Granule size distribution of wheat starch is an important characteristic that can affect its chemical composition and the functionality of wheat products. Two high-yield winter wheat cultivars were used to evaluate the effects of the application of exogenous ABA or GA during the reproductive phase of the initial grain filling on starch granule size distribution and starch components in grains at maturity. The results indicated that a bimodal curve was found in the volume and surface area distribution of grain starch granules, and a unimodal curve was observed for the number distribution under all treatments. The exogenous ABA resulted in a significant increase in the proportions （both by volume and by surface area） of B-type （〈9.9 Ixm in diameter） starch granules, with a reduction in those of A-type （〉9.9 ~tm） starch granules, while, the exogenous GA3 led to converse effects on size distribution of those starch granules. The exogenous ABA also increased starch, amylose and amylopectin contents at maturity but significantly reduced the ratio of amylose to amylopectin. Application of GA3 significantly reduced starch content, amylopectin content but increased the ratio of amylose to amylopectin. The ratio of amylose to amylopectin showed a significant and negative relationship with the volume proportion of granules 〈9.9 μm, but was positively related to the volume proportion of granules 22.8-42.8 μm.

Arylalkylamine N-acetyltransferase(aaNAT),considered a potential new insecticide target,catalyzes the acetylation of arylalkylamine substrates such as serotonin and dopamine and,hence,mediates diverse functions in ***,the origin of insect aaNATs(iaaNATs)and the evolutionary process that generates multiple aaNATs in mosquitoes remain largely ***,we have analyzed the genomes of 33 species to explore and expand our understanding of the molecular evolution of this gene family in *** show that aaNAT orthologs are present in Bacteria,Cephalochordata,Chondrichthyes,Cnidaria,Crustacea,Mammalia,Placozoa,and Teleoste,as well as those from a number of insects,but are absent in some species of Annelida,Echinozoa,and Mollusca as well as ***,more than 10 aaNATs were detected in the Culicinae subfamily of *** evolutionary analysis of aaNAT/aaNAT-like genes in mosquitoes reveals that tandem duplication events led to gene expansion in the Culicinae subfamily of mosquitoes more than 190 million years *** selection analysis demonstrates that mosquito aaNATs evolved under strongly positive pressures that generated functional diversity following gene duplication ***,this study may provide novel insights into the molecular evolution of the aaNAT family in mosquitoes.

Tree peony is a famous ornamental plant,while the low propagation rate is the main hurdles hindering the industry *** now,the highly efficient regeneration system for tree peony is not *** this study,using Paeonia ostii’Fengdan’mature embryos,the effects of variations in inoculation method,initiating culture,adventitious shoot induction,rooting media,plant growth regulators(PGRs),and a nonconventional PGR(plant extracts)on regeneration from explants were *** embryo cultures,embryonic callus induction rate of 1/4 embryos was the highest among those of embryos with other three technical treatments(whole embryos,1/2 embryos,and pieces of embryos).The woody plant medium(WPM)containing 1.0 mg·L^(-1)6-BA,0.5 mg·L^(-1)GA3,30.0 g·L^(-1)sucrose,and 3.0 g·L^(-1)phytagel significantly improved shoot induction and multiplication.3.0 mg·L^(-1)plant extracts promoted hypocotyl germination,rooting,and root growth,in direct embryo culture,and a combination of 3.0 mg·L^(-1)plant extracts+2.0 mg·L^(-1)IBA+1.5 mg·L^(-1)IAA produced optimal rooting induction rate for multiple shoots in direct embryo culture and indirect somatic *** the three in vitro micropropagation methods,the highest shoot proliferation coefficient(5.4±0.2)was obtained with indirect somatic ***,the propagation ability of shoots remains high,even when culture propagation was continued for more than two ***,a reliable system for plant regeneration from mature embryos derived from ***’Fengdan’callus and two direct embryo culture systems have been *** novel regeneration system could facilitate uniform seedling production.

Planktonic ostracods are small crustaceans abundant in marine ecosystem worldwide as appreciable part of marine zooplankton. Family Halocyprididae is a large group of halocyprid ostracods, and the tribe Conchoeciini has contained 21 genera previously. We described a further genus with a species Polyconchoecia commixtus gen. et sp. nov. from the middle of South China Sea in this study. The new species can be distinguished from related genera and species in having a unique combination of these characteristics： a lateral gland placed over right asymmetric gland and open near posterior margin; dense edge glands placed along ventral margin of carapace in line; shape of frontal organ; e-seta of first antenna is bare; endopod of second antenna has one small oval hump with central concave on mid-ventral margin, instead of processus mamillaris, exopod 1 has a small disto-dorsal spine, exopod 1 and 2 are fused, exopod 3 and 4 are bare; endopod 1 of mandible has one long ventral seta, endopod 2 has one ventral seta; maxillary endopod 1 has two basal setae; endopod 1 of fifth limb has only one ventral seta; endopod 1 of sixth limb and endopod 2 have no ventral seta. The definite distinctions in locations of major glands are the key characteristics of the new genus.

