Recurrence quantification analysis on pulse morphological changes in patients with coronary heart disease

OBJECTIVE： To show that the pulse diagnosis used in Traditional Chinese medicine, combined with nonlinear dynamic analysis, can help identify car- diovascular diseases. METHODS： Recurrence quantification analysis （RQA） was used to study pulse morphological changes in 37 inpatients with coronary heart dis- ease （CHD） and 37 normal subjects （controls）. An in- dependent sample t-test detected significant differ- ences in RQA measures of their pulses. A support vector machine （SVM） classified the groups accord- ing to their RQA measures. Classic time-domain pa- rameters were used for comparison. RESULTS： RQA measures can be divided into two groups. One group of measures [ecurrence rate（RR）, determinism （DEL）, average diagonal line length （L）, maximum length of diagonal structures （Lmax）, Shannon entropy of the frequency distribu- tion of diagonal line lengths （ENTR）, laminarity （LAM）, average length of vertical structures （TT）, maximum length of vertical structures （Vmax）] showed significantly higher values for patients with CHD than for normal subjects （P〈0.0S）. The other measures （RR_std, L_std, Lmaxstd, TT_std, Vmax_std） showed significantly lower values for the CHD group than for normal subjects （P〈0.05）. SVM classification accuracy was higher with RQA measures： With RQA （16 parameters） accuracy was at 88.21%, and with RQA（12 parameters） accuracy was at 84.11%. In contrast, with classic time-do- main （15 parameters） accuracy was 75.73%, and with time-domain （7 parameters） accuracy was 74.7O%. CONCLUSION： Nonlinear dynamic methods such as RQA can be used to study functional and struc- tural changes in the pulse noninvasively. Pulse sig- nals of individuals with CHD have greater regulari- ty, determinism, and stability than normal subjects, and their pulse morphology displays less variabili- ty. RQA can distinguish the CHD pulse from the healthy pulse with an accuracy of 88.21%, thereby providing an early diagnosis of cardiovascular dis- eases such as CHD.

Obesity and associated metabolic disorders represent a major societal challenge in health and quality of life with large psychological consequences in addition to physical disabilities. They are also one of the leading causes of morbidity and mortality. Although, different etiologic factors including excessive food intake and reduced physical activity have been well identified, they cannot explain the kinetics of epidemic evolution of obesity and diabetes with prevalence rates reaching pandemic proportions. Interestingly, convincing data have shown that environmental pollutants, specifically those endowed with endocrine disrupting activities, could contribute to the etiology of these multifactorial metabolic disorders. Within this review, we will recapitulate characteristics of endocrine disruption. We will demonstrate that metabolic disorders could originate from endocrine disruption with a particular focus on convincing data from the literature. Eventually, we will present how handling an original mouse model of chronic exposition to a mixture of pollutants allowed demonstrating that a mixture of pollutants each at doses beyond their active dose could induce substantial deleterious effects on several metabolic end-points. This proof-of-concept study, as well as other studies on mixtures of pollutants, stresses the needs for revisiting the current threshold model used in risk assessment which does not take into account potential effects of mixtures containing pollutants at environmental doses, e.g., the real life exposure. Certainly, more studies are necessary to better determine the nature of the chemicals to which humans are exposed and at which level, and their health impact. As well, research studies on substitute products are essential to identify harmless molecules.

cardiovascular disease is the most common cause of death in the diabetic population and is currently one of the leading causes of death in the United States and other industrialized countries. The health care expenses associated with cardiovascular disease are staggering, reaching more than US$350 billion in 2003. The risk factors for cardiovascular disease include high fat/cholesterol levels, alcoholism, smoking, genetics, environmental factors and hypertension, which are commonly used to gauge an individual's risk of cardiovascular disease and to track their progress during therapy. Most recently, these factors have become important in the early prevention of cardiovascular diseases. Oxidative stress, the imbalance between reactive oxygen species production and breakdown by en- dogenous antioxidants, has been implicated in the onset and progression of car- diovascular diseases such as congestive heart failure and diabetes-associated heart dysfunction (diabetic cardiomyopathy). Antioxidant therapy has shown promise in preventing the development of diabetic heart complications. This review focuses on recent advances in oxidative stress theory and antioxidant therapy in diabetic cardiomyopathy, with an emphasis on the stress signaling pathways hypothesized to be involved. Many of these stress signaling pathways lead to activation of reactive oxygen species, major players in the development and progression of diabetic cardiomyopathy.

Proteome-wide alterations in an industrial clavulanic acid producing strain of Streptomyces clavuligerus

The usefulness of genetic/metabolic engineering for further improvement of industrial strains is subject of discussion because of the general lack of knowledge on genetic alterations introduced by iterative cycles of random mutagenesis in such *** industrial clavulanic acid(CA)-overproducer Streptomyces clavuligerus DEPA was assessed to understand proteome-wide changes that have occurred in a local industrial CA overproducer developed through succesive mutagenesis *** proteins that could be identified corresponded to 33 distinct ORFs for underrepresented ones and 60 ORFs for overrepresented *** CA biosynthetic enzymes were overrepresented in *** DEPA;carboxyethylarginine synthase(Ceas2),clavaldehyde dehydrogenase(car)and carboxyethyl-arginine betalactam-synthase(Bls2)whereas the enzymes of two other secondary metabolites were underrepresented along with two important global regulators[two-component system(TCS)response regulator(SCLAV_2102)and TetR-family transcriptional regulator(SCLAV_3146)]that might be related with CA production and/or differentiation.g-butyrolactone biosynthetic protein AvaA2 was 2.6 fold underrepresented in *** *** levels of two glycolytic enzymes,2,3-bisphosphoglycerate-dependent phosphoglycerate mutase and phosophoglycerate kinase were found decreased while those of dihydrolipoyl dehydrogenase(E3)and isocitrate dehydrogenase,with two isoforms were found as significantly increased.A decrease of amino acid metabolism,methionine biosynthesis in particular,as well as S-adenosylmethionine synthetase appeared as one of the prominent mechanisms of success of *** DEPA strain as a prolific producer of *** levels of two enzymes of shikimate pathway that leads to the production of aromatic amino acids and aromatic secondary metabolites were also *** of the overrepresented stress proteins in *** DEPA included polynucleotide phosphorylase/polyadenylase(PNPase),ATP

