NECAB family of neuronal calcium-binding proteins in health and disease

The N-terminal EF-hand calcium-binding proteins 1–3(NECAB1–3) constitute a family of predominantly neuronal proteins characterized by the presence of at least one EF-hand calcium-binding domain and a functionally less well characterized C-terminal antibiotic biosynthesis monooxygenase domain. All three family members were initially discovered due to their interactions with other proteins. NECAB1 associates with synaptotagmin-1, a critical neuronal protein involved in membrane trafficking and synaptic vesicle exocytosis. NECAB2 interacts with predominantly striatal G-protein-coupled receptors, while NECAB3 partners with amyloid-β A4 precursor protein-binding family A members 2 and 3, key regulators of amyloid-β production. This demonstrates the capacity of the family for interactions with various classes of proteins. NECAB proteins exhibit distinct subcellular localizations: NECAB1 is found in the nucleus and cytosol, NECAB2 resides in endosomes and the plasma membrane, and NECAB3 is present in the endoplasmic reticulum and Golgi apparatus. The antibiotic biosynthesis monooxygenase domain, an evolutionarily ancient component, is akin to atypical heme oxygenases in prokaryotes but is not wellcharacterized in vertebrates. Prokaryotic antibiotic biosynthesis monooxygenase domains typically form dimers, suggesting that calcium-mediated conformational changes in NECAB proteins may induce antibiotic biosynthesis monooxygenase domain dimerization, potentially activating some enzymatic properties. However, the substrate for this enzymatic activity remains uncertain. Alternatively, calcium-mediated conformational changes might influence protein interactions or the subcellular localization of NECAB proteins by controlling the availability of protein–protein interaction domains situated between the EF hands and the antibiotic biosynthesis monooxygenase domain. The Citron homology domain of MAP4Ks improves outcomes of traumatic brain injury

A supramolecular nanoprodrug for prevention of gallstone formation

Cholelithiasis affects approximately 10%-20% of the adult population globally. And cholesterol accumulation and nucleation of cholesterol crystals are commonly recognized as the primary process in the initiation and progression of gallstones. Hydroxypropyl-β-cyclodextrin(HPCD) is a supramolecular host compound that can solubilize cholesterol, potentially serving as a preventative or therapeutic agent for cholelithiasis. However, we found that the administration of HPCD treatment did not impede the formation of gallstones in mice, mainly attributed to the pre-complexation of its cavity during the transition process. Here we synthesized a prodrug of HPCD and prepared a HPCD nanoparticle(HPCD-NP), which can be transported efficiently to the gallbladder through the hepatobiliary system following an intravenous injection. In the bile, the HPCD-NP degraded into free HPCD, bound to cholesterol crystals and gallstones within the gallbladder and effectively increased cholesterol solubilization, leading to gallstones regression. Given the established safety of both HPCD and cyclodextrin-based nanoparticles in numerous animal and human studies, HPCD-NP shows considerable promise for the prevention and treatment of human cholelithiasis.

Flavones in pomelo peel resist fibril formation of human islet amyloid polypeptide

OBJECTIVE: To evaluate the effect of Persian shallot extract on the serum markers and blood pressure of patients with metabolic syndrome(MetS). METHODS: Fifty patients with Met S diagnosis were randomly assigned to the intervention(Persian shallot extract) and the control(placebo) group. Both groups received treatment for three months. Before the study and at the end of the study, 5 mL peripheral blood was taken from each patient. The measured factors included total antioxidant capacity(TAC), superoxide dismutase enzyme(SOD), malondialdehyde, oxidized low-density lipoprotein(Ox-LDL), apolipoprotein H(Apo-H), fasting blood glucose(FBS), total cholesterol, triglycerides, high-density lipoprotein(HDL), LDL, and systolic and diastolic blood pressure. RESULTS: At baseline, the evaluated parameters were not significantly different between the intervention and control groups. At the end of the study, the mean serum levels of malondialdehyde and ox-LDL were significantly lower in the intervention group. The mean FBS, cholesterol, triglycerides, and LDL were significantly lower in the intervention group. The mean TAC and HDL were significantly higher in the intervention group(P < 0.05). Moreover, the intervention group significantly reduced systolic and diastolic blood pressure. No other significant association was observed. CONCLUSION: Persian shallot extract has several beneficial effects in Met S patients, including optimizing oxidative balance, reducing blood pressure, fasting blood sugar, and blood lipid profile.

Biomechanical study of the visual system by ocular response analyzer investigates the inter-structural biological relationships,mechanics,and function of the visual *** review aimed to investigate the changes in corneal biomechanical parameters with age and *** articles published in PubMed between 2000 and 2021 were investigated and critiqued,and valid scientific evidence was collected,reviewed and concluded according to the inclusion and exclusion *** studies showed that corneal biomechanical changes occur infrequently in children up to the age of 20y,and with increasing age and wider age range,there was a significant decrease in corneal biomechanical indices,especially corneal *** children and adults,most studies have shown that these biomechanical indicators,especially corneal resistance factor,were higher in *** hormonal changes may contribute to this finding,the role of axial length and other biometric indicators should not be *** axial length,the intraocular pressure,and the corneal thickness are other factors associated with biomechanical parameters that should be taken into account in clinical diagnosis and management especially for patients undergoing refractive surgery as well as keratoconus patients.
