Error-free mitosis depends on accurate chromosome attachment to spindle microtubules,which is monitored by the spindle assembly checkpoint(SAC)*** an upstream factor of SAC,the precise and dynamic kinetochore localization of Mps1 kinase is critical for initiating and silencing SAC ***,the underlying molecular mechanism remains ***,we demonstrated that the multisite interactions between Mps1 and Ndc80 complex(Ndc80C)govern Mps1 kinetochore ***,we identified direct interaction between Mps1 tetratricopeptide repeat domain and *** further identified that Mps1 C-terminal fragment,which contains the protein kinase domain and C-tail,enhances Mps1 kinetochore ***,Mps1 C-terminal fragment mediates its *** of C-tail attenuates the kinetochore targeting and activity of Mps1,leading to aberrant mitosis due to compromised SAC *** together,our study highlights the importance of Mps1 dimerization and multisite interactions with Ndc80C in enabling responsive SAC signaling.

Rarely,scientific developments centered around the patient as a whole *** multidisciplinary group,headed by gastrointestinal surgeons,applied this research philosophy considering the most important aspects of thediseases“colon-and rectal cancer”in the long-term *** expertcooperation/knowledge at the Comprehensive Cancer Center Ulm(CCCU)wereapplied in several phase III trials for multimodal treatments of primary tumors(MMT)and metastatic diseases(involving nearly 2000 patients and 64 centers),fortreatment individualization of MMT and of metastatic disease,for psychooncology/quality of life involving the patients’wishes,and for disease *** of the targets initially were heavily rejected/discussed in thescientific communities,but now have become standards in treatments andnational guidelines or are topics in modern translational research protocolsinvolving molecular biology for e.g.,“patient centered individualized treatment”.In this context we also describe the paths we had to tread in order to realize ournew goals,which at the end were highly beneficial for the patients from manypoints of *** description is also important for students and youngresearchers who,with an actual view on our recent developments,might want toknow how medical progress was achieved.

Studies found new-born microglia replenished the whole brain after selective elimination of microglia（＞99%） in adult mice. Immunohistochemical evidences suggested that repopulated microglia were differentiated from de novo progenitors expressing Nestin in the brain, which raised the possibility that the cross-lineage differentiation occurs in the mature brain. However, the origin of repopulating microglia has been hotly debated. In the present study, we investigated the origin of repopulating microglia by fate mapping. We first excluded that repopulated microglia were from blood cells. We then identified that repopulated microglia were not differentiated from Nestinpositive cells. Next we demonstrated all new-born microglia were derived from the proliferation of surviving microglia（＜0.90%）. Moreover, we confirmed that the whole brain transcriptome were not largely altered among stages of microglial elimination and repopulation. In summary, we concluded that de novo microglial progenitor cells were not existed in the adult brain.

The treatment scenario of colorectal cancer(CRC)has been evolving in recent years with the introduction of novel targeted agents and new therapeutic strategies for the metastatic *** extensive effort has been directed to the identification of predictive biomarkers to aid patients selection and guide therapeutic *** represents an irreplaceable tool to individualize patients treatment based on germline and tumor acquired somatic genetic variations able to predict drugs response and risk of *** growing knowledge of CRC molecular characteristics and complex genomic makeup has played a crucial role in identifying predictive pharmacogenomic biomarkers,while supporting the rationale for the development of new drugs and treatment *** validation of promising biomarkers,however,is often an *** recently,a deeper understanding of resistance mechanisms and tumor escape dynamics under treatment pressure and the availability of novel technologies are opening new perspectives in this *** review aims to present an overview of current pharmacogenomic biomarkers and future perspectives of pharmacogenomics in CRC,in an evolving scenario moving from a single drug-gene interactions approach to a more comprehensive genome-wide approach,comprising genomics and epigenetics.
