Background:Intracellular accumulation of the microtubule-associated protein tau and its hyperphosphorylated forms is a key neuropathological feature of Alzheimer’s disease(AD).Melatonin has been shown to prevent tau hyperphosphorylation in cellular and animal ***,the molecular mechanisms by which melatonin attenuates tau hyperphosphorylation and tau-related pathologies are not fully ***:Immunofluorescence,immunoblotting analysis and thioflavin-S staining were employed to examine the effects of early and late treatment of melatonin on tau-related pathology in hTau mice,in which nonmutated human tau is overexpressed on a mouse tau knockout ***-throughput microRNA(miRNA)sequencing,quantitative RT-PCR,luciferase reporter assay and immunoblotting analysis were performed to determine the molecular ***:We found that both early and late treatment of melatonin efficiently decreased the phosphorylation of soluble and insoluble tau at sites related to ***,melatonin significantly reduced the number of neurofibrillary tangles(NFTs)and attenuated neuronal loss in the cortex and ***,using miRNA microarray analysis,we found that miR-504-3p expression was upregulated by melatonin in the hTau *** administration of miR-504-3p mimics dramatically decreased tau phosphorylation by targeting p39,an activator of the well-known tau kinase cyclin-dependent kinase 5(CDK5).Compared with miR-504-3p mimics alone,co-treatment with miR-504-3p mimics and p39 failed to reduce tau ***:Our results suggest for the first time that melatonin alleviates tau-related pathologies through upregulation of miR-504-3p expression by targeting the p39/CDK5 axis and provide novel insights into AD treatment strategies.

A novel Co^Ⅱ-Fenton-like heterogeneous catalyst,(H3O)2[Co^Ⅱ(phen)(H2O)2]2[Mo^Ⅵ5O15(PO4)2]·4H2O (phen=1,10-phenanthroline,C12N2H8)(1),is synthesized and utilized for photocatalytic degradation of organic dyes and antibiotic in a wide range of *** experimental results show that 1 acting as the Fenton-like catalyst with H2O2 exhibits remarkable activity at pH 3–9 under vis-light irradiation and is merited with excellent recyclability and reusability.A variety of analytical methods,including in-situ electron paramagnetic resonance (EPR) spectroscopy and density functional theory (DFT) calculations,are applied to study the mechanism on generation of·OH and O2^·-radicals for photocatalytic *** suggests that,unlike the classical Fenton process involving the redox transformation of the central cations,the generation of·OH and O2^·-radicals is associated with the substitution of the coordinating water molecules at Co(Ⅱ) by H2O2 and/or OOH^-,followed by the light-driven O–O bond cleavage and *** outcomes of this study are striking which overcome the obstacles in the classical Fe^Ⅱ-Fenton process,including the slow redox transformation between Fe(Ⅱ) and Fe(Ⅲ) and the production of massive iron precipitates especially at elevated *** opens up new avenues for the development of the high-performance Fenton(-like) catalysts for photocatalytic degradation over extended pH and provides new insight into the related process.

Treadmill exercise and mesenchymal stem cell transplantation are both practical and effective methods for the treatment of cerebral ***,whether there is a synergistic effect between the two remains *** this study,we established rat models of ischemia/reperfusion injury by occlusion of the middle cerebral artery for 2 hours and reperfusion for 24 *** models were perfused with bone marrow mesenchymal stem cell-derived exosomes(MSC-exos)via the tail vein and underwent 14 successive days of treadmill *** assessment,histopathology,and immunohistochemistry results revealed decreased neuronal apoptosis and cerebral infarct volume,evident synaptic formation and axonal regeneration,and remarkably recovered neurological function in rats subjected to treadmill exercise and MSC-exos *** effects were superior to those in rats subjected to treadmill exercise or MSC-exos treatment ***,further investigation revealed that the activation of JNK1/c-Jun signaling pathways regulated neuronal apoptosis and synaptic-axonal *** findings suggest that treadmill exercise may exhibit a synergistic effect with MSC-exos treatment,which may be related to activation of the JNK1/c-Jun signaling *** study provides novel theoretical evidence for the clinical application of treadmill exercise combined with MSC-exos treatment for ischemic cerebrovascular disease.

Somaclonal variation in tissue culture is a common phenomenon induced by various external or internal environmental conditions,resulting in heritable or non-heritable alterations in gene expression. One crucial mechanism involved in plant growth and development is epigenetic regulation. A highly dynamic epigenome can respond to environmental changes by regulating gene expression. DNA methylation is one of these epigenetic modifications that can alter gene expression in tissue-cultured pineapple plants. The underlying mechanism of such somaclonal variations in pineapple and the epigenetic regulation involvement in somaclonal variations has not been studied. This study performed DNA methylome and transcriptome sequencing of wild-type(WT) and mutant pineapple plants(WS, HW, and TW). We observed altered DNA methylation patterns in chlorophyll development in the mutants. Specifically, we noticed that the methylation levels in the CHG and CHH contexts were lower in the gene body regions compared to the upstream and downstream regions. We identified several thousand differentially methylated regions(DMRs) located at the gene body regions, some of which overlapped with the differentially expressed genes(DEGs). Functional enrichment analyses suggested that these genes were involved in chlorophyll metabolism. Thus, our results revealed that the transcriptional regulation of many chlorophyll metabolic essential genes could be regulated by DNA methylation caused by somaclonal variations and provided insights into epigenetic mechanisms underlying the regulation of chlorosis in pineapple plants.

