Comparative analysis of metabolite changes in two contrasting rice genotypes in response to low-nitrogen stress

Identification of metabolites responsible for tolerance to low nitrogen availability(low-N)will aid in the genetic improvement of rice yield under nitrogen *** this study,a backcross introgression line(G9)and its recurrent parent Shuhui 527(SH527),which show differential responses to low-N stress,were used to identify metabolites associated with lown tolerance in *** in metabolite contents in the leaves of G9 and SH527 at three growth stages under low-N stress were assessed by gas chromatography–mass *** metabolites,including amino acids and derivatives,were highly enriched in G9 compared with SH527 under the control condition,suggesting that the two genotypes had basal metabolite ***-N stress induced genotype-specific as well as growth stagedependent metabolite *** induced specifically in G9 that were involved in glycolysis and tricarboxylic acid metabolism were enriched at the tillering and grain filling stages,and metabolites involved in nitrogen and proline metabolism were enriched at the booting *** of pyroglutamate,glutamate,2-oxoglutarate,sorbose,glycerate-2-P,and phosphoenolpyruvic acid in G9 suggests that these metabolites could be involved in low-N stress *** results presented here provide valuable information for further elucidation of the molecular mechanisms of low-N tolerance in crops.

Nitrogen fertilizer is required for high productivity of wheat(Triticum aestivum).However,excessive fertilizer use is unlikely to be effective at increasing yields and can cause detrimental effects on the environment and human *** key solution to achieve high yield with bwer fertilizer input and reducing environment pollution is to improve nitrogen use efficiency(NUE) in crops.N-defieiency can reduce leaf chlorophyll content and grain yields of *** this study,we observed the wheat line 178 A had higher leafN concentratbn and chbrophyll content than the wheat line 178 B under lown conditions during whole life,and showed longer leaf greenness persistence than 178 B during grain *** results indicate 178 A is more tolerant to ***480 recombinant inbred lines(RILs) derived from 178 A and 178 B were evaluated under bw-and high-N conditbns in two growing seasons to map genetic locus controlling leaf cobr and greenness persistence duration(GPD).We identified three major QTLs for leaf cobr at flowering stage under bw-N conditbns and three QTLs for *** most significant bcus for leaf cobr on chromosome 1 A also has pleiotropic effects on ***,the major focus for GPD on chromosome 6 A within a 0.49 cM interval explains the highest proportbn of the phenotypie variation According to published wheat genome sequence,this interval focates 30-33 Mb regbn on 6 *** region confers a NAC transcript factor NAM-A1,a homologous allele of NAM-B1 controlling leaf senescence and grain protein concentratbn Genomic DNA and cDNA sequence cfoning reveals that NAM-A1 in 178 A has a 1-bp frame-shift deletion leading premature stop of coding ***,candidate genes for these major QTLs still need to be further configuration.
