Selection of phages and conditions for the safe phage therapy against Pseudomonas aeruginosa infections

The emergence of multidrug-resistant bacterial pathogens forced us to consider the phage therapy as one of the possible alternative approaches to treatment. The purpose of this paper is to consider the conditions for the safe, long-term use of phage therapy against various infections caused by Pseudomonas aeruginosa. We describe the selection of the most suitable phages, their most effective combinations and some approaches for the rapid recognition of phages unsuitable for use in therapy. The benefi ts and disadvantages of the various different approaches to the preparation of phage mixtures are considered, together with the specifi c conditions that are required for the safe application of phage therapy in general hospitals and the possibilities for the development of personalized phage therapy.

Isolation of γδT cells from mouse small intestine

Gamma/delta T cells (γδТ cells) are among the first lymphoid cells appearing in the ontogenesis. Many of γδТ cells are located in the small intestine/lamina propria of mice and human, where they cooperate with different cells (B-1 cells, in particular) and influence their activity. However, such interactions are studied rather scanty, and the functional role of γδТ cells in the intestine?is not yet fully elucidated. To study the interactions of mouse γδТ cells with other cells it is necessary to have purified cell populations. Unfortunately, most approaches used for isolation of human γδТ cells are not suitable for isolation of mouse γδТ cells. The aim of the present study is the modification of the method of mouse intestinal γδТ cell isolation in the quantities sufficient for the in vivo and in vitro experiments.

Common Preconditions for Safe Phage Therapy of <i>Pseudomonas aeruginosa</i>Infections

One of the approaches to the treatment of infections caused by multiply-(MDR) or extra drug- resistant (XDR) pathogenic strains may be application of bacterial viruses (bacteriophages)— phage therapy. Results of a long, but quite limited use of phage therapy in several Eastern European countries, as well as experiments on animal models in Western countries support the possibility to use phage therapy. However, given the role of phages in the evolution of bacterial pathogens, it is necessary to discuss and evaluate negative consequences of mass introduction of phage therapy, as the measures necessary for its safe use. We discuss some actions in case of transiting to world-wide use of phage therapy with purpose to prolong the active life of phage therapy and to diminish possible complications.

Enteric viruses are the most common cause of acute gastroenteritis(AGE) in young children and a significant public health problem globally. Hospital admissions of children under 5 years of age with diarrhea are primarily associated with group A rotavirus(RVA) infection. In this retrospective study, the population structure of viruses linked to AGE etiology in young children hospitalized with AGE in Moscow was evaluated, and molecular characterization of RVA strains was *** specimens were collected from children under 5 years old hospitalized with AGE between 2009 and 2014 in Moscow, Russia. Multiplex real-time reverse transcription PCR was used to detect enteric viruses and for G/[P]-genotyping of isolated RVAs. Sequencing of RVA VP7 and VP4 cDNA fragments was used to validate the data obtained by PCRgenotyping. The main causes for hospitalization of children with AGE were RVA(40.1%), followed by noroviruses(11.4%), while adenoviruses, astroviruses, sapoviruses, enteroviruses, and orthoreoviruses were detected in 4.7%, 1.9%,1.4%, 1.2%, and 0.2% of samples tested, respectively. Nosocomial infections, predominantly associated with RVAs and noroviruses, were detected in 24.8% of cases and occurred significantly more frequently in younger infants. The predominant RVA genotype was G4 P[8], detected in 38.7% of RVA-positive cases, whereas genotypes G1 P[8], G9 P[8],G3 P[8], and G2P[4] were found in 11.8%, 6.6%, 4.2%, and 3.3% of cases, respectively. Together, the presence of circulating RVA strains with rare VP7 and VP4 gene variants(G6 and P[9]) highlights the need to conduct continuous epidemiological monitoring of RVA infection.
