Trophic Interaction in a Portunus rituberculatus Polyculture Ecosystem Based on Carbon and Nitrogen Stable Isotope Analysis

Trophic interaction among various biomass groups in a swimming crab Portunus trituberculatus polyculture pond was investigated using carbon and nitrogen stable isotope analysis. The polycultured animal species also included white shrimp Litopenaeus vannamei, short-necked clam Ruditapes philippinarum, and redlip mullet Liza haematochila. The mean δ^(13)C value for all the biomass groups in polyculture ecosystem ranged from -25.61‰ to -16.60‰, and the mean δ^(15)N value ranged from 6.80‰ to 13.09‰. Significant difference in the δ^(13)C value was found between particulate organic matter(POM) and sediment organic matter(SOM)(P < 0.05), indicating that these two organic matter pools have different material sources. Assuming that a ^(13)C-enrichment factor of 1.00‰ and a ^(15)N-enrichment factor of 2.70‰ existed between consumer and prey, diets of the four cultured animals were estimated using a stable isotope mixing model. The estimated model results indicated that P. trituberculatus mainly feed on Aloidis laevis; L. vannamei mainly feed on shrimp feed; while A. laevis, R. philippinarum and L. haematochelia mainly feed on POM. Shrimp feed was also an important food source of R. philippinarum and L. haematochelia. The diets of P. trituberculatus, L. vannamei, R. philippinarum, and L. haematochila showed complementary effects in this polyculture ecosystem. Our finding indicated that the polyculture of these four organisms with suitable farming density could make an effective use of most of the food sources, which can make a highly efficient polyculture ecosystem.

Karyological analysis of the sea cicada Blepharipoda liberate Shen from the rizhao intertidal zone, China

Blepharipoda liberate Shen is a commercially valuable seafood species that has important ecological signi?cance in Shandong Province, China. Although B. liberate is crustacean, its external characteristics are not entirely those of shrimps or crabs. The question of whether B. liberate is a shrimp or a crab has been debated in recent years. We studied the karyotype of B. liberate by light microscopy using air-drying and spreading methods. We obtained mitotic chromosomal plates from B. liberate larvae, and from adult B. liberate females subsequent to egg-laying. The results revealed that B. liberate has 53 pairs of chromosomes(i.e., n =53 and 2 n =106), a characteristic shared with four species of crab. The karyogram of B. liberate consists of 25 metacentric, 14 submetacentric, 11 subtelocentric and 3 telocentric pairs. We did not ?nd any heteromorphosis sex chromosomes. Tissue from larvae, gills and ovaries can be used for chromosomal investigations, and we found similar lampbrush chromosomes in ovary cells. Comparatively speaking, larvae tissue is more practical, and ovary tissue is more suitable for the preparation of lampbrush chromosomes. B. liberate is more closely related to crabs than to shrimps, based on the numbers of chromosomes. The B. liberate karyotype reported here provides a basis for further comparative cytogenetic studies of species populations.

The pen shell,Atrina pectinata,distributes *** is one of the most important edible bivalves in east Asian ***,there are multiple difficulties in rearing pen shell larvae and juveniles because of their high *** understand the mechanism underlining such high mortality at the early embryonic development stage,we obtained approximately 100 million larvae during later breeding season in June,2017,and checked a large amount of mitotic chromosomal plates of the early embryos and post-spawning gonads tissue slices of their *** results showed that most diploid *** embryos have 17 pairs of chromosomes(2n=34)as their parents *** first pair of particularly large chromosomes are heterotypic in some diploid embryos while they are homomorphic in *** primary sex-determination chromosome type is XX/XY.A lot of triploid,pentaploid and aneuploid embryos with different numbers of the largest homomorphic or heteromorphic chromosomes were found due to the degeneration of overmatured parent gonads which hold normal *** larvae will die even though most of them may develop into the trochophore stage with 34 *** deficiency of chromosomes will cause a high rate of mortality in early embryos in late breeding *** findings should enrich the current knowledge of juvenile pen shell aquaculture.

