Effect of miR-124 Overexpression under Wnt/β-catenin signaling Pathway on Inflammatory Factor and Epidermal Growth Factor in Rats with Precancerous Gastric Lesions

Objective: To investigate the effects of miR-124 overexpression on inflammatory factors and epidermal growth factor in rats with gastric precancerous lesions by modulating the Wnt/β-catenin signaling pathway. Methods: Eighty sPF-grade Wistar rats were selected as the research subjects. After adaptive feeding,they were divided into 4 groups using a random number table method. The control group was fed a regular diet, while the model group, low expression group, and overexpression group of rats were treated with N-methyl-N'-nitro-N-nitrosoguanidine to establish a model of gastric cancer precancerous lesions. After successful modeling, the control group and model group rats were injected intraperitoneally with 0.9% sodium chloride solution, the low-expression group rats were injected intraperitoneally with miR-124-5p inhibitor, and the over-expression group rats were injected intraperitoneally with miR-124-5p mimic. After 8 weeks of continuous injection, the levels of miR-124,Wnt/β-catenin signaling pathway-related proteins, inflammatory factors, and epidermal growth factor were observed and compared among the four groups of rats. Results: The differences in the relative expression of miR-124 among the four groups of rats were statistically significant(Ps in the expression of Wnt protein andβ-catenin protein among the four groups of rats were statistically significant(Pssion of Wnt protein and β-catenin protein in the overexpression group was significantly higher than that of the control group, and significantly lower than that in the model group and the low-expression group; the levels of IL-1β, IL-6, and TNF-αof rats in the four groups of rats IL-1β, IL-6 and TNF-α levels were statistically different(Ps of IL-1β, IL-6 and TNF-α in rats in the overexpression group were higher than those in the model group and significantly lower than those in the model group and the low-expression group(P<0.05); the differ

Objective To observe the value of preoperative CT radiomics models for predicting composition of in vivo urinary *** Totally 543 urolithiasis patients were retrospectively enrolled and divided into calcium oxalate monohydrate stone group(group A,n=373),anhydrous uric acid stone group(group B,n=86),carbonate apatite group(group C,n=30),ammonium urate stone group(group D,n=28)and ammonium magnesium phosphate hexahydrate stone group(group E,n=26)according to the composition of calculi,also divided into training set and test set at the ratio of 7∶*** features were extracted and screened based on plain CT images of urinary *** binary task models(model A—E corresponding to group A—E)and a quinary task model were constructed using least absolute shrinkage and selection operator algorithm for predicting the composition of calculi in *** receiver operating characteristic curves were drawn,and the area under the curves(AUC)were calculated to evaluate the predictive efficacy of binary task models,while the accuracy,precision,recall and F1 score were used to evaluate the predictive efficacy of the quinary task *** All binary task models had good efficacy for predicting the composition of urinary calculi in vivo,with AUC of 0.860—0.948 in training set and of 0.856—0.933 in test *** accuracy,precision,recall and F1 score of the quinary task model for predicting the composition of in vivo urinary calculi was 82.25%,83.79%,46.23%and 0.596 in training set,respectively,while was 80.63%,75.26%,43.48%and 0.551 in test set,*** Binary task radiomics models based on preoperative plain CT had good efficacy for predicting the composition of in vivo urinary calculi,while the quinary task radiomics model had high accuracy but relatively poor stability.

Post-traumatic peritendinous adhesion presents a significant challenge in clinical *** study proposes the use of diamond-like carbon(DLC)deposited on polylactic acid(PLA)membranes as a biophysical mechanism for anti-adhesion barrier to encase ruptured tendons in tendon-injured *** results indicate that PLA/DLC composite membrane exhibits more efficient anti-adhesion effect than PLA membrane,with histological score decreasing from 3.12±0.27 to 2.20±0.22 and anti-adhesion effectiveness increasing from 21.61%to 44.72%.Mechanistically,the abundant C=O bond functional groups on the surface of DLC can reduce reactive oxygen species level effectively;thus,the phosphorylation of NF-κB and M1 polarization of macrophages are ***,excessive inflammatory response augmented by M1 macrophage-originated cytokines including interleukin-6(IL-6),interleukin-1β(IL-1β),and tumor necrosis factor-α(TNF-α)is largely *** biocompatibility evaluation,PLA/DLC membrane is slowly absorbed within tissue and displays prolonged barrier effects compared to traditional PLA *** studies show the DLC depositing decelerates the release of degradation product lactic acid and its induction of macrophage M2 polarization by interfering esterase and PLA ester bonds,which further delays the fibrosis *** was found that the PLA/DLC membrane possess an efficient biophysical mechanism for treatment of peritendinous adhesion.