Recently it is a hot topic to make full use of high abundant Y element in Nd_(2)Fe_(14)B-type permanent *** contrast to Pr and Nd elements,Y shows different metallurgical behaviors during preparation *** this paper,we have explored the magnetic properties,microstructures and corrosion performance of Pr-Nd-Y-Fe-B magnets fabricated by spark plasma sintering(SPS)technique from the ribbons of nanocrystalline and amorphous precursors,*** coercivity and maximum energy product were improved for the magnets prepared from amorphous precursor materials(denoted as SPS-A hereafter)compared with the magnets prepared from crystalline precursor materials(denoted as SPS-C hereafter).Magnetic properties of Jr=0.79 T,Hci=864 k A/m,and(BH)_(max)=102 k J/m^(3)were obtained for SPS-A *** contrast with SPS-C magnets,the magnetic properties of SPS-A magnets are not so sensitive to the preparation conditions,which is quite beneficial to the homogeneity of microstructure and enhancement of coercivity for large-scale production of the designated ***(Pr,Nd,Y)-rich phase was found out in SPS-C *** and Nd elements are rich at grain boundary while Y is distributed uniformly at main phase and grain boundary *** strip grains and equiaxed grains exist in SPS-C and SPS-A magnets,*** enhanced magnetic properties for SPS-A magnets are accredited to the uniform distribution of rare-earth-rich phase and low demagnetization *** is revealed by electrochemical test and dipping test that the corrosion potential is more positive and the corrosion rate is slower for the SPS-A magnets in 3.5 wt.%Na Cl *** work is also expected to shed light on developing the nanocrystalline Pr-Nd-Y-Fe-B SPSed high-performance magnets in industry.

The East Tianshan contains many late Paleozoic magmatic and polymetallic *** studies demonstrate that the early Paleozoic volcanic rocks discovered in the northern region of East Tianshan can be subdivided into the Daliugou,Hongliuxia and Kalatag ***,we report zircon U-Pb ages of volcanic rocks and quartz diorite porphyry,together with whole-rock geochemical and Sr-Nd isotope data of early Paleozoic volcanic rocks,in order to investigate their petrogenesis and geodynamic *** U-Pb analyses of the rocks suggest that the Kalatag Formation formed at ca.438-413 *** characteristics of the early Paleozoic volcanic rocks exhibit typical subduction-related *** have high Mg^(#)(44-75),positiveε_(Nd)(t)values(0.25-7.88),low^(87)Sr/^(86)Sr ratios(0.70457 to 0.70588)and young two-stage Hf isotope model ages(551-446 Ma),consistent with a depleted mantle origin with limited crustal *** basaltic andesite and andesite have relatively high MgO contents(6.1-9.4 wt%),suggesting that they belong to high-magnesian *** were most likely derived from the partial melting of mantle peridotite caused by the addition of fluids released by subducted oceanic *** on regional geology,geochemical characteristics and previous studies,we infer that the southward subduction of the Junggar Ocean slab resulted in the generation of early Paleozoic volcanic rocks and arc setting-related metallogenic systems in the Kalatag area.

Background:Although many causative genes of hereditary spastic paraplegia(HSP)have been uncovered in recent years,there are still approximately 50% of HSP patients without genetically diagnosis,especially in autosomal recessive(AR)HSP *** studies have been performed to determine the genetic spectrum and clinical profiles of recessive HSP patients in the Chinese ***:In this study,we investigated 24 Chinese index AR/sporadic patients by targeted next-generation sequencing(NGS),Sanger sequencing and multiplex ligation-dependent probe amplification(MLPA).Further functional studies were performed to identify pathogenicity of those uncertain significance ***:We identified 11 mutations in HSP related genes including 7 novel mutations,including two(p.V1979_L1980delinsX,p.F2343 fs)in SPG11,two(p.T55 M,p.S308 T)in AP5Z1,one(p.S242N)in ALDH18A1,one(p.D597fs)in GBA2,and one(p.Q486X)in ATP13A2 in 8 index patients and their family *** in ALDH18A1,AP5Z1,CAPN1 and ATP13A2 genes were firstly reported in the Chinese ***,the clinical phenotypes of the patients carrying mutations were described in *** mutation(p.S242 N)in ALDH18A1 decreased enzyme activity of P5CS and mutations(p.T55 M,p.S308 T)in AP5Z1 induced lysosomal ***:Our results expanded the genetic spectrum and clinical profiles of AR-HSP patients and further demonstrated the efficiency and reliability of targeted NGS diagnosing suspected HSP patients.

Identification of aeolian loess deposits on the Indo-Gangetic Plain(India)and their significance

Aeolian loess deposits contain abundant information about the evolution of the *** example,paleoclimate changes recorded in Chinese loess area obtained significant achievement in the past few *** to Chinese loess,research on Indian loess is ***,most studies focus on the Kashmir area located in the southern Himalayas,and studies on other areas are ***,field observations demonstrate that the sediments around the New Delhi-Agra-Jaipur Plain are similar to Chinese loess-paleosol *** example,the boundary between two strata is transitional and without horizontal ***,obvious pedogenic horizons developed among sediment sequences,probably indicating unrecognised aeolian deposits in the Indo-Gangetic Plain(IGP).To confirm this,pilot samples were obtained from the IGP and detailed indoor measurements *** results indicate that the distribution patterns of particle size and rare earth elements(REE) of the pilot samples are similar to Chinese ***,the scanning electronic microscopy(SEM) images of pilot samples show obvious conchoidal fractures,dash-shaped concavities,and abundant small pits that usually form through mechanical *** are typical characteristics of aeolian *** addition,environmental and rock magnetic measurements indicate that the dominant magnetic minerals in the pilot samples are magnetite and maghemite,and that they likely contain small amounts of ***,conventional magnetic parameters are comparable with Chinese *** on this,aeolian loess deposits are widely distributed in the IGP,which may have promoted the development of Indian farming and contributed towards the prosperity of ancient Indian *** study also provides a new and valuable record for the research on paleoclimate changes in the study area in the future.