The ultrastructure of the spermatozoon of Octopus ocellatus Gray, 1849 (Cephalopoda: Octopoda)

Morphology of the spermatozoon of Octopus ocellatus was studied by light, scanning electron, and transmission electron microscopes. Sperm are 600-700 um long, with a large number of granules in diameter about 130 um. Each spermatozoon is composed of a head, neck, and tail. The head is made up of an acrosomal complex anterior to the nucleus. The spiral acrosomal complex consists of an electron-lucent vesicle, lacuna, and an electron-dense acrosomal vesicle. Additionally, the spiral acrosomal vesicle has numerous equidistant striations, and is surrounded by many small granules （20 nm diameter）. A long straight nucleus, which is electron-densed, has a deep posterior concavity, the nuclear vacuole. At the terminal end of the nucleus is a sleeve-like structure with a concave posterior nuclear fossa （PNF）. The neck is short connecting the PNF. The basal body is located in the PNF and gives rise to the axoneme. This structure connects the head, neck, and tail. The tail is divided into a middle piece and a principal piece. The middle piece, having a 9＋9＋2 arrangement, is surrounded by a mitochondrial sheath and terminates by an electron-dense fibrous sheath. The principal piece is the longest part of the sperm with coarse fibers tapering posteriorly. The results of this study shall provide some useful information for artificial breeding of this species.

Embryonic and larval development in barfin flounder Verasper moseri (Jordan and Gilbert)

Broodstock of Verasper moseri （Jordan and Gilbert） aged 3-4 years old were selected, and reinforced cultivation was conducted to promote maturation under controlled water temperature and photoperiod conditions. Fertilized eggs were obtained by artificial fertilization, and the development of embryos, larvae and juveniles was observed continuously. The results showed that the fertilized eggs of V. moseri were spherical, with transparent yolk and homogeneous bioplasm, and had no oil globule inside. The average diameter of the eggs was 1.77＋0.02 mm. The eggs of V. moseri were buoyant in water with salinity above 35. The cleavage type was typical discoidal. Young pigment cells appeared when olfactory plates began to form. Hatching occurred at 187 h after fertilization at a water temperature of 8.5℃. The newly hatched larvae, floating on the water surface, were transparent with an average total length of 4.69±0.15 mm. During the cultivation period, when the water temperature was raised from 9 to 14.5℃, 4-day old larvae showed more melanophores on the body surface, making the larvae gray in color. The pectoral fins began to develop, which enabled the larvae to swim horizontally and in a lively manner. On days 7-8, the digestive duct formed. The yolk sac was small and black. The yolk sac was absorbed on day 11. Larvae took food actively, and body length and body height clearly increased. The rudiments of dorsal and anal fin pterygiophores were discernible and caudal fin ray elements formed on day 19. On day 24, the larval notochord flexed upwards, and the rays of unpaired fins began to differentiate. Pigment cells converged on the dorsal and anal fin rays, and the mastoid teeth on the mandible appeared. On day 29, the left eyes of juveniles began to move upwards. Depigmentation began in some juveniles and they became sandy brown in color on day 37. Most juveniles began to settle on the bottom of the tank. The left eyes of juveniles migrated completely to the right side on

Genetic identification of two species of Pleuronichthys by DNA barcoding

DNA barcoding is a new method for biological taxonomy, offering the ability to identify species from fragments in any life-history stage. Pleuronichthys cornutus and P. japonicus are two morphologically similar species. Pleuronichthys japonicus has never been found previously in China. However, in this study, we identified both species using DNA barcoding （cytochrome c oxidase subunit I （COI））, the mtDNA control region and cytochrome b. The results reveal that： l） intraspecific variation in the DNA barcode is much less than interspecific variation; 2） the two morphologically similar species were placed into separate clades distinguishable by high bootstrap values; 3） COI barcodes are more powerful for identifying the two species than the other two mtDNA fragments.

Evaluation of phenolic contents and antioxidant activities of brown seaweeds belonging to Turbinaria spp.(Phaeophyta,Sargassaceae) collected from Gulf of Mannar

Objective:To evaluate the antioxidant activities and total phenolic contents of brown seaweeds belonging to Turbinaria spp.[Turbinaria conoides(***)and Turbinaria ornata(***)collected from Gulf of Mannar of southeastern coast of India in various in vitro ***:The antioxidant activity was evaluated using different in vitro systems,viz.,I,I-diphenyl-2-picrylhydrazyl(DPPH),2,2'-azino-bis-3 ethylbenzothiozoline-6-sulfonic acid diammonium salt(ABTS),H_20_2/H0 radical scavenging,Fe^(2+)ion chelating ability,and reducing ***-Ciocalteu method was used to determine the total phenolic content of the extracts,and the results were expressed as mg of gallic acid equivalents(GE)/g of the seaweed *** acid-reactive substances assay was employed to assess the ability of the seaweed extracts to inhibit lipid ***:Ethyl acetate(EtOAc)fraction of *** registered significantly higher phenolic content(105.97 mg GE/g)than that of ***(69.63 mg GE/g).Significantly higher antioxidant potential as determined by DPPH(64.14%)radical scavenging activity was registered in EtOAc fraction of ***.A higher ABTS(?)radical scavenging(IC_(50)3.16μg/mL),Fe^(2+)chelating(IC_(50)0.46 mg/mL),H_2O_2 scavenging(IC_(50)4.25 mg/mL),lipid peroxidation inhibitory(TBARS,IC_(50)0.21 mg/mL),and reducing abilities(IC_(50)52.67 mg/mL)(P<0.05)were realized in EtOAc fraction of *** than other ***:This study indicated the potential use of *** and *** as candidate species to be used as food supplements/functional foods to increase shelf-life of food items for human consumption,and nutraceuticals to deter deleterious free radical-induced life-threatening diseases.