Involvement of phosphatidate phosphatase in the biosynthesis of triacylglycerols in Chlamydomonas reinhardtii

Lipid biosynthesis is essential for eukaryotic cells, but the mechanisms of the process in microalgae remain poorly understood. Phosphatidic acid phosphohydrolase or 3-sn-phosphatidate phosphohydrolase(PAP) catalyzes the dephosphorylation of phosphatidic acid to form diacylglycerols and inorganic orthophosphates. This reaction is integral in the synthesis of triacylglycerols. In this study, the mRNA level of the PAP isoform CrPAP2 in a species of Chlamydomonas was found to increase in nitrogen-free conditions. Silencing of the CrPAP2 gene using RNA interference resulted in the decline of lipid content by 2.4%–17.4%. By contrast, over-expression of the CrPAP2 gene resulted in an increase in lipid content by 7.5%–21.8%. These observations indicate that regulation of the CrPAP2 gene can control the lipid content of the algal cells. In vitro CrPAP2 enzyme activity assay indicated that the cloned CrPAP2 gene exhibited biological activities.

A group of lipoproteins with molecular sizes of approximately 30 kDa, referred to as 30K proteins, axe synthesized in fat body cells in the fifth instar larvae of silkworm, Bombyx mori. Analyzing the silkworm genome and its expressed sequence tags （ESTs）, we found 10 genes encoding 30K proteins, which are mainly distributed in three subfamilies. Of these, seven coding proteins were found to harbor the degrading sites of 30kP protease A, although the number of degrading sites may be different. As some potential core promoters and regulatory elements were supposed to be essential for gene transcription, the expression profiles of these genes were examined by semi-quantitative reverse transcription polymerase chain reaction. Eight 30K protein genes were detected to express luxuriantly in the fat body, while two were hardly expressed. Such results suggest that these 30K proteins may have different functions, and their adjacent regulatory elements play a crucial role in regulating their transcription.

TP53 mutation(TP53^(mut))occurs in 10–20%of diffuse large B-cell lymphoma(DLBCL)cases and serves as an unfavorable biomarker of DLBCL *** confers resistance to immunochemotherapy,high-dose chemotherapy,autologous stem cell transplantation,and anti-CD19 chimeric antigen receptor T-cell *** targeting of TP53^(mut) remains a significant challenge in DLBCL *** we assessed TP53^(mut) in 667 patients with newly diagnosed DLBCL,including 576 patients treated with immunochemotherapy rituximab,cyclophosphamide,doxorubicin,vincristine,and prednisone(R-CHOP)and 91 patients with decitabine plus R-CHOP(DR-CHOP,NCT02951728 and NCT04025593).TP53^(mut) independently predicted an inferior prognosis in R-CHOP-treated DLBCL,although this could be mitigated by DR-CHOP *** TP53^(mut) patients,multiple viral regulation pathways were repressed,resulting in the inhibition of immune modulation,as revealed by gene set enrichment ***53^(mut) DLBCL exhibited increased methyltransferase SUV39H1 expression and H3K9 trimethylation(H3K9me3),contributing to repression of endogenous retroviruses(ERVs)and immunosuppressive tumor *** TP53^(mut) DLBCL cell lines,decitabine down-regulated SUV39H1,inhibited H3K9me3 occupancy on ERVs,and triggered ERV expression,thereby unleashing interferons program and CD4^(+)T/CD8^(+)T cell *** silencing of SUV39H1 significantly abrogated decitabine-induced H3K9me3 inhibition and ERV *** TP53^(mut) patient-derived xenograft models and TP53^(mut) patients,the anti-tumor effect was improved upon the use of combined treatment of decitabine and doxorubicin via SUV39H1-H3K9me3-ERVs ***,our findings highlight an ERV regulatory circuitry in TP53^(mut) DLBCL and the crucial roles ERVs for epigenetically reprogramming tumor microenvironment for treating TP53^(mut)-driven cancers.

To understand how the nervous system develops from a small pool of progenitors during early embryonic development,it is fundamentally important to identify the diversity of neuronal subtypes,decode the origin of neuronal diversity,and uncover the principles governing neuronal specification across different *** single-cell analyses have systematically identified neuronal diversity at unprecedented scale and speed,leaving the deconstruction of spatiotemporal mechanisms for generating neuronal diversity an imperative and paramount *** this review,we highlight three distinct strategies deployed by neural progenitors to produce diverse neuronal subtypes,including predetermined,stochastic,and cascade diversifying models,and elaborate how these strategies are implemented in distinct regions such as the neocortex,spinal cord,retina,and ***,the identity of neural progenitors is defined by their spatial position and temporal patterning factors,and each type of progenitor cell gives rise to distinguishable cohorts of neuronal *** cues,spontaneous activity,and connectional pattern further reshape and diversify the fate of unspecialized neurons in particular *** illumination of how neuronal diversity is generated will pave the way for producing specific brain organoids to model human disease and desired neuronal subtypes for cell therapy,as well as understanding the organization of functional neural circuits and the evolution of the nervous system.

Review and Analysis on the Meridian Research of China over the Past Sixty Years

The meridian research situation and various meridian hypotheses of China in the past sixty years between 1950 and 2013 are summarized in the paper; possible existed problems in the process of current meridian research are analyzed. Based on previous research results, we proposed that the essence of meridian can not be explained by the reductive analysis method, meridian research should be carried out under the guidance of overall concept of Chinese medicine theory. In this paper, combined with coherence theory of biophoton, we put forward the quantum interference hypothesis of meridian, which provides a possible research idea for meridian study.