Targeting Metabolism to Improve car-T Cell Immunotherapy

The transfer of genetically-engineered chimeric antigen receptor(car)-T cells has recently become one of the most promising approaches in the treatment of *** normal T cells,car-T cells are dependent on an adequate supply of nutrients and energy to be able to proliferate and mediate anti-tumor ***,car-T cell proliferation and cytotoxicity are limited by the elevated metabolic demands of actively dividing cancer cells,which deplete nutrients from the tumor microenvironment(TME).Thus,a variety of TME-imposed metabolic“barriers”must be overcome by artificially altering the metabolism of car-T cells to increase their clinical performance,especially in solid *** this review,we summarize the metabolic interactions between T cells and the TME,discuss the metabolic reprogramming of T cells,present recent progress in the application of metabolic interventions in car-T cell therapy,and finally allude to future directions in this field.

Transcriptomes of Litopenaeus vannamei reveal modulation of antioxidant system induced by dietary archaeal carotenoids

Oxidative stress induced by factors such as ammonia nitrogen has become a major issue in shrimp *** effects of carotenoids on the growth and antioxidant capability of Litopenaeus vannamei juveniles were investigated in this study using dietary archaeal carotenoids *** four weeks,shrimp were given diets containing 0 mg/kg(Ctrl)and 55.98 mg/kg(car)archaeal *** archaeal carotenoids significantly enhanced the astaxanthin content in shrimp muscles and carapaces,as well as the superoxide dismutase(SOD)and glutathione peroxidase(GSH-Px)activity(Pcar group significantly decreased(Pcarotenoids supplementation.A total of 1536 differentially expressed genes(DEGs)were detected,including 538 upregulated DEGs and 998 downregulated *** functional enrichment analysis between Ctrl and car indicated that 26 GO terms including extracellular region,metabolic process,and proteolysis were *** KEGG pathway enrichment analysis revealed that the amino sugar and nucleotide sugar metabolism,cysteine and methionine metabolism,glycine serine and threonine metabolism,and amino acid biosynthesis were *** carotenoids influenced the expression of several important genes involved in reactive oxygen species(ROS)generation,Nrf2 signaling,and antioxidant *** DEGs were chosen to confirm the RNA-Seq data using *** genes and pathways discovered in this work assist to elucidate the molecular processes through which archaeal carotenoid enhances *** antioxidative system.

Psychiatrists' approach to vascular risk assessment in Latin America

AIM: To explore the way in which Latin American psychiatrists approach the screening of vascular risk factors in patients receiving antipsychotic ***: This was a descriptive, cross sectional study that surveyed Latin-American physicians to evaluate differences between groups divided in three main sections. The first section included demographic and professional data. The second section asked about the available medical resources: weighing scales, sphygmomanometer and measuring tape. Finally, the third section aimed at looking into the attitudes towards car-diovascular prevention. The latter was also divided into two subsections. In the first one, the questions were about weight, blood pressure and waist perimeter. In the second subsection the questions asked about the proportion of patients:(1) that suffered from overweight and/or obesity;(2) whose lipids and glycemia were controlled by the physician;(3) that were questioned by, and received information from the physician about smoking; and(4) that received recommendations from the physician to engage in regular physical activity. The participants were physicians, users of the medical website Intramed. The visitors were recruited by a banner that invited them to voluntarily access an online self-reported structured questionnaire with multiple options. RESULTS: We surveyed 1185 general physicians and 792 psychiatrists. Regarding basic medical resources, a significantly higher proportion of general physicians claimed to have weighing scales(χ2 = 404.9; P metabolic indexes in the general population in a higher proportion than in patients treated with antipsychotics(Z =-11.91; P < 0.001), psychiatrists claimed to measure them in patients medicated with antipsychotics in a higher proportion than in the general population(Z =-3.26; P < 0.001). Diagnosis, treatment and prognosis of rhabdomyolysis

Background Statins are lipid-lowering drugs that are used widely around the world to treat and prevent car- diovascular diseases. Rare, but potentially fatal, cases of rhabdomyolysis （RM） have linked to the use of statins, especially in the presence of other risk factors. Methods We retrospectively analyzed the causes, clinical char- acteristics, treatments and prognosis of RM patients who were hospitalized between January 2005 and March 2016 at the Guangdong General Hospital, China. A total of 16 patients were included in the study （12 males and four females, mean age 60.69 years）. Amongst the patients, there were three cases of coronary heart disease, eight cases of dilated cardiomyopathy, six cases of atrial fibrillation, one case of pulmonary embolism, one case of aor- tic dissection, six cases of hypertension and one case of paroxysmal ventricular tachycardia. All patients had some degree of liver and kidney dysfunction, with low hemoglobin and myoglobinuria. Seven patients had a fe- ver and four had relevant neurological signs. All patients had digestive disorders and muscle weakness or pain during their stay in hospital. All patients received renal replacement therapy and 13 were placed on a ventilator to support breathing. Nine patients were eventually improved but seven patients died despite treatment. Results All sixteen cases of RM were associated with lipid-lowering drugs, with 14 patients taking statins and two pa- tients taking fibrates. Conclusions Statins were the main cause of RM in patients with poor prognosis and high mortality rate.
