Exploring the mechanism of Xiaoaiping Injection inhibiting autophagy in prostate cancer based on proteomics

Xiaoaiping(XAP) Injection demonstrates the anti-prostate cancer(PCa) effects, yet the underlying mechanism remains unclear. This study aims to investigate the impact of XAP on PCa and elucidate its mechanism of action. PCa cell proliferation was evaluated using a cell counting kit-8(CCK-8) assay. Cell apoptosis was assessed through Hoechst staining and Western blotting assays. Proteomics technology was employed to identify key molecules and significant signaling pathways modulated by XAP in PCa cells. To further validate potential key genes and important pathways, a series of assays were conducted, including acridine orange(AO)staining, transmission electron microscopy, and immunofluorescence assays. The molecular mechanism of XAP against PCa in vivo was examined using a PC3 xenograft mouse model. Results demonstrated that XAP significantly inhibited cell proliferation in multiple PCa cell *** C4-2 and prostate cancer cell line-3(PC3) cells, XAP induced cellular apoptosis, evidenced by reduced B-cell lymphoma 2(Bcl-2) levels and elevated Bcl-2-associated X(Bax) levels. Proteomic, immunofluorescence, and quantitative reverse transcription-polymerase chain reaction(q RT-PCR) investigations revealed a strong correlation between forkhead box O3a(Fox O3a) autophagic degradation and the anti-PCa action of XAP. XAP hindered autophagy by reducing the expression levels of autophagy-related protein 5(Atg5)/autophagy-related protein 12(Atg12) and enhancing Fox O3a expression and nuclear translocation. Furthermore,XAP exhibited potent anti-PCa action in PC3 xenograft mice and triggered Fox O3a nuclear translocation in tumor tissue. These findings suggest that XAP induces PCa apoptosis via inhibition of Fox O3a autophagic degradation, potentially offering a novel perspective on XAP injection as an effective anticancer therapy for PCa.

Fisetin micelles precisely exhibit a radiosensitization effect by inhibiting PDGFRβ/STAT1/STAT3/Bcl-2 signaling pathway in tumor

It is of great significance to find safe and effective radiosensitizers. A primary investigation has been made on fisetin's modification of radiation effect, but its radiosensitization and related mechanisms still need to be deeply clarified. Furthermore, fisetin with high hydrophobicity is difficult to dissolve in water,severely limiting its research and application. In this study, we fabricated a safe and soluble radiosensitizer fisetin micelle for precisely enhancing radiotherapy by inhibiting platelet-derived growth factor receptor-β(PDGFRβ)/signal transducer and activator of transcription 1(STAT1)/signal transducer and activator of transcription 3(STAT3)/B cell lymphoma 2(Bcl-2) signaling pathway in the tumor. Systematic and detailed studies were performed to verify its radiosensitization effect in vitro and in vivo. On the cellular level, fisetin micelles selectively increased the radiosensitivity of tumor cells(CT26 and 4T1 cells)and had little effect on the sensitivity of normal mouse cells(L929 cells) to radiation. In the mouse models of colon and breast cancers, fisetin micelles showed an efficient radiosensitization capacity without apparent toxicity. Additionally, we first found that fisetin micelles played a radiotherapy sensitization role by inhibiting the PDGFRβ/STAT1/STAT3/Bcl-2 pathway activity. In general, this work not only confirmed that fisetin micelles precisely exhibit a radiosensitization effect in vitro and in vivo, but also profoundly explored its mechanisms underlying, to provide a theoretical and experimental basis for the clinical application of fisetin micelles.

Objective To investigate the therapeutic effect of Xiangshao Granules(XSG)on post-stroke depression(PSD)and explore the underlying *** Forty-three C57BL/6J mice were divided into 3 groups:sham(n=15),PSD+vehicle(n=14),and PSD+XSG(n=14)groups according to a random number *** PSD models were constructed using chronic unpredictable mild stress(CUMS)after middle cerebral artery occlusion(MCAO).The sham group only experienced the same surgical operation,but without MACO and CUMS *** XSG group received XSG(60 mg/kg per day)by gavage for 4 *** mice in the sham and vehicle groups were given the same volume of 0.9%saline at the same *** body weight and behavior tests including open field test,sucrose preference test,tail suspension test,and elevated plus-maze test,were used to validate the PSD mouse ***-time fluorescence quantitative polymerase chain reaction(RT-qPCR),enzyme-linked immunosorbent assay(ELISA),and immunofluorescence staining were used to evaluate the anti-inflammatory effects of *** potential molecular mechanisms were explored and verified through network pharmacology analysis,Nissl staining,Western blot,ELISA,and RT-qPCR,*** The body weight and behavior tests showed that MCAO combined with CUMS successfully established the PSD *** alleviated neuronal damage,reduced the expressions of pro-apoptotic proteins Caspase-3 and B-cell lymphoma-2(BCL-2)-associated X(BAX),and increased the expression of anti-apoptotic protein BCL-2 in PSD mice(P<0.05 or P<0.01).XSG inhibited microglial activation and the expressions of pro-inflammatory cytokines including tumor necrosis factor-α,interleukin(IL)-1β,and IL-6 via the toll-like receptor 4/nuclear factor kappa-B signaling pathway in PSD mice(P<0.05 or P<0.01).Furthermore,XSG decreased the expression of indoleamine 2,3-dioxygenase1(IDO1)and increased the concentration of 5-hydroxytryptamine in PSD mice(P<0.05 or P<0.01).Conclusion XSG could reverse

Lymphoid and myeloid neoplasms are common malignant tumors in China,which threaten people’s health(1).To improve the diagnosis and treatment of lymphoma,the National Health Commission of the People’s Republic of China revised Chinese guidelines for diagnosis and treatment of malignant lymphoma in 2018(2).The new guidelines are based on the Classification of Tumours of Haematopoietic and Lymphoid Tissues revised by the World Health Organization in *** new edition provided principles of accurate pathological diagnosis of lymphoma on the basis of various scales including histomorphology,immunohistochemistry,fluorescent immunohybridization and genetics.

