gbf 50 could be used to improve preservative factors for a controlled fermentation of non thermal treatment fermented food for their potential of acidification adds to BLIS production.

Bovine tuberculosis (bTB) is an endemic zoonosis significantly affects animal health in Burkina Faso. The primary causative agent is Mycobacterium tuberculosis (M. tuberculosis) complex, mainly M. bovis. Cattle are considered as natural reservoir of M. bovis. However, in Burkina Faso, the circulation of these strains remains poorly understood and documented. This study aimed to identify and characterize Mycobacterium strains from suspected carcasses during routine meat inspection at Bobo-Dioulasso refrigerated slaughterhouse. A prospective cross-sectional study was conducted from January 2021 to December 2022 on cases of seizures linked to suspected bovine tuberculosis. Microbiological and molecular analyzes were used for mycobacterial strain isolation and characterization. Out of 50 samples, 24% tested positive by microscopy and 12% by culture. Molecular analysis identified 6 strains of Mycobacteria, exclusively Mycobacterium bovis specifically the subspecies bovis (Mycobacterium bovis subsp bovis). In conclusion, M. bovis subsp bovis is the primary agent responsible for bovine tuberculosis in Bobo-Dioulasso. Continuous monitoring of mycobacterial strains is therefore necessary for the effective control of this pathology in the local cattle population.

Study of the Activation Process of a 2 m3 Pilot Biodigester on a Microfarm Site

A domestic balloon-type digester with 1200 liters of substrate and a 700-litre gas reserve was installed at the Université Gaston Berger pilot farm, which has 4 cows. After an initial load of 1000 L of water, 90 L of bovine rumen and 145 L of cow dung, the functional parameters of the reaction medium, i.e., temperature, pH, salinity and conductimetry, were regularly monitored at a rate of 3 tests per day until the thirtieth day, corresponding to the flame test and the start of analysis and daily loading of the biodigester. The analysis of the biogas obtained after the flame test showed us the considerable contribution of bovine hindquarters to the CH4 fraction, which reached 72.2% from the start of the production phase. As anaerobic digestion is both a complex and multiparametric process, microbiological analysis revealed the presence of several strains of bacteria in the substrate used. Among the most abundant were Escherichia coli, Klebsiella spp, non-fermentative Gram-negative bacilli and Enterococcus sp. However, the bacterial strain that interests us most in anaerobic digestion is the non-fermentative Gram-negative family. We see the identification and temporal monitoring of these families of bacteria as a major step forward in the control of anaerobic fermentation processes in the Sahelian context and in Senegal in particular.

First Research on Bat Bacteria in Burkina Faso (West Africa)

The world is in turmoil with the emergence of various diseases in which bats play a key part. Indeed, bats are known to host bacteria that can create major public health problems. The investigation into bat bacteria was carried out from December 2020 to September 2021 at seven sites in Burkina Faso. Bat specimen collection occurred from 6 pm to 5 am with mist nets. On each bat captured, an oral and rectum swab was taken to search for bacterial species using standard bacterial culture methods. A total of 204 bats representing 11 species were captured. 183 bat specimens were infected by at least one bacterium with a prevalence of 89.7%. 54 species of bacteria divided into 30 genera were identified from the 183 specimens. Bacterial species richness was the highest in the bat Mops condylurus (A. Smith, 1833) followed by Epomophorus gambianus (Ogilby, 1835). Genus Escherichia was the most frequent of the bat species. Genus Pseudomonas alone is represented by six species. The most infected site was the rectum, from which we isolated 44 species of bacteria out of the 54 species. The most infected locality was Ouagadougou. Bacteria are highly pathogenic to humans and may be responsible for public health problems, such as Shigella sp. was identified and bacteria known to cause harm to bats such as Yersinia and Pasteurella were also isolated. From this study, decisions on the management of public health problems can be considered drown to avoid the emergence and re-emergence of certain zoonotic diseases.

Molecular Characterization and Technological Properties of Lactic Acid Bacteria, Bacillus and Yeast of Probiotic Interest Isolated from Fermented Porridges

Cereal-based porridges are among fermented foods with a composite microbiota. The objective of this work is to characterize the microbiota of porridge. Samples of porridge were collected in Ouagadougou and analyzed according to standard methods in microbiology. The presumed strains obtained were characterized by polymerase chain reaction (PCR). Technological abilities were estimated by tests for resistance to acid pH, bile, antibiotics and antimicrobial, proteolytic and lipolytic activities. All presumptive Bacillus, lactic acid bacteria and yeast were characterized by PCR. Four (BC1a;BC9b;BC2a and BC1b) strains were confirmed to Bacillus by PCR, 6 strains to Lactobacillus and only to Saccharomyces cerevisiae. All strains were sensitive to two antibiotics gentamycin and imipenem. In contrast, all strains were resistant to oxacillin, amoxicillin-clavulanic acid, streptomycin, penicillin and ticarcillin. Strains tested were resistant to bile but in terms of pH this resistance was relative. Potential probiotic strains have been shown to be effective in inhibiting pathogens. Proteolytic and lipolytic activities were positive on all strains. The characterization of strains, although concerned with a non-exhaustive list of primers, has made it possible to confirm the strains that may be of good quality probiotics if a quantitative study is carried out on technological aptitudes.