Biodiversity and seasonal variation of benthic macrofauna in Minicoy Island, Lakshadweep,India

From the Indian coast only limited data are available on the benthic fauna of the seagrass communities. In this study, seasonal variation in the distribution of macrobenthos and influence of environmental parameters was explored at four seagrass beds and two mangrove stations along the Minicoy Island, Lakshadweep, India, from September 1999 to August 2001. A total of 160 macrobenthic species from eight major groups represented the macrofauna of the Minicoy Island. Of the identified taxa, molluscs 70（gastropods 41.46%, bivalves 7.5%）, polychaetes 27（16.88%）, crustaceans 30（18.75%）, echinoderms 11（6.88%） and remaining others. Average seasonal abundance of benthic macrofauna ranged from 219 to 711 ind./m2, species diversity varied from 1.45 to 3.64 bits per individual, species richness index ranged from 4.01 to 26.17, evenness 0.69 to 1.66. In general, the higher abundance and species diversity was noticed in southern seagrass stations and northern seagrass stations, but in the mangrove stations comparatively low species diversity was observed. Three-way analysis of variance indicated that all communities resulted as being significantly different between seagrass and mangrove station, mainly when the seasonal interaction was considered. Multivariate analyses were employed to help define benthic characteristic and the relationship between environmental parameters at the six monitoring stations. Results of cluster analyses and multidimensional scale plot suggest that for mangrove region, different physiographic provinces, lower salinity, dissolved oxygen and sediment biotic structure have a higher influence on the species composition and diversity than other oceanographic conditions.

Genetic diversity of populations and clones of Rhopilema esculentum in China based on AFLP analysis

Amplified fragment length polymorphisms(AFLP) markers were developed to assess the genetic variation of populations and clones of Rhopilema esculentum Kishinouye(Scyphozoa,Rhizostomatidae).One hundred and seventy-nine loci from 56 individuals of two hatchery populations and two wild populations were genotyped with five primer *** polymorphic ratio,Shannon's diversity index and average heterozygosity were 70.3%,0.346 and 0.228 for the white hatchery population,74.3%,0.313,and 0.201 for the red hatchery population,79.3%,0.349,and 0.224 for the Jiangsu wild population,and 74.9%,0.328 and 0.210 for the Penglai wild population,***,all populations had a relatively high level of genetic diversity.A specific band was identified that could separate the white from the red hatchery *** was 84.85% genetic differentiation within *** cluster analysis using unweighted pair-group method with arithmetic mean(UPGMA) suggested that hatchery populations and wild populations could be *** the hatchery populations,the white and red populations clustered separately;however,for the wild populations,Penglai and Jiangsu populations clustered *** genetic diversity at the clone level was also *** data suggest that there are relatively high genetic diversities within populations but low genetic differentiation between populations,which may be related to the long-term use of germplasm resources from Jiangsu Province for artificial seeding and *** findings will benefit the artificial seeding and conservation of the germplasm resources.

Virus-phytoplankton adhesion:a new WSSV transmission route to zooplankton

The pathogenicity of white spot syndrome virus （WSSV） to zooplankton species, rotifer Brachionus urceus （Linnaeus）, copepod Acartia clausi （Giesbrecht） and mysid shrimp Neomysis awatschensis （ Brandt ）, was estimated by immersion challenge and virus - phytoplankton adhesion mute to investigate a potential new transmission mute of WSSV to zooplankton. WSSV succeeded in infecting these zooplankton species and nested-PCR revealed positive results for the virus - phytoplankton adhesion mute, whereas WSSV cannot infect zooplankton by immersion challenge. These results indicated that virus - phytoplankton adhesion route is a successful new transmission mute of WSSV to zooplankton and also implied that phytoplankton could be a carrier in WSSV transmission.
