Hepatoprotective effect of Averrhoea carambola fruit extract on carbon tetrachloride induced hepatotoxicity in mice

Objective:To investigate the hepatoprotective activity of Averroha carambola fruit extract against carbon tetrachloride induced hepatic ***:Hepatotoxicity was induced on albino mice by intraperitoneal administration of CCl4.half an hour after the administration of the last dose of the extract of Averroha carambola *** extract of the fruit of Averroha carambola was administered at a dose of 0.9 g/kg body weight once daily for seven *** hepatic injury and its prevention was assessed by the estimation of serum activities of alanine aminotransferase（ALT）,aspartate aminotransferase（AST）,alkaline phosphates（ALP）,glutathione level and histopathological studies of ***:Pre-treatment of mice with the fruit extract of Averrhoea carambola significantly reduced serum levels of ALT,AST and ALP enzyme and significantly increased the liver reduced glutathione levels 24 h after the administration of carbon tetrachloride.A marked improvement in the enzyme activities and the liver reduced glulathione level was observed in the pre-treated mice 4 days after the administration of carbon tetrachloride. Histopathological studies provided supportive evidence for the biochemical analvsis. Conclusions:The aqueous extract of the fruit of Averrhoea carambola has hepatoprotective effect against carbon tetrachloride induced liver damage in mice.

OBJECTIVE: To evaluate in vitro and in vivo antiarthritic potential of Solanum nigrum (S. nigrum). METHODS: Aqueous methanolic (70∶30) extract of S. nigrum was prepared. The in vitro antiarthritic effect was evaluated in terms of its inhibition of protein denaturation and membrane stabilization. While, formaldehyde, complete Freund's adjuvant (CFA) and Collagen induced arthritis rat models were used to study in vivo antiarthritic activities of S. nigrum at dose level of 200, 400 and 800 mg/kg. RESULTS: The extract exhibited inhibition of protein denaturation and protected red blood cell by stabilizing the membranes in a concentration dependent manner, with maximum effect attained at 800 μg/mL. Moreover, there was a marked reduction in paw edema observed in extract treated animals, when compared to arthritic control animals in all in vivo models and 800 mg/kg dose got maximum reduction of paw edema. The low survival rate of Kidney renal clear cell carcinoma(KIRC)patients is largely attributed to cisplatin *** than focusing solely on individual proteins,exploring protein-protein interactions could offer greater insight into drug *** this end,a series of in silico and in vitro experiments were conducted to identify hub genes in the intricate network of cisplatin resistance-related genes in KIRC *** genes involved in cisplatin resistance across KIRC were retrieved from the National Center for Biotechnology Information(NCBI)database using search terms as“Kidney renal clear cell carcinoma”and“Cisplatin resistance”.The genes retrieved were analyzed for hub gene identification using the STRING database and Cytoscape *** and promoter methylation profiling of the hub genes was done using UALCAN,GEPIA,OncoDB,and HPA ***,survival,functional enrichment,immune cell infiltration,and drug prediction analyses of the hub genes were performed using the cBioPortal,GEPIA,GSEA,TIMER,and DrugBank ***,expression and methylation levels of the hub genes were validated on two cisplatin-resistant RCC cell lines(786-O and A-498)and a normal renal tubular epithelial cell line(HK-2)using two high throughput techniques,including targeted bisulfite sequencing(bisulfite-seq)and RT-qPCR.A total of 124 genes were identified as being associated with cisplatin resistance in *** of these genes,MCL1,IGF1R,CCND1,and PTEN were identified as hub genes and were found to have significant(p<0.05)variations in their mRNA and protein expressions and effects on the overall survival(OS)of the KIRC ***,an aberrant promoter methylation pattern was found to be associated with the dysregulation of the hub *** addition to this,hub genes were also linked with different cisplatin resistance-causing ***,hub genes can be targeted with Alvocidib,Estradiol,Tretinoin,Capsaicin,Dronabinol,Metribolone,Calcitriol

Available reports have confirmed a link between bacterial infection and the progression of different types of cancers,including colon,lungs,and prostate *** we report the Chlamydia pneumonia proteins targeting in endoplasmic reticulum(ER)using in-silico approaches and their possible role in lung cancer *** predicted 48 proteins that target human ER,which may be associated with protein folding and protein-protein interactions during *** results showed *** proteins targeting human ER and their implications in lung cancer *** targeted proteins may be involved in competitive interactions between host and bacterial proteins,which may change the usual pathway functions and trigger the development of lung ***,*** unfolded protein accumulation in the human ER possibly induces ER stress,consequently activating the unfolded protein response(UPR),and providing a favorable microenvironment for cancer *** current study showed the *** protein targeting in ER of host cell and their implication in lung cancer *** results may help researchers better manage lung cancer and establish a molecular mechanism for *** lung cancer association.

Attenuation of experimentally induced diabetic neuropathy in association with reduced oxidative-nitrosative stress by chronic administration of <i>Momordica charantia</i>

Momordica Charantia (MC) is one of the most famous traditional plant worldwide, used for the treatment of diabetes and its complications. In the present study possible protective effect of MC in Streptozotocin (STZ) induced diabetic neuropathy in mice was evaluated. STZ induced diabetic mice were orally administered MC at various doses (200 - 800 mg/kg) for six weeks. Diabetes induced neuropathic pain was assessed by hot plate test, formalin test and tail flick test at the beginning and end of the study. Serum TBARS, NO and SOD levels were estimated at the end of the study as the markers of oxidative-nitrosative stress. Rotarod test was employed to assess the effect of treatment on motor coordination. The results showed that STZ induced diabetes significantly decreased the pain threshold as was indicated by increased flinching in formalin test and decreased withdrawal latency in hot plate and tail flick tests. Oxidative-nitrosative stress was significantly increased in diabetic animals. Chronic administration of MC significantly attenuated diabetes induced increase in flinches and decrease in withdrawal latency without impacting sensory and motor functions. MC administration also exhibited dose dependant reduction of hyperglycemia and serum TBARS, NO and SOD levels in diabetic mice. The results suggest that long term use of MC protects against diabetes induced neuropathy in association with attenuation of hyperglycemia and oxidative-nitrosative stress.