Background: Herpes Simplex Virus type-2 (HSV-2) infection is one of the most common worldwide sexually transmitted and female sex workers are most at risk for these infections. Beside HSV-2, Human T-lymphotropic virus (HTLV) is one of these infections with a high risk of sexual transmission. HTLV-1 causes T-cell leukaemia and myelopathy. Little is known about the HSV-2, HTLV-1 and HTLV-2 infections among female sex workers in West Africa in general, and particularly in Burkina Faso. The aim of this study aimed to estimate HSV-2 and HTLV-1/2 seroprevalence among female sex workers, in Ouagadougou in order to inform HIV prevention programs. Methods: Archived sera samples of a biological and behavioral cross-sectional study among female sex workers (FSWs) in Ouagadougou were tested for HSV-2 and HTLV-1/2. Sera samples collected from February to May 2013 were previously tested for HIV and syphilis and stored with participants’ additional consent for further infection testing. Antibodies against HSV-2 and HTLV-1/2 detection tests were performed using Enzyme-Linked Immuno-Sorbent Assay (ELISA) methods. Results: HSV-2 seroprevalence was approximately 75.7% (95% CI: 70.8-79.9) among FSWs. It ranged from 69.3% among FSWs under 24 years of age, to 88.5% among those who were at least 30 years old. Furthermore, HSV-2 seroprevalence was high among FSW with no formal education (87.7%) and decreased significantly among those with at least primary education (76.0%) and others with secondary education and above (64.9%). Regarding HTLV-1/2, 11.2% (95% CI: 8.3 - 15.1) of FSWs were positive. By age group, 11.8% were positive among FSWs under 25 years of age;15.2% among those 25 to 29 years of age, and 5.8% among those over 30 years of age. Conclusion: HSV-2 and HTLV-1&2 infections are common among FSWs, which confirms the need for effective and preventive interventions such as sex education and early screening.

Introduction: The epidemiology of both hepatitis B virus (HBV) and hepatitis C virus (HCV) infections among drug users (DUs) is little known in West Africa. The study aimed to assess the prevalence of hepatitis B and C viruses among drug users in Burkina Faso. Methodology: This was a cross-sectional biological and behavioral survey conducted between June and August 2022, among drug users in Ouagadougou and Bobo Dioulasso, the two main cities of Burkina Faso. A respondent-driven sampling (RDS) was used to recruit drug users. Hepatitis B surface antigen was determined using lateral flow rapid test kits and antibodies to hepatitis C virus in serum determined using an Enzyme-Linked Immunosorbent Assay. Data were entered and analyzed using stata 17 software. Weighted binary logistic regression was used to identify the associated factors of hepatitis B and C infections and a p-value Results: A total of 323 drug users were recruited with 97.5% males. The mean age was 32.7 years old. The inhaled or smoked mode was the most used by drug users. The adjusted hepatitis B and hepatitis C prevalence among study participants were 11.1% and 2.3% respectively. The marital status (p = 0.001), and the nationality (p = 0.011) were significantly associated with hepatitis B infection. The type of drug used was not significantly associated with hepatitis B infection or hepatitis C infection. Conclusion: The prevalence of HBsAg and anti-HCV antibodies among DUs are comparable to those reported in the general population in Burkina Faso. This result suggests that the main routes of contamination by HBV and HCV among DUs are similar to those in the population, and could be explained by the low use of the injectable route by DUs in Burkina Faso.

Background: Some national and international strategies for the detection and prevention of emerging infectious diseases have been established across sectors. The capacity to carry out these tasks varies from country to country, and that remains largely undervalued. The Pasteur Institute of C?te d’Ivoire has created and implemented the capacity of national reference centers to fight against emerging and other infectious diseases. Objective: Show on the one hand the strategies used to develop the National Reference Centers and the IPCI and on the other hand the results obtained by performing these strategies. Method: Datas collection by documentary analysis (published scientific articles and grey literature) was done on Google Scholar, PUBMED and institutional reference documents. The documentary research was carried out to have a better understanding of strategies used to create and develop the NRCs in microbiology of communicable diseases. Results: Seven integrated strategies were launched: 1-training and workforce development;2-investigation of epidemics or public health events;3-strengthening laboratory epidemiological research;4-strengthening surveillance systems;5-improving communication with partners and stakeholders;6-building national and international collaborations;and 7-strengthening technical and technological platforms. In two decades, the number of researchers has risen from 10 in 2004 to ninety (90) in 2021, with 12 senior researchers and 32 junior researchers. A number of health service staff had attended a qualifying training course, 27 investigations into outbreaks and other public health events had been carried out, 18 short-term research projects had been launched, major surveillance programs and epidemiological research efforts on vector-borne, food-borne and nosocomial infections had begun, and several scientific manuscripts had been published or were edited in the writting press.
