Antimicrobial Activity of Minocycline-Loaded Genipin-Crosslinked Nano-Fibrous Chitosan Mats for Guided Tissue Regeneration

Antimicrobial delivery has been advocated for guided tissue regeneration (GTR) or guided bone regeneration (GBR) therapies involving patients with aggressive or unresolved periodontitis/peri-implantitis. Electrospun chitosan membranes demonstrate several advantages over traditional GTR barrier membranes because they stimulate healing, mimic the topology of the extracellular matrix, and allow for diffusion of nutrients and wastes into/out of the graft site, and were shown to stimulate bone formation in a rabbit calvarial criticalsize defect model. Previously, we have shown improvements in mechanical properties and degradation kinetics by crosslinking electrospun membranes with 5 mM or 10 mM genipin. We have also demonstrated the ability of elecrospun chitosan membranes to inhibit lippopolysaccharide (LPS)-induced monocyte activation. In this study, minocycline was incorporated into the chitosan membrane by passive absorption at 5 or 10 mg/mL. The minocycline-loaded membranes and control membranes (carrier only) were tested against Porphyromonas gingivalis (P. gingivalis) by repeated zone of inhibition (ZOI) measurements. Testing showed that uncrosslinked and genipin-crosslinked membranes have similar capacity to absorb aqueous solutions (swelling ratio 1.7 - 2.2). Minocycline loading resulted in bacterial inhibition for up to 8 days from crosslinked membranes (with 11 mm initial ZOI) whereas uncrosslinked membranes loaded with minocycline only inhibited bacteria for 4 days (with 8 mm initial ZOI). These in vitro results suggest that genipin-crosslinked electrospun chitosan membranes loaded with minocycline may be able to reduce early bacterial contamination of GTR graft sites.

Regulation of TH17 cell differentiation by innate immune signals

Upon antigen stimulation, naive T helper cells differentiate into distinct lineages to attain specialized properties and effector functions. TH17 cells, a recently identified lineage of CD4＋ effector T cells, play a key role in the immune defense against fungi and extracellular bacteria, but also contribute to the pathogenesis of many autoimmune conditions. The differentiation of TH 17 cells is orchestrated by an intricate network of signaling pathways and transcriptional regulators in T cells. While the involvement of T cell-intrinsic pathways has been described extensively, we are just beginning to appreciate how TH17 cell development is shaped by extrinsic pathways, espec- ially the innate immune signals. Dendritic cells （DCs）, the most important cell type to bridge innate and adaptive immunity, drive TH17 cell differentiation by providing antigenic, costimulatory and cytokine signals. This is mediated by the recognition of innate and inflam- matory signals by DCs via pattern recognition receptors, cytokine receptors and other immunomodulatory receptors that in turn activate the intracellular signaling network. In particular, p38a MAP kinase has emerged as a critical pathway to program DC-dependent TH17 cell differentiation by integrating multiple instructive signals in DCs. Here, we summarize the current knowledge on the mechanisms by which DC-derived innate immune signals drive TH17 cell differentiation.

Cysteine Supplementation to Parenteral Nutrition Improves Red Blood Cell Glutathione Concentrations of Critically Ill Preterm Neonates

Premature neonates have immature antioxidant enzyme systems rendering them more susceptible to oxidative injury. One key antioxidant is glutathione (GSH). The rate limiting amino acid (AA) in GSH production is thought to be cysteine. Critically ill premature neonates who are parenterally fed are often supplemented with additional cysteine, yet the need for cysteine and optimal dose is unknown. This was a prospective, un-blinded, three-group, randomized crossover study aimed to evaluate three doses of cysteine by analyzing red blood cell (RBC) GSH, plasma AA, weight, and nitrogen balance. Neonates were randomized to receive 72 hours of each of the following cysteine doses: 10 mg/g AA, 20 mg/g AA, and 40 mg/g AA. GSH, plasma AAs, weight, and nitrogen balance were evaluated at baseline (after 72 hours of 0 mg/g AA), day three, day six, and day nine. Sixteen patients completed all doses of cysteine, which resulted in significantly increased RBC GSH concentrations over baseline. Plasma concentrations of cystine, total and free cysteine/cystine, glycine and serine increased with cysteine dose. All cysteine doses were associated with adequate weight gain, and positive nitrogen balance. These results are contrary to more recent studies of cysteine effect on RBC GSH concentrations in preterm neonates and infants, but may reflect the severity of illness in our study subjects, where cysteine requirements may be increased.

A tour-based analysis of travel mode choice accounting for regional transit service

The aim of this work is to explore the impact of regional transit service on tour-based commuter travel behavior by using the Bayesian hierarchical multinomial logit model, accounting for the spatial heterogeneity of the people living in the same *** two indicators, accessibility and connectivity measured at the zone level, the regional transit service is captured and then related to the travel mode choice behavior. The sample data are selected from Washington-Baltimore Household Travel Survey in 2007,including all the trips from home to workplace in morning hours in Baltimore city. Traditional multinomial logit model using Bayesian approach is also estimated. A comparison of the two different models shows that ignoring the spatial context can lead to a misspecification of the effects of the regional transit service on travel behavior. The results reveal that improving transit service at regional level can be effective in reducing auto use for commuters after controlling for socio-demographics and travel-related *** work provides insights for interpreting tour-based commuter travel behavior by using recently developed methodological approaches. The results of this work will be helpful for engineers, urban planners, and transit operators to decide the needs to improve regional transit service and spatial location efficiently.

Calcium Phosphate Coating over Silk Fibroin Film by Biomimetic Methods

To investigate the biomincralization behavior of silk fibroin and to valuate the biodegradation and biocompatibility of the hybrid biomaterial, the calcium phosphate deposits were identified with SEM, EDX, XRD and FTIR. The results reveal that supersaturated calcification solution is an effective method for the mineralization of fibroin film. Enzymatic degradation experiment demonstrates the biodegradability of the composites. Osteoblasts incubation shows an excellent cytocompatibility on the mincralized fibroin films.

Neonatal hyperbilirubinemia is a common phenomenon shortly after birth and a frequent cause of prolonged hospitalization[1].Increased enterohepatic circulation of indirect bilirubin is one of the elucidated mechanisms implicated in the pathophysiology of neonatal hyperbilirubinemia[2,3].According to various in vitro and in vivo evidence from animal[3]and human[2]trials,zinc salts have been depicted to reduce bilirubin levels.