Relapsed/refractory classical Hodgkin lymphoma effectively treated with low-dose decitabine plus tislelizumab:A case report

BACKGROUND Academic studies have proved that anti-programmed death-1(PD-1)monoclonal antibodies demonstrated remarkable activity in relapsed/refractory classical Hodgkin lymphoma(cHL).However,most patients ultimately experienced failure or *** is urgent and necessary to develop a novel strategy for relapsed/refractory *** aim of this case report is to evaluate the combination approach of low-dose decitabine plus a PD-1 inhibitor in relapsed/refractory cHL patients with prior PD-1 inhibitor *** SUMMARY The patient was a 27-year-old man who complained of enlarged right-sided cervical lymph nodes and progressive pain aggravation of the right shoulder over the past 3 mo before *** analysis of lymph node biopsy was suggestive of *** patient experienced failure of eight lines of therapy,including multiple cycles of chemotherapy,PD-1 blockade,and anti-CD47 antibody ***-enhanced CT showed that the tumors of the chest and abdomen significantly shrunk or disappeared after three cycles of treatment with decitabine plus *** patient had been followed for 11.5 mo until March 2,2021,and no progressive enlargement of the tumor was *** The strategy of combining low-dose decitabine with tislelizumab could reverse the resistance to PD-1 inhibitors in patients with heavily pretreated relapsed/refractory *** therapeutic effect of this strategy needs to be further assessed.

Immune checkpoint inhibitors in malignant lymphoma: Advances and perspectives

The Incidence of lymphoma in Beijing:Comparing Results from MIS-CASS(2019)and Beijing Cancer Registry(2017)

Introduction:This study aimed to evaluate the performance of the Medical-Insurance-System-based Cancer Surveillance System(MIS-CASS)in estimating cancer incidence by comparing the results with the Beijing Cancer Registry(BCR),which is one of the highest-quality population-based cancer registries in ***:Using lymphoma as an example,we extracted relevant claims data from the administrative systems of medical insurance in Beijing(2012–2020)and estimated the most current lymphoma incidence in Beijing(2019)using a standard data processing *** absolute number of new cases,crude incidence rate,and age-standardized incidence rate of lymphoma were compared with the latest data reported by the BCR(2017).Results:Both lymphoma incidence rates and age distribution of new cases estimated based on MISCASS were similar to the BCR data(crude incidence rate:9.8/100,000 vs.10.6/100,000).However,because MIS-CASS included more designated hospitals and covered a larger local stationary population irrespective of household registration(hukou),the absolute number of incident lymphoma cases identified by MIS-CASS was 39.1%higher than that reported by the BCR(2,002 vs.1,439).Conclusions:The MIS-CASS approach reflected the actual cancer burden in a more complete and timely manner as compared with the current BCR,providing new insights for improving cancer surveillance strategies in China.

Prognostic value of pre-and post-transplantation 18F-fluorodeoxyglucose positron emission tomography results in non-Hodgkin lymphoma patients receiving autologous stem cell transplantation

Objective： High-dose chemotherapy （HDC） followed by autologous stem cell transplantation （ASCT） is the standard of care in the upfront or relapsed/refractory setting in some patients with non-Hodgkin lymphoma （NHL）. However, a proportion of patients do not respond to ASCT. lSF-fluorodeoxyglueose （FDG） positron emission tomography （PET）/computed tomography （CT） has been widely used for staging, response evaluation, and prognosis prediction. Here, we investigated the prognostic role of PET/CT in NHL patients before and after ASCT. Methods： A retrospective study was conducted at Peking University Cancer Hospital. All NHL patients who underwent ASCT between March 2010 and July 2016 were identified. Patients who had PET/CT scan before and after ASCT were included. Deauville criteria （5-point scale） were used to interpret PET scans. Univariate and multivariate survival analyses were performed using Cox regression. The predictive value of PET scanning was estimated by comparing the area under the receiver operating characteristic （ROC） curve. Results： In total, 79 patients were enrolled in this study. In univariate analysis, pre- and post-ASCT PET result was identified as prognostic factors for 3-year progression-free survival （PFS） and overall survival （OS）. Patients with negative pre-ASCT PET result demonstrated significantly better PFS （84.2% vs. 54.2%） and OS （89.2% vs. 63.6%） than patients with positive pre-ASCT PET result. PFS （91.6% vs. 25.3%） and OS （96.5% vs. 36.8%） were also significantly different between patients with negative and positive post-ASCT PET result. Multivariate analysis also showed a significant association between survival and post-ASCT PET result. ROC analysis revealed that the predictive value of post-ASCT PET result was superior to that of pre-ASCT PET result alone.
